Klonální vývoj leukemických buněk a jeho úloha při progresi leukémií a preleukémií / Clonal evolution of leukemic cells and its role in the progression of leukemia and preleukemia

Clonal evolution is a multistep process characterized by progression of the disease, adverse prognosis and shortening of overall survival. The aim of the dissertation was a detailed characterization of identified changes in patients with myelodysplastic syndromes (MDS) and clonal evolution and evaluation of their prognostic impact. We performed detail cytogenomic analyses in 36/469 (8%) patients with confirmed linear clonal evolution. We described 57 primary abnormalities (32% MDS-specific) at the time of diagnosis, the most frequent was deletion of long arm of chromosome 5. We proved 156 secondary aberrations (21% MDS-specific) during the course of the clonal evolution, the most frequent were trisomies/tetrasomies of chromosome 8. We identified acquired uniparental disomies (aUPD) in 19% of patients. In MDS-specific aUPDs 4q, 11q and 17p, we proved homozygous mutations of TET2, c-CBL and TP53 genes. We found a statistically significant difference in overall survival between the groups of patients divided according to their diagnostic cytogenomic findings. In patients with clonal evolution before treatment 54% of aberrations were gains of whole chromosomes, by contrast 44% of abnormalities identified in patients with clonal evolution after treatment were monosomies or deletions. Pavlína Šumníková, Ph.D., Praha 2010 ANNOTATION: The work deals with identifying the level of olfactory perception among people with severe visual impairment. The aim is to compare the function of olfactory discrimination and olfactory threshold sensitivity among persons with congenital or acquired visual handicap. The theoretical section presents the basic anatomical and physiological characteristics of the olfactory system, the list of disorders of smell and olfactory testing methods in clinical practice, and a brief account of visual impairments. The empirical investigation is carried out using standardized tests, olfactory discrimination function is measured by a test of the University of Pennsylvania Smell Identification Test (UPSIT), olfactory perception threshold using the Smell Threshold Test (STT). The thesis describes in detail both the methods and characteristics of groups of test subjects. In the analytical part it focuses on the comparison of test results among persons with congenital or acquired severe visual impairment.

This study examines the role of innate and adaptive immunity in the immunotherapy based on the combination of the ligands stimulating phagocytosis anchored in the tumour cells membrane and the mixture of TLR agonists. This immunotherapy is primarily focused on the innate immunity activation and induces strong inflammatory infiltration, which neutrophils and NK cells are part of. Therefore, the next aim of this study was to evaluate the anti-tumour activity of neutrophils and NK cells. For examination malignant melanoma and pancreatic adenocarcinoma mouse tumour models were used.

The work deals with the choice and implementation of an open source tool for managing a project portfolio management in the organization of o.s. Prostor. The first part of the work aims at theory description including examination of trends in project management tools and open source software. Project management and project portfolio management is increasingly important and increasingly used by companies and organizations in recent years. According to a new study made by Gartner, the use of open source software continues to grow and becomes competitive differently. The main aim of the work was to successfully implement one selected open source tool to the organization that I have found at an early stage. After finding the suitable company, I started to do the main part of my work i.e. the implementation of a selected open source tool for managing projects and its portfolio in this company. This phase started from the choice of the most suitable instrument, by means of analysis of the selected organization, o.s. Prostor, to the final design and implementation itself. I made analysis of the organization and on its basis I chose suitable criteria and their weights for the later selection of a suitable tool. I performed the choice based on multi-criteria analysis in order to maximum objectively select a suitable open source tool for implementation. None of the existing instruments fully met the requirements of the organization o.s. Prostor. Therefore, I assessed finally selected web2project tool in detail and made a proposal for additional programming of missing functionality. After completion of the missing functionality additional programming, I made a proposal of a web2project tool implementation procedure. I suggested four stages of the implementation procedure, which consists of the preparation for the installation environment, the way how to install the tool on the server, planning necessary capacity, and finally training including creation of a demonstration project. Finally, based on this proposal, I installed the tool web2project on the server and run testing and validation of the tool. When performing this work, I gained a lot of valuable experience in all relevant phases of the project cycle management from management to the implementation itself. The organization intends to fully operate the tool early next year, 2012, when they can begin to enter new projects.

Title: Acquired flatfoot in adults Background: Flatfoot deformity is one of the common diagnoses in the orthopedic field and physiotherapy, its common in adults as acquired, there is still a large incidence of flatfoot in adolescence and many cases of adult flatfoot are frequently presented as residual pediatric flatfoot. The treatment also has wide field according to type of cause. Aims: The aim of this literature review was to enhance knowledge on the persistence of the acquired flatfoot condition in adult's population by determining the causes of flatfoot with contributing factors that affect the medial longitudinal arch and progression of the deformity, and review of the most common treatment. Method: An electronic database search was conducted to obtain articles from relevant journals (from early 1990 to end 2015). The information was collected also from textbooks. Results: Regarding the causes of acquired flatfoot in adult, a good base of evidence stems from a number of articles review, the posterior tibialis tendon dysfunction is the most cause of acquired flatfoot. The sensory motor stimulation technique is not the most treatment used to treat acquired flatfoot in adults and the most used treatment is the surgical procedure.
