Orientador: José Luiz Laus / Banca: Paola Castro Moraes / Banca: Alexandre Pinto Ribeiro / Resumo: Avaliou-se o efeito da instilação local de morfina 1% sobre a expressão do fator de necrose tumoral alfa (TNF-alfa), da interleucina-1 beta (IL-1beta), da prostaglandina E2 (PGE2) e da mieloperoxidase (MPO) e a turbidez do humor aquoso (flaremetria a laser) em olhos de coelhos com uveíte induzida por injeção intravítrea de endotoxina. Empregou-se 30 leporinos da raça Nova Zelândia Branco, separados em 5 grupos (n=6), a saber : basal, grupo controle (GC), grupo morfina (GM), grupo naloxona (GN), grupo morfina-naloxona (GMN). Sob anestesia dissociativa, injetou-se 0,1 mL, contendo 0,2 μg de endotoxina (parede celular de Salmonella typhimurium), na câmara vítrea do olho direito de cada animal para a indução da uveíte experimental. Para a avaliação, realizou-se a flaremetria a "laser" antes da injeção intravítrea de endotoxina (basal), e às 10 e 20 horas da indução da uveítes, ocasião em que os olhos tratados foram enucleados para a quantificação do fator de necrose tumoral alfa (TNF-alfa), da interleucina-1 beta (IL-1beta), da mieloperoxidase (MPO) e da prostaglandina E2 (PGE2) por ELISA. Os dados foram analisados estatisticamente, quanto contagem de fótons no humor aquoso (flaremetria a "laser"), aplicando-se análise de variância não paramétrica de Friedman, seguida do "post hoc test" de Dunn, ao nível mínimo de significância de p>0,05 e quanto à quantificação das citocinas, utilizando-se Análise de Variância de via única, seguida pelo teste de Tukey, nas avaliações entre grupos. Considerou-se o nível mínimo de significância p<0,05. Observou-se, após a indução da uveíte, elevação significativa no nível TNF-alfa e IL-1beta, comparativamente aos olhos basais (p<0,05) em todos os tratamentos estudados (GC, GM, GN, GMN). Os níveis dessas citocinas ... / Abstract: To evaluate the effects of instillation of morphine 1% on the expression of tumor necrosis factor alpha (TNF-alpha), interleukin-1 beta (IL-1beta), prostaglandin E2 (PGE2) and myeloperoxidase (MPO) and the turbidity of aqueous humor (laser flaremetry) in the eyes of rabbits with uveitis induced by intravitreal injection of endotoxin. Were used 30 New Zealand White rabbits, separated into 5 groups (n = 6), as follows: healthy control group (CG), morphine group (GM), naloxone group (GN), morphine-naloxone group (GMN). Under dissociative anesthesia it was injected 0.1 mL containing 0.2 mg of endotoxin (cell wall of Salmonella typhimurium) in the vitreous of the right eye of each animal to induce experimental uveitis. For the evaluation, it was used laser flarimetry before intravitreal injection of endotoxin (baseline) and at 10 and 20 hours after the induction of uveitis, at which time the treated eyes were enucleated for the quantification of tumor necrosis factor alpha (TNF-alpha), interleukin-1 beta (IL-1beta), mieloperoxidase (MPO) and prostaglandin E2 (PGE2) by ELISA. The data were treated statistically, the photon count in the aqueous humor (laser flarimetry), applying Friedman's nonparametric two-way analyses of variance test, then the Dunn's "post hoc test", the minimum level of significance was p> 0.05 and for quantification of cytokines using ANOVA one-way followed by Tukey test in ratings between groups. Considered the minimum level of significance p <0.05. It was observed after induction of uveitis, significant increase in the level of TNF-alpha and IL-1beta compared to baseline eyes (p <0.05) in all treatments (GC, GM, GN, GMN). The MPO activity showed a nonsignificant increase after induction of uveitis.
