This diploma thesis deals with personalities of the Keybord Department at the Academy of Performing Arts in Prague in the second half of the 20th century. This work focuses on making united and systematic summary of professors at the Academy in the first decades of working at the Piano Department. Author of this paper is trying to have a complex attitude to these personalities. They were not only great teachers but also active interpreters, composers, authors of didactic works and they made revisions of the piano compositions. All the articles are in chronological order that depends on the beginning of teacher´s academic working. Chapters are also divided into three basic parts that connect people of the same generation. Part that deals with the brief history of the Academy should help readers to get the wider context. This chapter includes historical and political information that are more than important for the total meaning of the thesis. Some of these happenings had an important influence on all the spheres of life. The purpose of the thesis is to show the situation in the community of pianists in Prague after the Second World War in a well-arranged way. The work is supposed to be an integrated overview for an informed reader and should give information about lives, performances and legacy of the greatest pianists of the Academy of Performing Arts in the post-war period.

The possibility to lattice matching GaAsPN with silicon makes this semiconductor interesting for studies. In this dissertation we investigated the optical and magneto optical properties of semiconductors nanostructures of the type GaP(N)/GaAsPN. Mesures of photoluminescence (PL), photoluminescence excitation (PLE) and magneto photoluminescence (Magneto-PL) under high fields (B≤ 15T) were performed in films of GaAsPN and multiple quantum wells (MQW) of GaAsPN/GaPN. We have studied localizations effects with measures of diamagnetic shift, stoke shift and the dependence of photoluminescence peak with temperature. We observed a negative diamagnetic shift for some samples, which is an anomalous effect in these systems. It was also seen a red shift of the PL peak when the MQW samples suffered a thermal treatment. Analyzing the spin polarization properties of this material, utilizing magneto photoluminescence resolved with circular polarization, we observed polarization of the samples as high as 30% on fields of 15T. / Ligas semicondutoras III-V nitreto diluídas possuem propriedades físicas que as tornam interessantes para aplicações em dispositivos optoeletrônicos. A possibilidade de casar liga de GaAsPN com silício faz com que esse semicondutor se torne interessante para estudos. Nesta dissertação investigamos as propriedades ópticas e magneto ópticas de nanoestruturas semicondutoras do tipo GaP(N)/GaAsPN. Foram realizadas medidas de fotoluminescência (PL), fotoluminescência de excitação (PLE) e magneto fotoluminescência (Magneto-PL) sob altos campos magnéticos (B ≤ 15T), em filmes finos de GaAsPN e múltiplos poços quânticos (MQW) de GaAsPN/GaPN. Estudamos efeitos de localização dos portadores através da análise do deslocamento diamagnético da PL, deslocamento Stoke e a dependência da posição do pico de fotoluminescência com a temperatura. Verificamos um deslocamento diamagnético negativo para algumas das amostras, o que é um comportamento anômalo nesse tipo de sistema. Foi verificado também um deslocamento para o vermelho do pico de PL quando realizado tratamento térmico nas amostras de MQWs. Analisando as propriedades de polarização de spin desse material, utilizando magneto-PL resolvida em polarização circular, foi observada uma polarização circular de até 30% num campo de 15 T.

Amostras de Semicondutores III-V foram crescidas usando a técnica de Epitaxia por feixes Moleculares. As propriedades elétricas das estruturas de GaAs com dopagem planar com Silício foram investigadas, e também a saturação e a difusão do Silício nestas amostras. As propriedades ópticas e elétricas das estruturas dopadas planarmente com Selênio foram analisadas, usando as técnicas de Capacitância-voltagem e a de Tunelamento resonante. As propriedades elétricas dos poços quânticos a base de InGaAs/GaAs foram investigadas, em função da posição da impureza planarmente dopada com Silício. / III-V semiconductor samples were grown using the Molecular beam epitaxy technique, the electrical properties of the GaAs structures planar doped with silicon were investigated as well as the Silicon saturation and diffusion in these samles. The optcal and electrical properties of structures planar doped with Selenium were analyzed using the Capacitance Voltage and resonant Tunneling techniques. The electrical properties of InGaAs/ GaAs based quantum wells were investigated as a function of the planar doped with Silicon impurity position.

Estudo da influência de elementos transponíveis nos genomas das algas C. reinhardtii e V. carteri / Influence of transposable elements in the genomes of C. reinhardtii and V. carteri algae

Elementos transponíveis (TEs) são sequências de DNA que possuem a capacidade de transposição no genoma hospedeiro. O principal objetivo deste trabalho reside na investigação em torno de possíveis contribuições de TEs nos genomas das algas C. reinhardtii e V. carteri, mais especificamente, na arquitetura dos genes ortólogos nestas espécies. Neste contexto, análises em sílico em larga escala foram realizadas, buscando-se identificar associações entre TEs e os genes ortólogos. Os resultados indicaram que os genes em C. reinhardtii tendem a acumular mais cópias de TEs em relação aos seus ortólogos em V. carteri. C. reinhardtii apresentou maior densidade de cópias de TEs para as regiões flanqueadora 5´ , flanqueadora 3´ e intrônica quando comparada a V. carteri; o inverso foi verificado quando analisada a densidade de TEs nas regiões codificantes. Análises para apurar a distribuição dos elementos em regiões intergênicas e intragênicas foram estabelecidas, nas quais a frequência observada dos elementos foi comparada à frequência esperada segundo a distribuição randômica de TEs no genoma, simulada computacionalmente. Foram constatadas regiões em que a presença dos elementos encontra-se significativamente abaixo do esperado, a exemplo de intervalos adjacentes ao início e ao término dos genes, o que provavelmente reflete a seleção negativa de eventos de integração nestas delimitações, em virtude dos efeitos deletérios associados à disrupção de estruturas de regulação da expressão gênica. De forma geral, nas regiões flanqueadoras 5´ e 3´, foi identificada a tendência de elevação da frequência padronizada de TEs à medida que a classe de distância avaliada se distancia do início e do término do gene, respectivamente. A baixa representatividade dos elementos também foi constatada em regiões intragênicas. O estudo da distribuição de TEs nos íntrons dos genes ortólogos indicou a preservação destas regiões quanto à fixação de TEs, sendo a representatividade abaixo do esperado mais evidente em intervalos adjacentes ao éxon, o que minimiza a chance de ruptura no padrão de splicing dos genes. Em sequências codificantes, a escassez de TEs - esperada devido ao provável efeito deletério destes eventos para a função do gene - foi constatada nos ortólogos das duas espécies. No entanto, inovações decorrentes da integração dos elementos em regiões codificantes podem resultar em efeitos evolutivos positivos, embora estes eventos sejam raros. Nas espécies analisadas foram identificados dois casos, de especial interesse, em que um domínio da sequência peptídica encontra-se localizado em região derivada de TE: o primeiro refere-se ao gene Cre06.g262800, em C. reinhardtii, no qual foi identificado o domínio PHD-finger associadao ao elemento Gypsy-5-LTR_CR; o segundo remete ao gene Vocar20001092m.g, em V. carteri, no qual o domínio zinc knuckle foi reconhecido em região derivada do elemento Gypsy3-LTR_VC. Estes genes constituem exemplos da contribuição de TEs na evolução de sequências codificadoras nas espécies C. reinhardtii e V. carteri, corroborando a hipótese de que os TEs podem contribuir na evolução da arquitetura dos genes, apesar do efeito disruptivo inerente à integração dos mesmos em regiões gênicas. / Transposable elements (TEs) are DNA sequences able to transpose in the host genome. The aim of this study resides in the investigation of TEs contributions in the algae C. reinhardtii and V. carteri genomes, more specifically in the architecture of orthologous genes in these species. In this context, large scale in silico analysis were performed to identify associations between TEs and orthologous genes. The results indicated that genes in the C. reinhardtii specie tend to accumulate more TEs copies than orthologous genes in V. carteri. C. reinhardtii showed higher density of TEs copies in the 5´ flanking, 3´ flanking and intronic regions when compared to V. carteri; the opposite was observed in coding regions. Investigation of the elements distribution in the intergenic and intragenic regions was performed, in which the observed TE frequency was compared to expected TE frequency from the simulated random distribution of the elements in the genome. It was verified regions where TE frequency was significantly lower than expected, as in gene boundaries adjacencies, probably reflecting a negative selection of the TE integration events in these delimitations due to deleterious effects associated with disruption of gene regulatory structures. In general terms, it was observed an increasing standardized frequency in the 5´ and 3´ flanking regions as the distance from gene start and gene end, respectively, increases. TEs underrepresentation was also verified in the intragenic regions. The study of TEs distribution in the introns of orthologous genes revealed the preservation of these structures in relation to TEs fixation, with a stronger underrepresentation near exon, which minimizes the chance of gene splicing pattern disruption. In the coding sequences, the TEs scarcity - expected due the likely deleterious effects to gene function - was verified in the orthologous of both species. However, in rare instances, innovations mediated by TEs integration in the coding regions can lead to positive evolutionary effects. In the species analyzed two instances of particular interest were observed, in which the domain of peptide sequence is located in the region derived from TE. The first one refers to the Cre06.g262800 gene, in the C. reinhardtii specie, which has a PHD-finger domain associated with Gypsy-5-LTR_CR element. The second one refers to the Vocar20001092m.g gene, in V. carteri, in which the zinc knuckle was recognized in region derived from Gypsy3-LTR_VC element. These genes are examples of TEs contributions in the evolution of coding sequences in the C. reinhardtii and V. carteri species, corroborating the hypothesis that TEs can contribute to the evolution of gene architecture, despite the inherent disruptive effect in their integration in the gene regions.

O conhecimento atual das trombofilias permite a associação com cerca de 40% dos casos de TEV. Há controvérsias quanto ao valor da pesquisa laboratorial de trombofilia para o benefício dos doentes com tromboembolismo venoso. Procuraram-se as variáveis preditivas para a pesquisa positiva de trombofilia e avaliar o impacto desses resultados nas decisões clínicas. Foram avaliados 84 doentes consecutivos com TEV confirmado por métodos de imagem no período entre janeiro de 2001 e novembro de 2003. Após o período previsto de anticoagulação definido por critérios clínicos, os doentes foram submetidos à pesquisa das principais causas de trombofilia. Os resultados laboratoriais permitiram a dois examinadores independentes reavaliar caso-a-caso a indicação de ¿mudança de conduta¿, caracterizada pela interferência no tempo de profilaxia secundária ou ¿atenção especial¿ para medidas de maior vigilância diante de situações de risco ou para a extensão da pesquisa aos familiares assintomáticos. A trombofilia foi encontrada em 35 dos 84 casos (41,66%), sendo que em 27 (32,12%) havia uma causa genética. O fator V Leiden foi a alteração mais freqüente (15,47%), seguida do conjunto de deficiência dos anticoagulantes naturais (11,9%). Não houve diferença significativa da freqüência de trombofilia relacionada à faixa etária nem diferença de idade de aparecimento do primeiro evento trombótico entre doentes trombofílicos e não trombofílicos. Houve significância estatística para ocorrência de trombofilia nos doentes com tromboflebite superficial, recorrência e na associação com fatores de risco não cirúrgicos. A ¿mudança de conduta¿ foi atribuída a 6 dos 84 doentes (7,14%), estatisticamente significativa para aqueles com recorrência em relação aos que tiveram apenas um episódio de TEV. A ¿atenção especial¿ foi atribuída a 34 dos 84 casos (40,47%).A tromboflebite superficial de aparecimento espontâneo, a ocorrência de TEV relacionada a causas não cirúrgicas e a recorrência foram os principais achados preditivos de trombofilia. A ¿mudança de conduta¿ aplicou-se a uma pequena porcentagem de doentes, e refletiu predominantemente a confirmação da necessidade de prolongamento da profilaxia secundária. A ¿atenção especial¿ diante de situações de risco e a extensão da profilaxia primária a familiares de primeiro grau assintomáticos expostos a situações de risco parecem-nos a melhor indicação para a pesquisa laboratorial da trombofilia. Nowadays its possible to associated VTE with a cause of thrombophilia in about 40% of patients. There are some inconclusive points about the real benefit of the laboratorial investigation on thrombophilia for patients with VTE. We tried to identify the variables that point to the positive test results and the impact of these results on clinical decisions.The screening for the more common causes of thrombophilia was applied to 84 consecutive patients with VTE confirmed by image examination between January 2001 and November 2003. After test results, two independent observers evaluated, in a case by case basis, the indication of a ¿change on prophylaxis¿, in order to modify the period of anticoagulant intake (secondary prophylaxis), or ¿special attention¿ when considering to have a higher medical surveillance before risk situations or for the extension of the research to the first degree asymptomatic relatives.Thrombophilia was found in 41.66% (35/84), and in 32.12% (27/84) it involved agenetic cause. The factor V Leiden was the more prevalent alteration, identified in 15.47% of the cases, followed by the natural anticoagulants disfunction (11.9%). There was no significative difference of thrombophilia frequency between ages, nor a difference of age in the onset of the first thrombotic event between thrombophilic and non-thrombophilic patients. There was a higher prevalence of thrombophilia in patients with superficial thrombophlebitis of spontaneous onset, in cases of recurrence and when associated with non-surgical predisponent factors. The ¿change on prophylaxis¿ resulted in 7.14% (6/84), and there was statistically significance for patients with recurrent episodes when compared to patients with just one. The ¿special attention¿ was applied in 40.47% (34/84). Spontaneous superficial thrombophlebitis, occurrence of VTE related to nonsurgical causes and recurrence, were the main findings which suggested thrombophilia. The ¿change of prophylaxis¿ was applied to a small percentage of patients. The ¿special attention¿ for risk situations and the extension of the primary prophylaxis to the asymptomatic family members seem to be the best indication for the laboratorial research on thrombophilia. Investigando o efeito da temperatura na dinâmica de crescimento de nanofios de GaP encontramos uma grande variedade de nanofios em cada amostra, caracterizadas por diferentes direções de crescimento e/ou morfologias, apresentando sempre uma população dominante. Com o aumento da temperatura (510°C) observamos uma transição drástica na morfologia da população dominante, de nanofios típicos para uma nova e inesperada morfologia assimétrica. Mostramos ainda que modificando o tamanho da nanopartícula catalisadora de 5nm para 20nm os nanofios assimétricos ainda são favorecidos a alta temperatura. Procurando compreender o mecanismo de formação dos nanofios assimétricos, mostramos que estas estruturas cristalizam-se na fase WZ, com baixa densidade de defeitos, comparadas às outras amostras. Além disso, mostramos que a assimetria destes nanofios não é oriunda de diferenças de polaridade nas facetas laterais ou formação de defeitos cristalográficos que pudessem modificar a dinâmica de crescimento de modo a levar à morfologia assimétrica. Desta forma, propomos um cenário de crescimento simplificado, no qual a estrutura assimétrica é formada pela combinação do crescimento de nanofios ordinários, via VLS, junto com estruturas que crescem livre de catalizador via mecanismo VS (Vapor-Sólido), estas duas estruturas então se juntam, transferindo material e dando lugar a facetas de menor energia, resultando na formação da estrutura assimétrica. Por fim, medidas de fotoluminescência mostram a emissão característica no verde do GaP WZ para os nanofios assimétricos, mesmo com a estrutura não estando passivada, confirmando a boa qualidade cristalina das nossas amostras / Abstract: In this work, we study the growth dynamics of GaP nanowires grown by VLS (Vapor-Liquid-Solid) method via Chemical Beam Epitaxy (CBE) using gold nanoparticles as catalyst. Investigating the effect of temperature on the growth dynamics of GaP nanowires we find a wide variety of nanowires in each sample, characterized by different growth directions and/or morphologies, always having a dominant population. With increasing temperature (510°C), we observed a dramatic transition in the morphology of the dominant population of typical nanowires to a new and unexpected asymmetric morphology. We also show that modifying the size of the catalyst nanoparticle from 5nm to 20mn the asymmetric nanowires are still favored at high temperature. Looking forward to understand the mechanism of formation of the asymmetric nanowires, we show that these structures crystallize in the WZ phase with low defect density when compared to the other samples. Furthermore, we show that the asymmetry of these nanowires is not derived from differences in polarity in side facets or the formation of crystallographic defects which might modify the growth dynamics so as to bring the asymmetric morphology. Thus, we propose a simplified growth scenario, in which the asymmetric structure is formed by growth combination of ordinary nanowires via VLS, along with structures that grow catalyst-free via VS (vapor-solid) mechanism, these two structures joined and by transferring materials facets with lower energy facets appear, resulting in the formation of asymmetrical structure. Finally, photoluminescence measurements show the characteristic emission in the green of WZ GaP for asymmetric nanowires, even with no passivation, which confirms the good crystalline quality of our samples / Mestrado / Física / Mestre em Física

InfluÃncia do fator V de Leiden e da mutaÃÃo g20210a no gene da protrombina no desenvolvimento de eventos trombÃticos no MunicÃpio de Fortaleza / Influence of factor v leiden and prothrombin g20210a mutation gene in the development of thrombosis at Fortaleza city

CoordenaÃÃo de AperfeiÃoamento de NÃvel Superior / CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior / RESUMO As doenÃas trombÃticas constituem um sÃrio problema de saÃde pÃblica. Diversas desordens hereditÃrias e ambientais, que afetam o sistema fisiolÃgico anticoagulante, estÃo atualmente estabelecidas como fatores de risco para a ocorrÃncia do evento trombÃtico. Dentre os fatores hereditÃrios, as mutaÃÃes G1691A do gene do fator V e G20210A do gene da protrombina sÃo os mais freqÃentes. A associaÃÃo entre estas alteraÃÃes genÃticas e a ocorrÃncia de eventos trombÃticos desencadeou o desenvolvimento de diversas pesquisas. Neste estudo, 189 pacientes portadores de eventos trombÃticos, atendidos no ambulatÃrio de Hematologia do Centro de Hematologia e Hemoterapia do Cearà - HEMOCE/SESA/UFC, foram analisados para a detecÃÃo da presenÃa das mutaÃÃes G1691A do gene do fator V e G20210A do gene da protrombina. O grupo controle consistiu de 349 voluntÃrios. A freqÃÃncia encontrada na populaÃÃo controle foi de 2% (7/349) para a mutaÃÃo do fator V e 1,7% (6/349) para a mutaÃÃo da protrombina, enquanto que nos pacientes trombofÃlicos a freqÃÃncia destas mutaÃÃes foi de 9% (17/189) e 2,1% (4/349), respectivamente. Dentre os fatores hereditÃrios, apenas a mutaÃÃo do fator V foi significante (p<0,001). Considerando os fatores ambientais de risco, o tabaco, idade > 40 anos e sexo feminino apresentaram significÃncia estatÃstica (p<0,001). Os riscos foram estimados em anÃlises pareadas e nÃo pareadas para o fator V de Leiden (4,8; 5,3; 9,8), tabaco (17,6; 14,9; 33,3), idade idade > 40 anos (2) e sexo feminino (3,7 e 4,1). Os fatores de risco para eventos trombÃticos no Cearà foram tabagismo, idade > 40 anos, sexo feminino, e a mutaÃÃo G1691A do fator V.foram associados com o desenvolvimento de trombose no estado do CearÃ. / Thrombotic diseases are a serious problem for public health. Several hereditary and environmental factors, that affect physiological anticoagulant system, have been nowadays well established as risk factors for thrombosis. Among hereditary factor V Leiden and prothrombin G20210A mutation are the most frequents. The association between several modifications on factor V gene and prothrombin gene in the development of thrombotic events has brought about future searches. In this study, 189 patients with thrombosis attended at the Hematology and Hemoterapy Center of Cearà state âHEMOCE/ Brazil, were analyzed to find out the presence of factor V Leiden and prothrombin G20210A mutation The control group was made up 349 healthy volunteers. In this study, the frequency found of factor V Leiden the control population was of 2% (7/349) and in the patients was 9% (17/189) while the frequency found of prothrombin G20210A mutation the control population was of 1,7% (6/349) and in the patients was 2,1% (4/189). Among hereditary factors only factor V Leiden was significant statistic (p<0,001). Among environmental factors studied, tabagism, age > 40 anos and femele were significant statistic (p<0,001). The ODDS RATIO of the risk factors with significant statistic were factor V de Leiden (4,8; 5,3; 9,8), tabaco (17,6; 14,9; 33,3), age > 40 years old (2) and female (3,7 e 4,1). Our results demonstrate that factor V Leiden, tabagism, age > 40 years old and female were associated with development trombosis in CearÃ.

Context: Requirements engineering (RE) and Verification and validation (V&amp;V) areas are treated to be integrated and assure successful development of the software project. Therefore, activation of both competences in the early stages of the project will support products in meeting the customer expectation regarding the quality and functionality. However, this quality can be achieved by aligning RE and V&amp;V. There are different practices such as requirements, verification, validation, control, tool etc. that are followed by organizations for alignment and to address different challenges faced during the alignment between RE and V&amp;V. However, there is a requisite for studies to understand the alignment practices, challenges and factors, which can enable successful alignment between RE and V&amp;V. Objectives: In this study, an exploratory investigation is carried out to know the impact of factors i.e. RE process and organizational factors during the alignment between RE and V&amp;V. The main objectives of this study are: To find the list of RE practices that facilitate alignment between RE and V&amp;V. To categorize RE practices with respect to their requirement phases. To find the list of RE process and organizational factors that influence alignment between RE and V&amp;V besides their impact. To identify the challenges that are faced during the alignment between RE and V&amp;V. To obtain list of challenges that are addressed by RE practices during the alignment between RE and V&amp;V. Methods: In this study Systematic Literature Review (SLR) is conducted using snowballing procedure to identify the relevant information about RE practices, challenges, RE process factors and organizational factors. The studies were captured from Engineering Village database. Rigor and relevance analysis is performed to assess the quality of the studies obtained through SLR. Further, a questionnaire intended for industrial survey was prepared from the gathered literature and distributed to practitioners from the software industry in order to collect empirical information about this study. Thereafter, data obtained from industrial survey was analyzed using statistical analysis and chi-square significance test. Results: 20 studies were identified through SLR, which are relevant to this study. After analyzing the obtained studies, the list of RE process factors, organizational factors, challenges and RE practices during alignment between RE and V&amp;V are gathered. Thereupon, an industrial survey is conducted from the obtained literature, which has obtained 48 responses. Alignment between RE and V&amp;V possess an impact of RE process factors and organizational factors and this is also mentioned by the respondents of the survey. Moreover, this study finds an additional RE process factors and organizational factors during the alignment between RE and V&amp;V, besides their impact. Another contribution is, addressing the unaddressed challenges by RE practices obtained through the literature. Additionally, validation of categorized RE practices with respect to their requirement phases is carried out. Conclusions: To conclude, the obtained results from this study will benefit practitioners for capturing more insight towards the alignment between RE and V&amp;V. This study identified the impact of RE process factors and organizational factors during the alignment between RE and V&amp;V along with the importance of challenges faced during the alignment between RE and V&amp;V. This study also addressed the unaddressed challenges by RE practices obtained through literature. Respondents of the survey believe that many RE process and organizational factors have negative impact on the alignment between RE and V&amp;V based on the size of an organization. In addition to this, validation of results for applying RE practices at different requirement phases is toted through survey. Practitioners can identify the benefits from this research and researchers can extend this study to remaining alignment practices.
