This thesis is focused on the dendrochronology of the chosen bell headstocks in the churches in Boskovice region. This work describes the entire process of selecting churches, samplings, measurements and a tree-ring dating. The tree-ring dating was focused on the bell headstocks in the Church of St.Jacob Mayor in Boskovice, The Holly Trinity Church in Drnovice, the Church of St. Stanislav in Kunštát, the Church of St. Prokop in Letovice, the Church of St.Peter and St.Paul in Lysice and the Church of the Assumption of Mary in Sebranice. The found dates are compared with the awailable literature and internet sources. The chronology extends back 435years (1383-1818).This chronology was used to complete the Czech oak chronology CZGES 2016. Now the Czech oak chronology contains samplings from the entire location of natural occurrence of oaks in the Czech Republic.

Title: The importance of myofeedback rehabilitation in faecal incontinence Introduction: Faecal incontinence is a serious symptom significantly worsening quality of life. Following results of international studies myofeedback has been considered an effective option of conservative treatment of this disease. Despite that it has so far been used at just a few centres in the Czech Republic. Purpose: The main purpose of this thesis was to assess the relevance of myofeedback as part of recovery therapy. Other objectives included assessment of use of a vaginal electrode instead of an anal one and comparison of the result correlations between examination by anal manometry, surface EMG and subjective improvement of continence for the patients. Methods: The research project included 8 females. The objective method of assessment of effects of the treatment involved examination of pelvis bottom muscles with the help of anorectal manometry and surface EMG. Success of the therapy was evaluated through the St Marks incontinence score and by the test population themselves filling out a quality of life questionnaire. Determination of the concentration of α1 - antitrypsin in the stool is a diagnostic indicator of inflammatory diseases of the small and the large intestine, especially malabsorption syndrome. α1 - antitrypsin belongs to the family of plasma proteins with antiproteinase effect. α1 - antitrypsin is synthesized in liver, in small amount in macrophage and is a protease inhibitor of serine proteases sercected from neutrophils. α1 - antitrypsin is acute phase protein. Higher α1 - antitrypsin values are in early phase of inflammation associated with raised CRP and other pozitive acute phase proteins. Fecal α1 - antitrypsin clearance is a sensitive and specific marker of protein loss. For α1 - antitrypsin determination in stool samples ELISA method can be used. ELISA is noncompetetive immunoassay used to detect presence of antibody or an antigen in a sample. The aim of this work was to compare two ELISA sets (Immundiagnostik and Ridascreen) used for determination α1 - antitrypsin in the stool. Then examine stability of α1 - antitrypsin in the stool and in extract prepared from stool in various storing conditions temperature and time. This diploma thesis deals with a development of a strategy for data evaluation generated by next-generation sequencing. Using bioinformatics tools such as Galaxy, Velvet and Enterovirus genotyping tool new aproach of data processing was optimized. There were 22 samples analyzed which of 10 were grown on cell culture. Remaining 12 were obtained from real stool samples. All samples were taken from children at the highest genetic risk of type 1 diabetes. All of them were enterovirus positive. Enteroviruses and their following infections have been suspecting to be involved in ehiology of type 1 diabetes for a long time. That's a disease resulting to an absolut insulin deficiency due to autoimmune destruction of pancreatic beta cells. Genetic components seems to be relatively well defined (the HLA, INS, STLA4, PTPN22, CTLA4, IFIH1 and numerous other genes), the environmental part of the etiology remains obscured. We were able to assemble 22 genomes de novo. However, there were numerous gaps among the particular contigs. For the first nine samples these gaps were complemented by Sanger sequencing. The output of this diploma work (dissertation) will be design of a construction for a tester enabling simulation of riding a vehicle with mounted snow chain in winter season on road without snow. The result of testing will be abrasion of single link of the chain caused interaction with surface of road and strength test of weld link e.g. when is pass gravelly surface.

The project elaborated in terms of engineering studies concern with project of a manufactural process of separator weldment and of an economical analysis of a production calculated for one item of a weldment and one item of a casting. During elaboration this project there were used all obtainable norms and technical bibliography. In literery studies there are summarized important knowledges related to welding constructions, welding technology, heat treatment and checking fusion splices. There was choosed box construction for this separator weldment. That is designed of two types of material. First of materials S355J2G3, EN 10025 is used for structural parts and the second material S235JR, EN 10025 is used for stiffering pars. The propriety checking of this construction and used materials was performed by Finite Element Method. After calculating costs of production of both variants we can say, that the costs of production of one piece seperator weldment are considerably lower, than costs of production of one piece of casting.

Construction of these accessories isn’t simple and simulation its characteristics need inspection in real measurements. Test bench for automobile radiator can help to easily choice and the project cooling system of the vehicles.

This final diploma thesis deals with the issue of vibration and noise measurement of transmission devices. The research part describes the basic issues of noise and vibration generation, their measurement, as well as sources of vibrations in gear units. The practical part deals with the design proposal of the attachment of the experimental single-stage gearbox and the load dynamometer to the anechoic chamber including the drawings. Further, by checking the modal properties of the proposed test stand, using the finite element method, the analytical and numerical control of the torsional vibrations of the rotating parts with respect to the expected frequency ranges of use.

The intestinal microbiota is composed of up to 100 trillion microorganisms of which bacteria are overwhelming majority. The microbiota affects the development of the immune system, defence against pathogens, host nutrition, vitamin synthesis or fat storage and its composition is changing throughout life. Some studies point to an association between microbiota composition and the development of inflammatory bowel disease. One of the treatment options is anti-TNFα antibodies therapy, which uptake or antagonize the TNFα cytokine that otherwise mediates inflammation in the intestinal mucosa. The aim of the thesis was to examine how this treatment affects the composition of the intestinal bacteriome in paediatric patients with Crohn's disease, and to find specific bacterial taxa, whose abundance changes during the treatment. By inclusion of patients with juvenile idiopathic arthritis, also treated with anti-TNFα, the study aims to discern specific effects of therapeutically induced intestinal restitution (observable in patients with Crohn's disease) from general effects of anti-TNFα therapy.
