Previous issue date: 2015-03-18 / Anaplasma marginale is a vector-borne pathogen that causes a disease known as anaplasmosis. To date, there are no sequenced genomes of Brazilian strains available. The aim of this work was to compare whole genomes of Brazilian strains of A. marginale (Palmeira and Jaboticabal) with genomes of strains from other regions (North-American and Australian strains). Genome sequencing was performed by next-generation sequencing. Reads were mapped using the genome of the Florida strain of A. marginale as a reference sequence. Identification of single nucleotide polymorphisms (SNPs) and insertions/deletions (INDELs) was performed. Data showed significant differences between the two Brazilian strains, and their comparison with North-American strains (Florida and St. Maries) revealed more nucleotide differences than with Australian strains (Gypsy Plains and Dawn). These results shed light into the evolutionary history of A. marginale and provide the first genome information of South American isolates. Assessing sequences of the genomes of strains from different regions is essential to increase knowledge of the pan-genome of this rickettsia. / Anaplasma marginale ? um pat?geno transmitido por vetores que causa uma doen?a conhecida como anaplasmose. At? o momento, n?o h? genomas sequenciados de cepas brasileiras dispon?veis. O objetivo deste trabalho foi comparar genomas completos de cepas brasileiras de A. marginale (Palmeira e Jaboticabal) com genomas de cepas de outras regi?es (cepas norte-americanas e australianas). O sequenciamento do genoma foi realizado por sequenciamento de alto rendimento. As reads foram mapeadas utilizando o genoma da cepa Florida de A. marginale como uma sequ?ncia de refer?ncia. A identifica??o de polimorfismos de nucleot?deo ?nico (SNPs) e inser??es/dele??es (INDELs) foi realizada. Os dados mostraram diferen?as significativas entre as duas cepas brasileiras, e sua compara??o com as cepas norte-americanas (Fl?rida e St. Maries) revelou mais diferen?as de nucleot?deos do que com as cepas australianas (Gypsy Plains e Dawn). Esses resultados lan?am luz sobre a hist?ria evolutiva de A. marginale e fornecem a primeira informa??o gen?mica de isolados da Am?rica do Sul. Avaliar sequ?ncias dos genomas de cepas de diferentes regi?es ? essencial para aumentar o conhecimento do pangenoma desta rickettsia.
