Orientador: Carlos Rossa Júnior / Banca: Denise Madalena Palomari Spolidorio / Banca: Cassiano Kuchenbecker Rosing / Resumo: O objetivo do estudo foi avaliar "in vivo" o efeito do condicionamento com nicotina ou cotinina sobre a morfologia, densidade e viabilidade de fibroblastos. Na avaliação da morfologia e densidade, também foi determinado se a raspagem mecânica das superfícies condicionadas modifica estes resultados. Foram formados dois grupos experimentais : Apenas condicionamento e Raspagem após condicionamento. No grupo Apenas condicionamento, as amostras foram incluídas em placas para cultivo celular e subdivididas em 3 subgrupos segundo a substância testada: Nicotina, Cotinina e Controle negativo. No primeiro subgrupo cada orifício recebeu 2ml de solução de Nicotina (concentrações de 100ng, 1ug, 100 ug, 1mg/ml). No segundo subgrupo cada orifício recebeu 2 ml de solução de Cotinina (concentrações de 50ng, 100ng, 500ng, 1ug/ml). O terceiro subgrupo foi o controle. Após 24 horas de condicionamento as amostras foram lavadas com tampão fosfato e adicionado a 1ml de meio de cultura + 1 ml de meio de cultura contendo 1x10 células/ml. Para o grupo Raspagem após condicionamento, foram empregadas as mesmas concentrações de nicotina e cotinina descritas. O condicionamento foi realizado da mesma forma seguido de raspagem manual. As amostras obtidas foram fixadas, desidratadas, metalizadas e levadas ao microscópio eletrônico de varredura para exame e fotografias. Para a avaliação da viabilidade foram formados dois grupos experimentais, segundo a dose de nicotina: 0-controle, 10ug, 100ug, 0.5mg, 1mg; e segundo o tempo de condicionamento : 1 e 24 hora. Para a cotinina os valores de concentração e tempo de condicionamento foram: 0-controle, 50ng, 100ng, 500ng, 1ug/ml; e 1, 24 e 48 horas respectivamente. Cada um dos orifícios de uma placa para cultura celular recebeu 1 mL de meio de Eagle, 1 ml de Eagle contendo a nicotina ou cotinina nas diferentes... (Resumo completo, clicar acesso eletrônico abaixo). / Abstract: The aim of this study was to evaluate "in vitro" the effect of nicotine or cotinine, on the morphology, density and viability of fibroblasts. In was also assessed if performing root planing on conditioned root surface would alter the results. Exposure to nicotine or cotinine and Root planning after the exposure to nicotine or cotinine. In the first group, the specimens were set in plates for cellular cultures and divided in three subgroups according to the substance tested: nicotine, cotinine, and control (negative). The third group was control. After 24 hours, the specimens were washed with phosphate buffer and 1ml of a cell suspension containing 1x15 cells/ml was added to each well. For the group of root planning after the exposure there were used the same concentrations of nicotine and cotinine of the first group. The exposure was made with the same procedure of root planning. The specimens obtained were fixed, dehidratated, sputter coated with gold, photographed and observed to the eletronic microscope. For the viability, two experimental groups were formed according to drug dosage: 0- control, 10 ug, 100 ug, 0.5 mg, 1 mg and to drug exposure time 1 and 24 hours for the nicotine group. For the cotinine group, concentration values were: 0- control, 50 ng, 100 ng, 500 ng, and 1 ug/ml, the exposure times were 1, 24 and 48 hours. Twelve well microplates were used. Each well received 1 ml of culture medium containing nicotine or cotinine in different concentrations. One milliliter of culture medium containing 1 x 10 cells/ml was also added to each well.
