contemporâneas pensadas de maneira site specific, investigando se é possível criar um paralelo entre os processos de tradução e os processos de apropriação de um site para criação de um trabalho artístico. Para isso, foram estudados aspectos relevantes na mudança dentro do campo das artes plásticas a partir do final da década de 1950, além de ter sido feito um breve levantamento histórico sobre as questões relativas ao lugar e o aparecimento das práticas site specific. Os problemas no que diz respeito à tradução foram discutidos a partir de um apanhado de teorias relativas ao processo tradutório, bem como estudos de traduções pensadas de maneira livre. O conceito de tradução, portanto, tentou ser ampliado e, para esta pesquisa, foi pensado em campos além da lingüística, trabalhando com diferentes linguagens. Para esta dissertação, considerou-se o tradutor como um criador e assim, foi possível relacionar a tradução com as práticas contemporâneas ditas site specific, atentando-se para o site como um lugar de discurso. Do mesmo modo em que um artista se apropria de um site para pensar no seu trabalho, podendo trabalhar com diferentes linguagens em uma mesma obra, os tradutores também concentram-se em se apropriarem de contextos para criarem suas traduções. A partir desta pesquisa teórica inicial, buscou-se dois artistas contemporâneos - Raquel Garbelotti e Antoni Muntadas - para analisar de que maneira artistas-tradutores, que pensam de modo site specific, criam seus trabalhos.

The Experience of Place in Performance: A Survey of Site-Specific TheatreRorem, Jacob 29 September 2014 (has links)
For practitioners and scholars of site-specific theatre, attempts to understand the relationship between a site and performance have often focused on performance. The many ways a site can inform and enhance the audience's experience of performance has been thoroughly explored, but what about the reverse? How can performance facilitate an experience of place and inform audiences about the value and potential of the places around us? I contend that site-specific performance which privileges place--including its varied histories and meanings--can foster a more thorough consideration of the places we inhabit and equip us to make better decisions about them. This thesis uses three case studies to explore the experience of place in performance and its potential implications. My case studies are Rob Ashford and Kenneth Branagh's production of Macbeth at the 2013 Manchester International Festival, We Players of San Francisco, and PlaceBase Productions of Minneapolis.

Trabalha com a ressignificação de ícones e símbolos para provocar uma reflexão sobre o controle exercido por instituições e ideologias dominantes. Suas proposições artísticas avançam para além da tela e experimentam relações com o espaço e com o espectador para sugerir novas maneiras de olhar o mundo. A partir das reflexões teóricas de Miwon Kwon, Claire Bishop e Nicolas Bourriaud discutimos os nexos entre a especificidade do site e as possibilidades de participação nos trabalhos de Doitschinoff em que verificamos pontos de convergência entre esses eixos, a saber: Temporal (2008), Novo Asceticismo (2009), Brilho do Sol (2011) e Cras do Micélio (2013).

Johnny Drops the Bomb is a site-specific, historical play written to be performed in the Diefenbunker, a bomb shelter built to protect key members of the Canadian government in the event of a nuclear attack. As a practice-as-research project, Johnny Drops the Bomb explores the theories and techniques associated with performing history in situ: aura and atmosphere, empathy and witnessing, embodiment and being-in-the-world, proximity and site-specificity, and upsurges of the Real. As the written component of that practical project, this thesis contextualizes Johnny Drops the Bomb by situating it within these approaches, reflecting on the role that each technique played as I wrote, rehearsed, and ultimately performed it with two other actors.

Site-directed mutagenesis of hydrogenase genes in Azotobacter chroococcumTito, Donald January 1992 (has links)
Accessory hydrogen uptake genes have been identified in a region of the Azotobacter chroococcum genome about 5 kb downstream of the hydrogenase structural genes (hupSL). DNA sequencing has revealed six genes (hupABYCDE) in this region. These genes are probably transcribed in the same direction as hupSL but are probably in a different operon. Mutational analysis had shown that disruption of the hupB, hupY, hupD and hupE genes gives a Hup$ sp-$ phenotype. In the present work additional mutational analysis, using Tn5, a Tn5 -derivative containing a promoterless lacZ gene, and a kanamycin resistance gene, confirms the direction of transcription and the separate nature of the hupABYCDE operon, and extends the region known to be necessary for Hup activity to hupA and possibly to 1.6 kb upstream of hupA.

V-ATPases are enzymes found in all eukaryotic cells. They are organized into a peripheral membrane complex (V1) and an integral membrane complex (V0). VI is responsible for ATP hydrolysis and generates the energy used by Vo to pump protons from the cytosol into the vacuole. Subunit d is a component of Vo possibly located at the interface between V 1 and V. in the V-ATPase complex. We hypothesize that subunit d could be involved in the structural and functional coupling of VI and Vo. This was tested by generating point mutations along the open reading frame of subunit d from yeast. The mutations F94A, H128A, D173A, D217A, D261A, E317A, W325A, E328A and C329A, all in conserved regions of the protein sequence, were characterized by examining their growth phenotype and by assessing their ATPase specific activity, proton transport and V1Vo assembly in purified vacuolar membranes. The mutations E317A, W325A, E328A and C329A had reduced ATPase and proton transport activities. In addition, V1Vo assembly was compromised by the mutation W325A. Our results suggest that residues at the carboxyl-end of subunit d are important for ATPase activity, proton pumping and V1Vo assembly at the membrane. / Department of Chemistry
