Previous issue date: 2003-08-29 / Embora o aumento na concentra??o de Esp?cies Reativas de Oxig?nio (Eros), devido ao diabetes mellitus, pode alterar o metabolismo normal do osso, acarretando assim a osteopenia. Uma suplementa??o com c?lcio, f?sforo e vitamina E pode ter a??o ben?fica e prevenir as complica??es associadas ao estado diab?tico. Foram utilizados ratos Wistar machos (n=125), peso corporal 250-350g, sendo divididos em tr?s grupos: grupo 1- controles (n=45) grupo 2- diab?ticos experimentais (n=45), e grupo 3 - diab?ticos suplementados (n=35) em c?lcio, f?sforo e vitamina E. O diabetes experimental foi induzido por estreptozotocina (STZ) (45 mg/Kg) e sacrificados em per?odos de 1 e 5 dias ap?s a indu??o e 5, 30, 60 e 90 dias ap?s a instala??o do diabetes mellitus. Animais com glicemia ? 250 mg/dL foram considerados diab?ticos. Realizaram-se an?lises de Subst?ncias Reativas ao ?cido Tiobarbit?rico (SRAT) e Glutationa (GSH) em sangue e c?rebro, medidas histomorfom?tricas dos f?mures esquerdos, al?m de dosagens bioqu?micas de fosfatase alcalina, fosfatase ?cida, fosfato, magn?sio, c?lcio ioniz?vel, c?lcio total, creatinina e albumina em amostras de soro e glicose, fosfato, magn?sio, creatinina e clearence de creatinina em amostras de urina dos animais. As SRAT no plasma e c?rebro, apresentaram-se significativamente elevadas no grupo 2 em rela??o ao grupo 1, e diminu?da para o grupo 3, no per?odo de 90 dias. O conte?do de GSH em c?rebro, no per?odo de 90 dias, diminuiu no grupo 2 e aumentou no grupo 3, enquanto em sangue n?o foram observadas altera??es. Os resultados histomorfom?tricos mostraram uma perda em m?dia de 50% de osso trabecular avaliadas pela dist?ncia entre as trab?culas, espessura das trab?culas e volume ?sseo trabecular, nos animais diab?ticos experimentais em rela??o aos controles e, com a suplementa??o, essa perda diminuiu significativamente alcan?ando valores pr?ximos aos encontrados no grupo controle. A atividade da fosfatase alcalina apresentou valores superiores para os grupos 2 e 3 em rela??o ao grupo 1 e a fosfatase ?cida aumentou significativamente nos grupos 2 e 3, em rela??o aos controles, no per?odo de 90 dias. As concentra??es s?ricas de fosfato, magn?sio, c?lcio ioniz?vel, c?lcio total e creatinina, n?o apresentaram altera??es uma vez que os resultados est?o dentro dos valores de intervalos de refer?ncia. A albumina s?rica mostrou-se diminu?das para o grupo 2 e 3. A an?lise da urina mostrou glicos?ria significativa para os grupos 2 e 3 em rela??o ao grupo 1. Em rela??o a fosfat?ria observou-se um aumento significativo principalmente para o grupo 3 e em rela??o ao clearence de creatinina n?o foram observadas diferen?as significativamente para o grupo 2 e 3. Os resultados observados sugerem que a suplementa??o de c?lcio, f?sforo e vitamina E diminuiu as altera??es oxidativas causadas pelo diabetes, al?m de ter exercido a??o ben?fica sobre o metabolismo ?sseo, diminuindo a perda ?ssea que ocorre durante esta s?ndrome. / The increase of EROs concentration, through diabetes mellitus, can alter the normal bone metabolism, causing osteopenia. A calcium, fosforum and vitamin E supplementation, can be a very good action in bone. In thi study were used male Wistar rats (n=125), weight ranging from 250 ? 350g, and were divided in three groups: group 1 control (n=45) group 2 experimental diabetic (n=45) and group 3 supplementadies diabetic (n=35) in calcium, fosforum and vitamin E. The animals were inuced with streptozotocin (45mg/Kg ) and killed in periods of 1 to 5 days after the indution e 5, 30, 60 and 90 days after installation of experimental diabetes mellitus. Animals with glycemia ? 250 mg/ dL was considered diabetics.In each period was realized determinations of Thiobarbituric Acid Reactive Substances and levels of reduced glutathione in blood and brain, histomorfometric measurement of left fermurs, and dosages of alkaline phosphatase, acid phosphatase, ionizable calcium, total calcium, phosphorus, magnesium, glycemia, creatinin and albumin in serum and glucose, phosphorus, magnesium, creatinine and clearence of creatinin in urine. Thiobarbituric Acid Reactive Substances at brain and plasma, choose higher at group 2 in relation group 1, and reduced in group 3, in period of 90 days. The levels of reduced glutathione in brain and blood, at 90 days, was decreased in group 2 and increased at group 3. The histomorphometric results show a less in middle of 50% in trabecular bone volume analysed per distance between the trabeculaes thickness of trabeculae and bone quantity in experimental diabetic animals in comparison to the control group and for controls group and, supplemented animals this lose reduced very much arriving sizes near found in control group. The activity of alkaline phosphatase showed superior values in relation of groups 2 and 3, about comparing group 1 and acid phosphatase increased higher in groups 2 and 3, about all controls, in a 90 days. The serum concentration of phosphorus, magnesium, ionazle calcium, total calcium and creatinine, didn?t show alterations, and the results are well if compare the reference result. The albumin serum showed less to group 2 and 3. The urine analyse showed significative glycos?ria to groups 2 and 3 if compared with group 1. In relation phosphatury, observed a significative increase principally to group 3 and creatinine clearance relation wasn?t observed differences to group 2 and 3. The results observed suggest that calcium, phosphorus and vitamin E supplementation, decreased the oxidative alterations which the diabetics were the cause, and helped bone metabolism, decreasing the lose bone that ocurr in this syndrome
