L'interferon-γ (IFNγ) est une cytokine immunomodulatrice puissante, également dotée d'une activité antivirale. Il possède deux ligands de haute affinité : un récepteur par lequel il transmet ses signaux et des polysaccharides complexes de la famille des héparanes sulfates (HS), tous deux situés à la surface cellulaire. In vivo, la liaison aux HS permet de concentrer localement la cytokine et de réguler son activité biologique par le biais d'une protection partielle du domaine C-terminal de la protéine. Ce domaine C-terminal, caractérisé par deux domaines basiques D1 et D2, est impliqué dans la reconnaissance du récepteur et des HS. Dans ce contexte, nos travaux se sont attachés à définir les aspects structuraux de l'interaction de l'IFNg, et plus précisément de son extrémité C-terminale, avec ses deux ligands. Pour cela, de divers mutants ponctuels, multiples et de délétion de l'IFNg ont été produites, purifiées et étudiées. Leur capacité à lier les HS et le récepteur de l'IFNg est déterminée par SPR puis leur influence sur l'activité antivirale de l'IFNg est déterminée. Les paramètres thermodynamiques de l'interaction IFNg:HS-oligosaccharides sont investigués. Par ailleurs, nous avons préparé une banque oligosaccharidique dérivée d'HS. Le criblage de cette banque, pour sa capacité à lier l'IFNγ, a permis de démontrer que l'IFNg reconnaissait des motifs de sulfatation particuliers. Finalement, nous avons tenté de cristallisé le complexe IFNg:HS-oligosaccharide, jusqu'à présent sans obtention de cristaux qui diffractent. Ces différentes approches visent à élucider le mécanisme de reconnaissance d'IFNg par des HS. Ceci afin de concevoir un mime de ce site d'interaction inhibant la signalisation de l'IFNg. Enfin, une compréhension plus détaillé de l'interaction de l'IFNg avec les HS et son récepteur reste à établir afin d'entièrement comprendre comment l'IFNg migre des HS vers l'IFNgR.

Electrically Controlled Formation and Release of Admicelles for Solid Phase Extraction

Solid phase extraction is one of the most widely used methods to concentrate diluted compounds in a solution. Substances can be extracted into admicelles and hemimicelles, which are surface adsorbed micelles and surfactant monolayers, respectively. Investigations of the electrical control of surfactants on surfaces for the purpose of analyte preconcentration prior to chromatographic analysis are presented. The surfactant layer serves as the “stationary phase” in a solid phase extraction sorbent scenario. Analytes are adsorbed on this layer, and then released from the solid phase via surfactant removal. The attachment and removal of the surfactant are controlled by means of an electric field. Because the surfactant-analyte association is released by electrical control, organic solvents, which are used in conventional solid phase extraction, are not required. Therefore, this procedure is advantageous for method development and environmental concerns. Presented is the preconcentration of a test probe, 2-naphthol, using electrical control of the formation and release of dodecyl sulfate on planar gold, gold coated stainless steel, and a porous stainless steel frit, using impedance spectroscopy to observe the layer formation with various surfactant concentrations and applied potentials.

No. of bitstreams: 0 / O diabetes mellitus é uma doença que afeta o metabolismo de carboidratos, gorduras e proteínas e tem como aspectos característicos a hiperglicemia e a excreção da glicose pela urina. Estima-se que o número de casos da doença aumentará muito nos próximos anos, o que a torna preocupante para o sistema público de saúde dos países mais afetados. Além do uso de medicamentos alopáticos, tratamentos complementares como o uso de plantas medicinais pode contribuir para melhorar a qualidade de vida do paciente. Entre as ervas utilizadas, tanto no Brasil quanto em outras partes do mundo, encontram-se Bidens pilosa e Salvia officinalis. Os mecanismos responsáveis pela atividade antidiabética das plantas, geralmente estão associados aos seus metabólitos secundários, no entanto, a influência de micronutrientes não deve ser descartada. Estudos mostram que há uma correlação entre a concentração destes elementos com a presença e o desenvolvimento da doença. O objetivo deste trabalho foi estudar os elementos Cr, Fe, Mg, Mn, V e Zn em Bidens pilosa (picão) e Salvia officinalis (sálvia) cultivadas com tratamento normal (substrato comercial) e com adição dos elementos acima mencionados, e verificar a influência destes elementos na produção de metabólitos secundários que podem atuar como agentes hipoglicêmicos. A determinação e quantificação elementar foram feitas por meio da técnica de Análise por Ativação Neutrônica Instrumental, e para análise dos metabólitos secundários utilizou-se a técnica de Cromatografia Líquida de Alta Eficiência. O resultados indicaram que B. pilosa e S. officinalis podem ser usadas como fontes de Cr, Fe Mg, Mn, V e Zn. Foi observado que a B. pilosa absorveu maior quantidade de Fe no grupo tratamento, e que a S. officinalis foi capaz de acumular Zn nas folhas independente de ter ou não sido tratada. The effect of OsteoEze Gold™ on pain and functional ability in osteoarthritis of the knee

M.Tech. (Homoeopathy) / Osteoarthritis (OA) is a musculoskeletal condition affecting the synovial joints of the body, most commonly the knee and hip (Colledge et al., 2010). OA is the most prevalent joint disorder worldwide (Ickinger & Tikly, 2010). The prevalence of OA of the knee in developing countries, including South Africa, is expected to increase due to the increase in obesity and life-expectancy (Woolf & Pfleger, 2003). OA not only impacts negatively on many areas of the patient’s personal life, but it also has a considerable impact on health care systems and cost to the patient (Lapsley et al., 2001; Majani et al., 2005). The two main complaints in patients suffering from OA of the knee are knee pain and decreased daily functionality, such as walking (Samson et al., 2007). The main aim of conventional treatment is pain reduction. This treatment does not prevent progression of the OA, and may have negative side-effects (Day & Graham, 2005). Treatments for OA, such as OsteoEze GoldTM, may provide an effective and safer alternative. The aim of this study is to determine the effect of OsteoEze GoldTM on pain and functional ability in osteoarthritis of the knee using the Intermittent and Constant Osteoarthritis Pain (ICOAP) scale: knee version (Appendix D) and the Short Physical Performance Battery (SPPB) test (Appendix E). This was a 16-week study, conducted at the Homoeopathic Health Centre, Doornfontein campus (DFC), University of Johannesburg (UJ). The study was randomised, double blind placebo controlled, and matched pairs were utilised. Sixty-seven participants, who satisfied the inclusion and exclusion criteria, were recruited, and 48 of the participants completed the study. Participants were recruited by advertisements, placed in and around the UJ Homoeopathy Health Centre (with relevant permission given) and by word of mouth. The participants were split into two groups using matched pairs according to age, gender and severity of symptoms (Appendix H). The participants in group A received the OsteoEze GoldTM capsules, and the participants in group B received the placebo capsules. Each capsule of OsteoEze GoldTM contained 500mg glucosamine sulphate, 267mg of chondroitin sulphate, 50mg of vitamin C and 1mg of manganese. The OsteoEze GoldTM or the placebo capsules were distributed at the initial (week-0) and second (week-8) consultations.
