Pesquisa do Estado da Bahia - FAPEB / Chemical characterization and biological activity tests of plants are important approaches to drug discovery, rational use of elements of our biodiversity in view of the variety of plants reflects the diversity of their secondary metabolites and therapeutic potential. Brazil is an invaluable source of natural resources to obtain drugs, however, little study has been done on the pharmacological potential of native species. The Bromeliaceae family includes about 58 genera and about 3,352 species. Brazil comprises about 40% of the total species with Ananas comosus being the best known. Some plants of this family are used for the treatment of various diseases and there are reports on the presence of secondary metabolites such as triterpenoids, steroids, flavonoids, glycerols, cinnamic acid derivatives and others. This study aimed to know the chemical composition and biological activities of accessions and hybrids of different botanical varieties of Ananas genus in partnership with (EMBRAPA). The leaves of the harvested specimens were dried, ground, and subjected to an extraction procedure with methanol and were pre-selected via the screening and biological screening in order to obtain the best species profile. Three accessions and one hybrid were selected, they were subjected to extraction with methanol, then partitioned with solvents of different polarities. Artemia salina lethality, antioxidants and acetylcholinesterase activity tests were performed. The chromatographic fractionation of the species with better chromatography profile was performed using different techniques. The results revealed that 21 collected accessions have different secondary metabolites, and a wide variation among themselves showing no standard. In relation to biological testing several accessions showed lethality to Artemia salina, antioxidants and acetylcholinesterase activity. The results indicate that extracts of accessions contain active constituents that present such activities confirming its biological potential. As a result of preliminary studies the accessions BGA-739, BGA-804, BGA-820 and hybrid HE were selected for further study, and biological tests were conducted. The BGA- 804 access, chloroform fractions (CL) and ethyl acetate (ACT) has been shown to have high potential antioxidant represented by the EC50 in ?g.ml-1 804-CL (203,5?0,528) e 804-ACT 192,5?0,193, lethality represented by the LC50 in ?g.ml-1 804-CL (69,3?0,47) and 804-ACT (87,7?0,49) and inhibition of the enzyme cholinesterase in % 804-ACT (53.230 ? 4,44). Fractionation, chemical profile by HPLC / DAD and chemical characterization of the BGA-804 access, resulted in the finding of the presence of phenolic compounds, flavonoids and the isolation and identification of daucosterol originating from 804 CL fraction. / Caracteriza??o qu?mica de plantas e testes de atividades biol?gicas s?o abordagens importantes na descoberta de novos medicamentos e para o aproveitamento racional de elementos de nossa biodiversidade, tendo em vista que a variedade de plantas reflete a diversidade dos seus metab?litos secund?rios e seu potencial terap?utico. O Brasil ? fonte inestim?vel de recursos naturais para obten??o de farm?cos, contudo as esp?cies nativas t?m sido pouco estudadas quanto ao potencial farmacol?gico. A fam?lia Bromeliaceae inclui aproximadamente 58 g?neros e cerca de 3.352 esp?cies. O Brasil abriga cerca de 40% do total de esp?cies, sendo a mais conhecida o Ananas comosus. Algumas plantas desta fam?lia s?o utilizadas para tratamento de diversas afec??es e existem relatos da presen?a de metab?litos secund?rios, como: triterpenos, esteroides, flavonoides, glicer?is, derivados do ?cido cin?mico e outros. Este estudo teve como objetivo conhecer a composi??o qu?mica e as atividades biol?gicas de acessos e h?bridos de diferentes variedades bot?nicas do g?nero Ananas, em parceria com (EMBRAPA). As folhas dos esp?cimes coletadas foram secas, mo?das, submetidas ao procedimento de extra??o com metanol e pr?-selecionadas via screening e triagem biol?gica, com a finalidade de obter esp?cies com o melhor perfil. Foram selecionados 3 acessos e 1 h?brido, estes foram submetidos a extra??o com metanol, posteriormente particionados com solventes de diferentes polaridades e realizados testes de letalidade frente ? letalidade frente ? Artemia salina, atividade antioxidantes e anticolinester?sica, o fracionamento cromatogr?fico das esp?cies com melhor perfil foi realizado utilizando diferentes t?cnicas de cromatografia. Os resultados revelaram que os 21 acessos coletados apresentam diferentes metab?litos secund?rios e uma varia??o grande entre si, n?o apresentando nenhum padr?o. Com rela??o aos testes biol?gicos v?rios acessos mostraram letalidade frente ? Artemia salina, atividade antioxidantes e anticolinester?sica. Os resultados obtidos indicam que os extratos dos acessos cont?m constituintes ativos que apresentam tais atividades, confirmando seu potencial biol?gico. Como resultado dos estudos preliminares os acessos BGA- 739, BGA-804, BGA-820 e h?brido HE foram selecionados para continua??o dos estudos, sendo realizado os testes biol?gicos. O acesso BGA-804, fra??es clorof?rmio (CL) e acetato de etila (ACT) demonstrou ter alto potencial antioxidante representado pela EC50 em ?g.ml-1 804-CL (203,5?0,528) e 804-ACT 192,5?0,193, letalidade representado pela LC50 em ?g.ml-1 804-CL (69,3?0,47) e 804-ACT (87,7?0,49) e inibi??o da enzima colinesterase em % 804-ACT (53.230 ? 4,44). O fracionamento, perfil qu?mico por CLAE/DAD e a caracteriza??o qu?mica do acesso BGA-804, resultou na verifica??o da presen?a de compostos fen?licos, flavonoides e no isolamento e identifica??o do daucosterol oriundo da fra??o 804 CL.
