Previous issue date: 2009 / Coordena??o de Aperfei?oamento de Pessoal de N?vel Superior (Capes) / As sempre-vivas (Syngonathus spp) esp?cie pertencente ? fam?lia Eriocaulaceae, possui grande interesse comercial e recentemente medicinal para a popula??o da sua ?rea de ocorr?ncia. Com a inclus?o de algumas esp?cies desse g?nero na lista de esp?cies amea?adas de extin??o torna-se necess?rias medidas que garantam a conserva??o da esp?cie seja in situ ou ex situ. Diante disso, a crioconserva??o de sementes ? uma alternativa economicamente vi?vel e que exige um protocolo para cada esp?cie. Dessa forma, o objetivo do presente trabalho foi avaliar a qualidade fisiol?gica de sementes de sempre-viva em rela??o ao efeito do descongelamento ap?s imers?o em nitrog?nio l?quido e a influ?ncia dos diferentes locais e per?odos de armazenamento. A pesquisa foi dividida em duas partes. A primeira parte consistiu em avaliar o efeito do descongelamento em quatro ambientes (freezer, ambiente,banho-maria e microondas) sobre o comportamento fisiol?gico das sementes de sempre-viva Syngonanthus elegans e Syngonanhus arthrotrichus. Os tratamentos foram dispostos em com delineamento inteiramente casualizado e os dados foram submetidos ? an?lise de vari?ncia, com as m?dias comparadas entre si pelo teste de Tukey a 5% de probabilidade. Pelos resultados constatou-se que ambas as esp?cies podem ser crioconservadas. Para as Syngonanthus elegans n?o houve diferen?a estat?stica entre os tratamentos e para a Syngonanthus arthrotrichus o descongelamento em microondas e ? temperatura ambiente proporcionaram os melhores resultados. A segunda parte consistiu em avaliar os locais de armazenamento (nitrog?nio l?quido, geladeira e ambiente) durante quatro per?odos (0, 40, 80 e 120 dias) atrav?s dos testes de vigor (teste de primeira contagem da germina??o e ?ndice de velocidade de germina??o), germina??o, mat?ria fresca, mat?ria seca e comprimento de pl?ntulas. Os tratamentos foram dispostos em arranjo fatorial (3X4), com delineamento inteiramente casualizado e os dados foram submetidos ? an?lise de vari?ncia, com as m?dias comparadas entre si pelo teste de Tukey a 5% de probabilidade. Pelos resultados constatou-se que esta esp?cie pode ser crioconservada com sucesso e que todos os m?todos de descongelamento em temperatura ambiente e em microondas consistiram na melhor alternativa. O armazenamento em geladeira apresentou os melhores resultados para as sementes analisadas. Para todos os par?metros avaliados todos os per?odos de armazenamento foram eficientes para as sementes de S. elegans. As sementes de Syngonanthus arthrotrichus mantiveram a qualidade fisiol?gica quando armazenadas em geladeira. / Disserta??o (Mestrado) ? Programa de P?s-Gradua??o em Produ??o Vegetal, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2009. / ABSTRACT The Sempre-viva (Syngonathus spp.) Species belonging to the family Eriocaulaceae, has great commercial interest for medical and recently the people of their area of occurrence. With the inclusion of some species of this genus in the list of endangered species it is necessary measures to ensure the conservation of the species is in situ or ex situ. Thus, the cryoconservation seed is a viable alternative and requires a protocol for each species. Thus, the purpose of this study was to evaluate the physiological quality of seeds of sempre-viva on the effect of thawing after immersion in liquid nitrogen and the influence of different places and periods of storage. The research was divided into two parts. The first part was to assess the effect of thawing in three environments (freezer, environment and water bath) on the physiological behavior of seeds of sempre-viva Syngonanthus elegans and the effect of the period (0, 40, 80 and 120 days) under the local storage (liquid nitrogen, refrigerator and environment), through tests of force test (first count of the germination rate and speed of germination), germination, green mass, dry weight and length of seedlings. Treatments were arranged in a factorial arrangement, with a randomized design and data were submitted to analysis of variance, compared with the averages among themselves by Tukey test at 5% probability. By the results it was found that this species can be successfully criokept and that all methods of thawing studied in this work, may be used. The research was divided into two parts. The store in freezer showed the best results for seeds analyzed .
