Despite the potential and advantages, there are technological barriers that prevent microalgae biodiesel production on a large scale. This research aimed to develop innovative technical and scientific studies to assess the feasibility of using freshwater microalgae as a raw material source for biodiesel production. This study allowed evaluating the biodiesel production from microalgae biomass, from the selection of strains, the production and biodiesel quality analysis, and thus enabling to raise important parameters on the cultivation and production of microalgae biodiesel. This research identified a freshwater microalga with potential for biodiesel production - Choricystis minor var. minor - which has content and fatty acid profile suitable for biofuel production, providing more biodiesel (115%) than soybean, by biomass direct transesterification. In studies of culture medium, was observed that the WC medium is the best for biodiesel production. The BBM medium is most suitable for pigments production, such as chlorophyll and carotenoids. Scale pilot production of Choricystis and microalgal biomass conversion into biodiesel showed that this microalgae presents volumetric productivity about 11 times greater than soybeans, with production estimated at 6 tonnes of biodiesel per hectare per year. A few adjustments are required for produced biodiesel quality parameters can achieve the specification adopted by the ANP. Thus, can be concluded that the biodiesel production using microalgae can become a reality on a large scale. / As microalgas, ricas em lipídios, carboidratos e outros bioprodutos, têm sido consideradas como uma promissora fonte de matéria-prima graxa para a produção de biodiesel. Apesar do grande potencial e das vantagens normalmente apontadas para seu uso, ainda existem entraves científicos e tecnológicos que inviabilizam a produção de biodiesel de microalgas em larga escala. Desta forma, essa pesquisa teve como objetivo elaborar estudos técnico-científicos inovadores que possam contribuir para avaliar a viabilidade do uso das microalgas dulcícolas como fonte de matéria-prima graxa para a produção de biodiesel. Os trabalhos realizados permitiram avaliar a produção de biodiesel a partir da biomassa microalgal desde a seleção de cepas até a produção e análise da qualidade do biodiesel, e dessa forma possibilitou levantar parâmetros importantes sobre o cultivo e a produção do biodiesel de microalgas. Nesse trabalho de tese foi identificada uma microalga dulcícola com potencial para a produção de biodiesel - Choricystis minor var. minor – a qual apresentou teores e perfis de ácidos graxos adequados para a produção desse biocombustível, fornecendo 115% mais biodiesel que a semente de soja, via transesterificação direta da biomassa. Nos estudos sobre meios de cultivos observou-se que o meio WC é o melhor para a produção de biodiesel, em comparação ao meio BBM, sendo este último o mais adequado para a produção de pigmentos, tais como carotenoides e clorofila. A produção em escala piloto da microalga C. minor e a conversão da biomassa microalgal em biodiesel permitiu concluir que esta microalga apresentou produtividade volumétrica cerca de 11 vezes superior à soja, com a produção estimada em 6 toneladas de biodiesel por hectare ao ano. Alguns poucos ajustes são necessários para que todos os parâmetros de qualidade do biodiesel produzido possam alcançar a especificação adotada pela ANP. Dessa forma, conclui-se que a produção de biodiesel utilizando microalgas pode vir a ser uma realidade em grande escala.

This thesis deals with interrogation of minors in course of criminal proceedings. In particular, the aim of the thesis is to basically describe differences between interrogations of minors and interrogation of adults categorised by individual procedural positions. The text focuses on interrogation of youngsters less than 18 years old in the position of witnesses. The thesis also discourses upon interrogation of minor perpetrators of criminal activity under the Juvenile Justice Act (in Czech zákon o soudnictví ve věcech mládeže) and takes into account provisions of the Criminal Victims Act (in Czech zákon o obětech trestných činů) focusing on right of interrogated victims on protection from secondary victimisation. The first chapter introduces point of view of individual branches of science in respect of the described topic of the thesis. The chapter determines fundamental terms used in the thesis, in particular the following terms: minor, interrogation and testimony. The characteristics of the possible procedural positions of minors follows, as the procedural position of the interrogated person strongly influences the choice of interrogation tactics. My master thesis primary focuses on proceedings in matrimonial cases and its related issues. Precisely, it deals with the marriage itself and it is supported by individual proceedings of marriage authorization and other specific status matrimonial case proceedings. The largest part of my thesis is however dedicated to divorce due to arising negative impacts divorce has on humanity worldwide and it is considered as one of the most recent and most discussed issue nowadays. The goal of my thesis is to provide the overview of law changes in individual matrimonial proceedings and more importantly their characters, supported by both theoretical and practical examples. While exploring above mentioned problematic, at the same time I engage in substantive law as well as in procedural law due to their mutual cohesion. For the sake of the differentiation of proceeding in matrimonial cases, text of this work is divided into four main chapters, where every chapter includes several subchapters to specify the concrete proceeding's character and its process. Guardianship and other forms of care of minors Abstract The objective of this thesis is to analyse the legal possibilities in problematic situations when parents are not capable of caring for their children due to objective or subjective barriers on their sides. These possibilities are enshrined in the second part, Title III., § 928 - § 975 of the Civil Code of the Czech Republic. In order to derive recommendations for the Czech legal system, particularly with regard to the work overload of judges for minors in the Czech Republic, these regulations will subsequently be compared to the legal situation in France which is mainly determined by the Code civil and the Code de l'action sociale et des familles. The comparison namely includes critical juxtapositions of tutorship, curatorship, entrusting a child to the care of another person, foster care and placing a child in the children's home. Hereby, the content, purpose and requirements of the legal bases for these areas in both countries are presented and contrasted. Thus, the differences are highlighted, evaluated and analysed with regard to the effectiveness and the level of protection of rights which is assured in the countries. Production and Purification of Synthetic Minor Ampullate Silk Proteins

Spider silks are incredible natural materials that have a wide variety of properties that can rival or outperform even common synthetic materials like Nylon and Kevlar. As nature’s architects, orb-weaving spiders spin seven different silks that are used for very specific roles throughout the spider’s lifecycle. These silks are comprised of proteins called spidroins. Each of these spidroins has evolved to have properties such as strength and/or stretch that make these silks successful and highly adapted in their designated roles in web construction, prey capture and reproduction. This study involves the production of minor ampullate silk by genetically modifying the bacteria Escherichia coli. Minor ampullate is a lesser studied silk that is used for the first spiral of the orb web. This spiral is a template that the spider uses to finish the web and provides stability during the web construction. Minor ampullate silk is strong, however it does not stretch so it may be well-suited for certain applications such as ballistic materials. By producing and purifying different arrangements of minor ampullate silk protein, it is possible to learn how this protein can be expressed without using the spider itself. This investigation sheds light on how deviations in the protein sequence and motif arrangement can produce different properties, which can potentially be used to make new materials.

Conquering through Non-Conformity: Answering Christ’s Call to Counterculturalism

The book of Revelation reflects a story of the past that speaks to the present. The Apocalypse presents a discourse that highlights a variety of Christians in Asia Minor who must decide whether they will remain faithful in their allegiance to Jesus Christ or demonstrate unfaithfulness to Christ and his kingdom by conforming to the majority culture by engaging in practices of the broader society that stand in contradiction to their faith. When faced with such a dilemma, various people, including Christians, respond in a variety of ways. For some, their responses bring glory to God while others bring disgrace to the name of the God they say they serve. Writers often discuss Christ’s confrontation and judgment of the Roman Empire. Few, if any, address Jesus’s confrontations with the seven churches of Asia Minor. This study combines Norman Fairclough’s theory of Critical Discourse Analysis and Jay Lemke’s theory of intertextuality to demonstrate that Rev 1-3, the incorporation of concepts and ideas appearing in other resources, the further development of the Apocalypse, and the historical context in which John composed the work present the kingdom of God as an alternative empire to which he belongs and commits himself as a citizen in resisting the power, practices, and allure of Rome. Though the assemblies to which he writes experience a variety of social, political, and religious circumstances, the messages to the churches come together to reveal that Christians, no matter their power or position in society, have a mandate to live as citizens of Christ’s kingdom who maintain a countercultural stance for the sake of the gospel. John depicts Christ as king of this alternative empire, who confronts each of his communities regarding their allegiance to him, demonstrated by their non-conformity to the practices of the broader culture regarding how they function in the various power structures in which they live. He then uses the later visions of the Apocalypse to depict Christ’s vindication of those who maintain a countercultural posture in relation to the Roman Empire and highlights the advantages and blessings of non-conformity, while noting the disadvantages and curses of conformity. Throughout the history of the Christian faith, various segments of the church have faced opposition from the surrounding culture. Christians today maintain the same mandate as the early church to resist the allure of the worldly powers that surround them and remain loyal to Jesus.
