Previous issue date: 2015-02-04 / The PACAP neuropeptide (pituitary adenylate cyclase-activating polypeptide) has a wide spectrum of biological functions, such as neurotransmitter, neuromodulator, neuroprotective and neurotrophic factor. Furthermore, studies suggest that PACAP plays an important role in the modulation of social behavior, learning and memory, as well as the stimulation of nitric oxide (NO) production and modulation of the activity of the NMDA-type glutamatergic receptors. This study aimed to investigate, in the CA1 region of the dorsal hippocampus and the basolateral amygdala (BLA), the participation of PACAP in the contextual fear conditioning memory (CFC) and social recognition memory (SR). For this, adult male Wistar rats with cannulas implanted bilaterally in the CA1 region of the hippocampus or the BLA, were trained in contextual fear conditioning or social recognition tasks. It was observed that the PACAP antagonist (PACAP 6-38, 40 pg/side), when infused intra-CA1 or intra-BLA immediately after the training session of the CFC, impairs consolidation of memory. This effect was not observed when NMDA receptor agonist (D-serine, 50 ?g/side) was infused concomitantly with PACAP 6-38. It was also found that when infused intra-CA1 immediately after the training session the extinction of the CFC, the PACAP 6-38 impairs memory of extinction, however, this effect was not observed in BLA or when the PACAP 6-38 was infused intra-CA1 concomitantly with D-serine. Further, it was found that when infused intra-CA1 or intra-BLA immediately after the social recognition task, PACAP 6-38 impairs consolidation of memory and this effect was not observed when nitrous oxide donor (SNAP; 5 ?g/side) was infused concomitantly with PACAP 6-38. The results obtained in this study suggest that PACAP participates in the consolidation and extinction of CFC memory, and that this effect seems to occur through NMDA receptors. Furthermore, the results also indicate that PACAP participates in the consolidation of the social recognition memory, and that this effect seems to occur through the action of nitric oxide / O neuropept?deo PACAP (polipept?deo ativador da adenilato ciclase pituit?ria) possui um amplo espectro de fun??es biol?gicas, como neurotransmissor, neuromodulador, neuroprotetor e fator neurotr?fico. Al?m disso, estudos sugerem que o PACAP desempenha um importante papel na modula??o do comportamento social, aprendizagem e mem?ria, bem como na estimula??o da produ??o de ?xido n?trico (NO) e na modula??o da atividade dos receptores glutamat?rgicos do tipo NMDA. O presente estudo teve como objetivo investigar, na regi?o CA1 do hipocampo dorsal e na am?gdala basolateral (BLA), a participa??o do PACAP nas mem?rias de medo condicionado ao contexto (MCC) e de reconhecimento social (RS). Para isso, ratos Wistar machos adultos, com c?nulas implantadas bilateralmente na regi?o CA1 do hipocampo ou na BLA, foram treinados nas tarefas de medo condicionado ao contexto ou reconhecimento social. Verificou-se que, o antagonista do PACAP (PACAP 6-38; 40 pg/lado), quando infundido intra-CA1 ou intra-BLA imediatamente ap?s a sess?o de treino do MCC, prejudica a consolida??o dessa mem?ria. Contudo, esse efeito n?o foi observado quando o agonista dos receptores NMDA (D-serina; 50 ?g/lado) foi infundido concomitantemente com o PACAP 6-38. Verificou-se tamb?m que, quando infundido intra-CA1 imediatamente ap?s a sess?o de treino da extin??o do MCC, o PACAP 6-38 prejudica a mem?ria de extin??o, entretanto, esse efeito n?o foi observado na BLA ou quando o PACAP 6-38 foi infundido intra-CA1 concomitantemente com D-serina. Ainda, verificou-se que, quando infundido intra-CA1 ou intra-BLA imediatamente ap?s a tarefa de reconhecimento social, o PACAP 6-38 prejudica a consolida??o dessa mem?ria, por?m, esse efeito n?o foi observado quando o doador de ?xido n?trico (SNAP; 5 ?g/lado) foi infundido concomitantemente com o PACAP 6-38. Os resultados obtidos no presente trabalho sugerem que o PACAP participa da consolida??o e da extin??o da mem?ria de MCC, e que esse efeito parece ocorrer atrav?s dos receptores NMDA. Al?m disso, os resultados tamb?m indicam que o PACAP participa da consolida??o da mem?ria de reconhecimento social, e que esse efeito parece ocorrer atrav?s da a??o do ?xido n?trico
