Previous issue date: 2009-05-26 / FAPEMIG - Fundação de Amparo à Pesquisa do Estado de Minas Gerais / A Toca de Assis é uma fraternidade católica de aliança e de vida fundada no Brasil, em 1994, pelo Pe. Roberto José Lettieri. Está a Toca de Assis, grosso modo, no lastro das novas comunidades eclesiais católicas de vida e aliança, surgidas, a maior parte delas, a partir das sensibilidades encontradas no movimento de renovação carismática católico. A tese visa investigar, particularmente, a comunidade de vida vinculada à Fraternidade Toca de Assis, isto é, o Instituto dos Filhos e Filhas da Pobreza do Santíssimo Sacramento. O instituto – de inspiração neofranciscana – tem atraído a adolescentes e jovens que a ele se filiam a partir da consagração religiosa, através dos votos de pobreza, castidade e obediência. Intentando ser uma nova congregação religiosa na Igreja Católica, o instituto, entretanto, tem apresentado características bastante particulares. Dentre elas destacamos: vivência radical da pobreza entre seus membros, que viveriam tão somente de doações e da fé na providência divina; trabalho junto à população em situação de rua, convivendo com ela e a acolhendo em suas casas de apoio; desconfiança e rejeição quanto aos estudos formais e a seus possíveis significados de racionalização; adoração ininterrupta aos elementos eucarísticos expostos; valorização de elementos litúrgicos, estéticos, teológicos e devocionais que representariam um modelo de Igreja Católica anterior ao do Concílio Vaticano II; acesso à liturgia e à vivência religiosa católica através de formas eivadas de características como as da emoção, afeto, e de expressões lúdicas e juvenis. A tese quer visualizar, através da pesquisa de campo e do aporte das ciências sociais da religião, estas e outras características, e percebê-las como expressões religiosas que acessam memórias e elementos religiosos pré-modernos através de sensibilidades modernas e pós-modernas, em configurações por vezes paradoxais. Finalmente, partindo de uma análise da dialética relação entre secularização e reencantamento, pretende a tese apontar para a busca a um universo de nova totalização católica, ou a um dossel sagrado, que protege e nominiza vidas e mundo na modernidade reflexiva da sociedade de risco, ainda que através de reflexividade e escolhas que comportam, elas mesmas, risco. / Die Fraternidade Toca de Assis (“Gemeinschaft der Zuflucht Von Assisi”) ist eine katholische religiöse Gemeinschaft, die 1994 in Brasilien von P. Roberto José Lettieri ins leben gerufen worden ist. Vereinfachend kann die Toca de Assis zu den neuen Gemeinschaften dês gemeinsamen Lebens und Glaubens innerhalb der katholischen Kirche gerechnet werden, die grösstenteils auf die in der katholischen charismatischen Erneuerungsbewegung vorgefundenen Bedürfnisse nach erlebter, emotionaler Religiosität zurückgehen. Gegenstand der vorliegenden Arbeit sind insbesondere die mit der TocaBewegung verbundenen Institute gemeinsamen religiösen Lebens, nämlich die Gemeinschaft der “Söhne und Töchter der Armut vom Allerheiligsten Altarsakrament” (männlicher und weiblicher Zweig). Dieser – neo-franziskanisch inspirierte – Verband übt Anziehungskraft auf Jugendliche und junge Erwachsene aus, die sich durch Gelübde der Armut, der Keuschheit und des Gehorsams als religiosen na die Gemeinschaft binden. Der Verband, der sich gern als neue Ordensgemeinschaft innerhalb der katholischen Kirche sehen möchte, weist allerdings einige recht ungewöhnliche Züge auf, darunter im Besonderen folgende Merkmale: Die Mitglieder leben in radikaler Armut und sollen ihren Lebensunterhalt ausschliesslich durch Spenden im Vertrauen auf die göttliche Vorsehung erwerben. Einen Schwerpunkt bildet die Arbeit mit Obdachlosen und Bedürftigen auf der Strasse, die in den “Zufluchtsstätten” (tocas) gennanten Häusern der Gemeinschaft aufgenommen werden. Geordneten theologischen Studien und wissenschaftlicher Bildung begegnet man aus Furcht vor einer Überbetonung der Ratio mit Misstrauen und Ablehnung. Praktiziert wird die ewige Anbetung der dauerhaft ausgesetzten eucharistischen Gestalten. Man legt Wert auf Frömmigkeitspraktiken und theologische, ästhetische und liturgische Vollzüge und Elemente, die ein Modell der katholischen Kirche repräsentieren, das hinter das 2. Vatikanische Konzil zurückgeht. Der Zugang zur Liturgie und zur katholischen religiösen Praxis ist sehr stark emotional und gefühlsbetont und erfolgt mithilfe jugendlich-spielerischer Ausdrucksformen und Gestaltungsmittel. Auf der Grundlag von Felduntersuchungen und unter Einbeziehung religionssoziologischer Ansätze und Forschungsbeiträge möchte die vorliegende Arbeit diese und weitere Kennzeichen der Gruppe sichtbar und als Ausdruck einer Religiosität erkennbar machen, die auf vormoderne religiöse Ideengebäude und Frömmigkeitselemente zurückgreift und dabei in manchmal paradoxer Weise modernen und postmodernen Wiederverzauberungsbedürfnissen entgegenkommt. Ausgehend von einer Analyse der dialektischen Beziehung zwischen Säkularisierung und Wiederverzauberung möchte die
Arbeit schliesslich ausblickende Hinweise auf ein Universum der totalisierung khatolischen oder eines heiligen canopy geben, das Schutzräume und Benennungsmuter für die Biografien in der Risikogesellschaft und die Welt der reflexiven Moderne bereithält, wenngleich durch Reflexivität und biografische Wahlentscheidung selbst wieder Risiken geschaffen werden.

Previous issue date: 2015-07-13 / Coordena??o de Aperfei?oamento de Pessoal de N?vel Superior (CAPES) / A porosidade e permeabilidade em zonas fraturadas podem aumentar devido a fluidos ascendentes em sistemas c?rsticos. Este trabalho apresenta uma an?lise estrutural de duas cavernas hipog?nicas da Am?rica do Sul . A ?rea de estudo inclui a Toca da Boa Vista (TBV) e Toca da Barriguda (TBR), que possuem 115 km e 32 km respectivamente. Este sistema c?rstico ocorre em carbonatos neoproterozoicos da Forma??o Salitre, por??o norte do cr?ton S?o Francisco. Durante o Brasiliano foram formados cintur?es de dobramento ao redor do cr?ton com padr?o compressivo desenvolvido entre 740-580 Ma. A metodologia inclui mapas de condutos do Grupo Bambu? de Pesquisas Espeleol?gicas, a partir dos quais foram elaborados mapas estruturais das cavernas e compara??o com a deforma??o regional. Os softwares usados para processamento de dados estruturais foram: Openstereo, Stereonet 8, Georient 9.4.5 e Tectonic FP. A An?lise da deforma??o do sistema TBV-TBR e o contexto regional da Forma??o Salitre permitiu concluir que: i) os condutos se desenvolveram ao longo de eixos de anticlinais N-S e ENE para E-W; ii) o desenvolvimento do carste foi formado pela dissolu??o de juntas subverticais; iii) o primeiro evento de dobramento F1 corresponde a trend N-S compressional e provavelmente se desenvolveu no Brasiliano, e, o segundo evento F2 est? relacionado a compress?o E-W, provavelmente mais recente que o Brasiliano. Portanto, ? poss?vel confirmar que existe rela??o direta dos fluidos ascendentes e dissolu??o hipog?nica com a deforma??o regional. / Porosity and permeability along fractured zones of carbonate rocks could be enhanced by ascending fluid flow in karst regions. This work presents a structural analysis of two largest hypogenic caves in South America. The study area encompasses the Toca da Boa Vista (TBV) and Toca da Barriguda (TBR) caves, which are 115 km and 32 km long, respectively. This karst system occurs in the Neoproterozoic carbonates of the Salitre Formation, in the northern part of the S?o Francisco craton. The fold belts around and at the craton edges were deformed during the Brasiliano orogeny in a compressive setting at 740-580 Ma. Methodology includes maps of conduits available by Bambu? Group of Speleological Research, which were used to constructo structural maps of caves and comparison to regional deformation. The following softwares were used to process structural data: Openstereo, Stereonet 8, Georient 9.4.5 e Tectonic FP. Analysis of deformation of TBV-TBR system and the context of Salitre Formation allowed to conclude that: i) Cave passages developed subhorizontally along (a) N-S and (b) ENE to E-W-oriented anticlines hinges; ii) Karst development were formed by dissolution of subvertical joints, iii) The first folding event F1 corresponds to a N-S-trending compression, probably during Brasiliano event whereas the second event F2, is related a E-W-compression, probably more recent than Brasiliano event. So, it is possible to confirm that fluid flow and related dissolution pathways have a close relationship with regional deformation.

Previous issue date: 2011-03-25 / Esta pesquisa objetivou estudar as práticas funerárias realizadas nos nove enterramentos evidenciados no sítio Toca da Baixa dos Caboclos/PI. Fundamentada em uma perspectiva conservadora, este estudo defendeu a hipótese de que a manutenção do sítio como espaço funerário poderia apontar a manutenção das próprias práticas funerárias dispensadas aos enterramentos nele evidenciados. Buscando validar esta hipótese, este trabalho se utilizou de uma metodologia ordenada e sistemática que permitiu reconstituir o Perfil Funerário do Sítio Toca da Baixa dos Caboclos, a partir da reconstituição dos seus respectivos Perfis das Unidades Funerárias. Segundo esta metodologia foi possível tecer considerações sobre as práticas funerárias dispensadas particularmente a cada enterramento e, ao mesmo tempo, entre todos os enterramentos estudados. Concomitantemente, o estudo da distribuição crono-espacial dos enterramentos possibilitou inferências acerca do aproveitamento do espaço interno do sítio, em distintos momentos cronológicos. Como resultado, foram identificadas recorrências relacionadas às práticas funerárias e também ao próprio agenciamento do espaço funerário. / This research aimed to study the funerary practices realized in nine burials evidenced in the archaeological site Toca da Baixa dos Caboclos/PI. Based on a conservative perspective, this study supported the hypothesis that maintaining the site as a burial space could point to maintenance of their own funerary practices. Seeking to validate this hypothesis, this study used a methodology systematic and ordered to reconstruct the Funerary Profile of site Toca da Baixa dos Caboclos, from the reconstitution of their respective Profiles of Funerary Units. According to this approach it was possible to comment on the funerary practices dispensed to every burial and at the same time, among all burials studied. At the same time, the study of chrono-spatial distribution of burials allowed inferences about the use of internal space of the site at different moments chronological. As a result, recurrences identified were related to funerarypractices and also in their own funerary use of internal space of the archeological site.

Toca-1 driven actin polymerisation at membranesFox, Helen Mary January 2018 (has links)
Regulation of the actin cytoskeleton is key to cellular function and underlies processes including cell migration, mitosis and endocytosis. Motile cells send out dynamic actin protrusions that enable them to sense and interact with their environment, as well as generating physical forces. Linking of the actin cytoskeleton to the cell membrane is essential for the formation of these protrusions. The proteins that are thought to fulfil such a role have a membrane interacting domain (such as the PH domain in lamellipodin, or I-BAR protein in IRSp53) and a domain which interacts with actin regulatory proteins (such as the SH3 domain of IRSp53, which binds Ena and VASP). I investigated the contribution of the F-BAR protein Toca-1 in linking actin polymerisation to membranes, by characterising a new protein-protein interaction and the interaction of Toca-1 with giant unilamellar vesicles. FBP17, a homologue of Toca-1, can oligomerise to form 2D flat lattices and 3D tubules on membranes. Proteins of the Toca-1 family have previously been implicated in actin polymerisation in cell-free systems and during endocytosis. However, there is emerging evidence that Toca-1 family proteins could also be involved in the formation of outward facing protrusions, lamellipodia and filopodia. In an in vitro system that recapitulates the formation of filopodia-like structures (FLS) on supported lipid bilayers, Toca-1 is recruited early, suggesting a Toca-1 scaffolding mechanism could precede the recruitment of other actin regulators. One prediction of this model is that Toca-1 would bind proteins previously implicated in filopodia formation, such as formins. I found that extracts depleted of Toca-1 binding partners no longer forms filopodia-like structures and subsequently optimised pull-down assays to identify Toca-1 binding partners by mass-spectrometry. I identified four formins, Diaph1, Diaph3, FHOD1 and INF2, and as well as the actin elongation factors and filopodia proteins, Ena and VASP. I further characterised these interactions and found that Toca-1 binds Ena and VASP via its SH3 domain. The interaction is direct and is strongly reduced if the proline-rich region in Ena is deleted. VASP was still able to bind without its proline rich region, suggesting there could be additional binding sites. I discovered that the binding of Ena and VASP was dependent on the clustering state of Toca-1, whilst the binding of the previously identified Toca-1 binding partner N-WASP was not. This further supports the importance of Toca-1 oligomerisation in actin polymerisation. I tested these interactions in the FLS system and found that increasing Toca-1 concentration leads to increased recruitment of N-WASP, as well as the novel binding partner Ena to the structures, whereas an increase in VASP was not observed. SH3-domain mediated interactions are required for Toca-1 recruitment to FLS, suggesting that its membrane and protein binding activities act cooperatively. I showed that unlike N-WASP, which promotes the formation of branched actin, Ena and VASP are not required for actin polymerisation on supported lipid bilayers, suggesting that they are redundant with other factors in the elongation step of FLS formation. Ena and VASP are known to be important for the formation of neuronal filopodia and so I began to further test the role of these interactions in a cellular context using a neuronal cell culture system. As well as recruiting protein binding partners, F-BAR family proteins are implicated in stabilising lipid microdomains and can induce the clustering of phosphoinositides. I investigated the role of Toca-1 in actin polymerisation on PI(4,5)P2-rich giant unilamellar vesicles (GUVs). Actin-rich tails formed on the GUVs only when excess Toca-1 was supplemented into the extracts, and I propose that this is due to lipid organisation by Toca-1. In summary, my work suggests a model in which Toca-1 clusters, stabilises the membrane lipids and recruits regulators of actin polymerisation, such as Ena. This mechanism could be used to link actin polymerisation to the membrane in cellular protrusions, such as filopodia.
