Previous issue date: 2015 / O Transtorno do Espectro Autista (TEA) e um disturbio neurocomportamental complexo desenvolvido na infancia precoce, caracterizado por varios graus de deficiencia, incluindo prejuizo nas areas do desenvolvimento pertinentes a interacao social, comunicacao, comportamento e interesse. O TEA apresenta uma prevalencia global estimada de 1 para cada 68 criancas, seu diagnostico se concentra basicamente nas avaliacoes comportamentais, nao havendo ate o momento biomarcadores para identifica-lo. Tanto fatores geneticos quanto ambientais, vem sendo ligados diretamente ao TEA. A heterogeneidade de suas manifestacoes faz com que os tratamentos tornam-se ineficazes. Novas alternativas terapeuticas tem sido sugeridas para melhorar os sintomas, entre eles o acido graxo poliinsaturado (AGP) omega 3 (¿Ö-3), tem apresentado resultados positivos no tratamento de doencas neurologicas e psiquiatricas. O objetivo deste estudo foi avaliar os efeitos da suplementacao de AGP ¿Ö-3, sobre os parametros comportamentais e bioquimicos em um modelo experimental de autismo induzido por lipopolissacarideo (LPS) a partir da ativacao imune materna. A suplementacao AGP ¿Ö-3 ou solucao salina foi administrada na prole a partir do dia pos-natal (PND) 30, na dose de 0,8 g/kg, via oral. No PND 52 os animais realizaram testes comportamentais, apos foram eutanasiados e as estruturas do encefalo dissecado e analisado em niveis de: enolase especifica do neuronio (NSE), fator neurotrofico derivado do cerebro (BDNF) e fator de transformacao do crescimento beta (TGF-¿À). Os resultados deste estudo indicaram que a ativacao imune materna causou na prole alteracoes comportamentais e neuroquimicas condizentes com o TEA, e a suplementacao com o AGP ¿Ö-3 apresentou resultados positivos, tanto comportamentais como neuroquimicos. / The Autism Spectrum Disorder (ASD) is a complex neurodevelopmental disorder developed in early childhood, characterized by several degrees of deficiency, including deficits in the areas of the development relevant to social interaction, communication, behavior and interest. The ASD has an estimated global prevalence of 1 per 68 children , its diagnosis focuses basically on behavioral assessments, and there is no biomarkers to identify it so far. Both genetic and environmental factors, have been linked directly to the ASD. The heterogeneity of its manifestations causes the treatments to become ineffective. New therapeutic approaches have been suggested to improve symptoms, including polyunsaturated fatty acid (PUFA) omega-3 (ù-3), has shown positive results in the treatment of neurological and psychiatric diseases. The objective of this study was to evaluate the effects of PUFA ù-3 supplementation on the behavioral and biochemical parameters in an experimental model of autism induced by lipopolysaccharide (LPS) from the maternal immune activation. The PUFA ù-3 supplementation or saline solution was administered in offspring from postnatal day (PND) 30, at a dose of 0.8 g/kg, orally. At PND 52, animals underwent behavioral tests, were euthanized after and the brain structures dissected and analyzed for levels: neuron specific enolase (NSE) , brain-derived neurotrophic factor (BDNF) and transforming growth factor beta (TGF-â). The results of this study indicate that maternal immune activation caused behavioral and neurochemical changes in the offspring consistent with the ASD, and supplementation with PUFA ù-3 showed positive results, both behavioral and neurochemical.
