The diploma thesis is focused on the issues of financial planning. In the introductory chapter the basic theoretical terms, which are important to understanding the issue, are explained. The aim of this thesis is to create a financial plan for selected company for the next 3 years, which is also the subject of the main part of this diploma thesis. The financial plan is compiled in two variants: optimistic and pessimistic. In both options of the financial plan the consequences of the company's planned investment will incorporate. At the end of the thesis, the evaluation and control of the results of both variants of the financial plan is made.

The subject of the thesis was the design of a multi-storey office building. In the transverse direction the building was designed as a three-tract 6.0 + 4.0 + 6.0 = 16 m. In the longitudinal direction 9x6.6 m = 59.4 m. The design was designed in several variants: articulated and frame in transverse directions. The joint variant was selected for detailed analysis.

Překlad dialektů v hraných filmech a seriálech do češtiny / Czech translation of dialects in feature films and TV series

The thesis studies Czech dubbing of Misfits and Futurama TV series with special attention to the method of translation of regional varieties of English. The first part summarizes the latest theoretical approach to translation of marginal varieties in dubbing and literature. This overview is followed by an analysis of language varieties used in the original series and their dubbed versions in order to define their role and function. The second part of the thesis examines decision-making processes employed in the process of translation of the regional varieties and presents the results of a survey of spectators' opinion on the translation method employed. Based on the survey results we compared spectators' evaluation of the original series with their evaluation of the dubbed versions, as well as markings of characters using regional varieties with markings of those using standard language. Last but not least, markings of the dubbed versions in the regions where the variety in question is spoken were compared with markings of these versions in the rest of the Czech Republic. The aim of the comparisons was to get an overview of recipients' opinions on the method of translation applied.

(EN) The doctoral thesis Varianta v poetice Bohumila Hrabala [The process of variation in Bohumil Hrabal's poetics] studies the creative process of writing variants, which the author employed in the whole of his literary production. The main theoretical focus of the thesis is the concept of semantic "openness" introduced by Italian semiotician Umberto Eco (1932-2016) as the possibility of inter­ preting a literary work on the basis of the aestetic information (i.e. the formal construction) of the text. A further elaboration of Eco's concept is to be found in a case study conducted by Michael Špirit (1965-) on one text written by Hrabal in the 50s and presented by Špirit as an example of a concluded text yet open in meaning, and its variant published in the 60s, which according to Špirit has the features of an open text, but is semantically closed. Through the textual analysis of three groups of variant texts written by Hrabal, the thesis argues the development of the process of variation from texts written in the 50s towards the semantic closed­ ness of their variants published in the 60s (epos Krásná Poldi- the short story Krásná Poldi; short stories Kain and Fádní stanice-novella Ostře sledované vlaky-short story Legenda o Kainovi), whi­ le demonstrating the opposite direction of the same process...

Carriers of apparently balanced chromosomal aberrations (BCA) are usually phenotypically normal. However, it has been estimated that up to 27% of these BCA may be associated with an abnormal phenotype, most often caused by cryptic imbalances at the breakpoints, gene disruption by the breakpoint or via the position effect. In contrast to conventional karyotyping, molecular cytogenetic techniques enable more detailed BCA characterization and better correlation between genotype and phenotype of the patient. The aim of this thesis was to evaluate the presence of copy number variants (CNVs) at breakpoints or elsewhere in the genome in patients with abnormal phenotype who carry de novo or inherited BCA. 54 BCA were investigated using array CGH (20 de novo cases, 27 inherited and 7 cases of unknown origin) including 32 reciprocal translocations, 6 robertsonian translocations, 12 inversions and 4 complex chromosomal rearrangements. If possible, the parents were also examined to ascertain the inheritance of the relevant CNVs. In order to specify microarray findings or exclude gene disruption, FISH was used in selected patients. 3 Abstract The doctor thesis is composed of two major studies, both of them focused on the mantle cell lymphoma (MCL). The first part deals with the verification of the prognostic influence of Mantle Cell Lymphoma International Prognostic Index (MIPI) and of the proliferative activity in 235 patients with MCL diagnosed in 1996-2008 in the Czech Republic. This population study was performed in the collaboration with the Czech Lymphoma Study Group. The clinical data of patients were completed in April 2012. The diagnosis of MCL was confirmed by our central histopathologic examination of pretherapeutic histological samples. The median overall survival (OS) was 47 months, median progression free survival (PFS) was 22 months. We demonstrated the influence of proliferative activity, MIPI and of the therapy type (intensive/non-intensive) on OS and PFS in univariate and multivariate analysis. Using univariate analysis we showed the prognostic influence of aggressive/other cytomorphological variants of MCL, nodal/extranodal localization of primary sample and also of the variants of MIPI - s-MIPI, MIPIb and a completely new variant of MIPI - combined MIPI. The work addresses the problem of evaluating investment options, both in theoretical and practical terms. In theoretical terms, there are firstly defined the basic terms, but especially the complex issue of implementation of investment projects in all stages of life of a project. Attention is also paid to the evaluation of investment alternatives, selecting individual assessment methods (ROI, PP, NPV, IBR, etc.), analysis of financial risks, and the methods of multi-criteria evaluation of investment options, including software tools that use these methods in practice much easier. The topic of this thesis is to create a financial plan for a selected company, which is the company ZEMO, spol. s r.o. with its headquarters in Bohaté Málkovice, which is engaged in agricultural activities, especially plant production. In the first part of the thesis there are the theoretical starting points, then the selected company is introduced, the strategic and financial analysis is performed and the financial plan for the years 2018 - 2021 in the optimistic and pessimistic variant is processed. Finally, an evaluation of both variants of this financial plan is made.
