Previous issue date: 2015-03-27 / CAPES / No Brasil, o descarte de papel representa cerca de 40% do total de dejetos sólidos municipais. Visto que boa parte desses resíduos não é reciclada, e que a composição do papel contém mais de 60% de carboidratos, o papel apresenta um forte potencial para a produção de açúcares. O objetivo deste trabalho foi comparar a hidrólise enzimática de papel de escritório descartado, após pré-tratamento químico (com ácido sulfúrico 1% V/V; 50 rpm) com a hidrólise realizada
após pré-tratamento físico, no qual o papel foi processado em moinho de facas a 10 mesh. Para tal estudo, foi aplicado um planejamento fatorial 2⁴, cujos fatores e níveis inferior e superior foram, respectivamente: tempo da hidrólise (24 e 72 h); carga enzimática (15 e 30 FPU/g celulose); carga de papel (4,5 e 7,5% m/V); e pré-tratamento (químico ou físico). Todas as hidrólises foram realizadas a 50ºC e 150rpm, utilizando as enzimas comerciais Celluclast 1.5L e β-glucosidase Novozymes. As concentrações de celobiose e glicose foram determinadas por meio de cromatografia líquida de alta eficiência e a análise do planejamento pelo software Statistica 7.0. O material foi caracterizado por meio das técnicas de espectroscopia de infravermelho com transformada de Fourier (Fourier Transmission Infrared – FTIR) e Difração de raios X (DRX). Análises estatísticas mostraram melhores resultados para o planejamento cujo material foi quimicamente pré-tratado, atingindo concentração de glicose de cerca de 25 g/L para o processo com 53,5 horas de duração, com carga enzimática igual a 20,8 FPU/g de substrato e 9% de carga de papel. / In Brazil, paper disposal represents about 40% of all municipal solid waste. Since much of this waste is not recycled, and that more than 60% of waste paper is composed by carbohydrates, the paper has great potential for the production of sugars. The objective of this study was to compare the enzymatic hydrolysis of office paper discarded after chemical pretreatment (with sulfuric acid 1% V/V, 50 rpm) with the hydrolysis performed after physical pretreatment, in which the paper was processed in mill to 10 mesh. For this study, an experimental design was applied, whose factors and lower and upper levels were: time of hydrolysis (24 and 72 h); enzyme loading (15 and 30 FPU/g cellulose); paper loading (4.5 and 7.5% w/v); and pre-treatment (chemical or physical). All hydrolyses processes were carried out at 50 °C and 150 rpm using commercial enzymes Celluclast 1.5L and β-glucosidase Novozymes. Concentrations of cellobiose and glucose were determined by high performance liquid chromatography analysis and the experimental design was analysed using the Statistica 7.0. The material was characterized by Fourier transform techniques (Fourier Infrared Transmission - FTIR) and X-ray diffraction (XRD). Statistical analysis showed better results for the material of which has been chemically pretreated, reaching glucose concentration of about 25 g/L to process during 53.5 hours, with enzyme loading of 20.8 FPU/g of substrate and paper loading of 9%.
