My PhD. thesis is dealing with the problem of excessive blooms of cyanobacteria at the Brno reservoir. Due to the constant supply of nutrients, this recreation area is faced with the problem of eutrophication for many years. Partial solution was found after thorough limnological research and the first interventions carried out in 2008. Project started with the application of lime on exposed shore which was followed by the coagulation of phosphorus by ferric sulphate and installation of aeration equipment in water column. Part of the measure was the change of the fish stock as well. This project followed the whole process from 2006 to 2011. Water samples were taken from a motor boat at three localities. Physico - chemical parameters of water (pH, dissolved oxygen, water temperature) and amount of chlorophyll were measured in situ by FlouroProbe and YSI Probe. Microscopic analysis of phytoplankton performed in living samples or after fixing with formaldehyde. Our results show significant changes in the composition of phytoplankton biomass and reduce the number of cells per ml of water. New conditions in the aquatic environment are reflected especially during the vegetation seasons 2010 and 2011. Also new results from summer 2012 confirm the effectiveness of this project.

This work deals with the possibility of Raman spectroscopical identification of selected biomarkers of extremophile species. It focuses mainly on selected cyanobacteria, algae and lichens with an emphasis on the ability to detect carotenoids. These pigments exhibit three characteristic bands of Raman spectra which represent stretching vibrations C=C; C-C and bending vibration C-CH3 in molecules of carotenoids. Raman spectra were measured not only by laboratory microspectrometers (λ - 514 nm and 532 nm), but also by portable and handheld spectrometers (λ - 532 nm, 785 nm and 700 - 1100 nm). In the case of cyanobacteria, the spectroscopical analysis was performed also on the fractions obtained by high performance liquid chromatography (HPLC). This work critically evaluates the possibilities of Raman spectroscopy to identify the carotenoids of cyanobacteria, algae and lichens. Besides the signal of carotenoids, interpretation of other bands in the Raman spectra corresponding to the presence of other biomarkers is given here for selected samples. This diploma thesis deals with monitoring of water quality in the reservoirs which form the Litovicko- Šárecká cascade. It is a cascade of ponds, waterworks and retention reservoirs situated on the Litovicko-Šárecký stream and is composed of 9 major reservoirs. These reservoirs have different functions: from landscaping, retention, biological as well as recreation: fishing and swimming. Many of these reservoirs flow through human settlements, which have a negative impact on the quality of water within the whole cascade. The goal of this thesis is to monitor physical and chemical indicators of water as well as to determine the composition of phytoplankton populations, assess the state of water depending on the human activities in the basin and also to monitor how has the water quality changed in time and what are the main pollution sources. The water quality was determined from 11 samples collected with monthly intervals during March 2012 to May 2013. Samples were collected on all the reservoirs from the inflow, in the middle and at the outflow for chemical analysis and only from the middle for biological analysis. The phytoplankton samples from 45 localities in Vysočina region were collected in 2009. Species composition and relative abundance of species were studied; ecological factors like pH, conductivity, temperature, water transparency and dissolved ions were measured. Ecological indices (Shannon and Simpson index of diversity) were calculated and compared with other ecological factors. Checklist of all species present in Vysočina region was made. There were found 262 species and 4 species new for the Czech Republic.

Cyanobacterial communities of the splash zone of two Croatian islands, Veruda and Ugljan, were surveyed on 16 localities both in horizontal and vertical directions. In 336 samples 42 cyanobacterial species, 1 green alga, and 1 red alga were found. On both islands Hyella spp. and Gloeocapsopsis crepidinum dominated. Solentia spp., Kyrtuthrix dalmatica, and Mastigocoleus testarum were also found frequently. According to statistical analyses 3 horizontal parts of the splash zone were shown to host different cyanobacterial communities. In the horizontal direction there were no statistical differences among sampling points; however, for uncovering the whole cyanobacterial diversity more samples from various distances are necessary. The species diversity and distribution were not dependent on the orientation of the locality toward cardinal points.
