The traditional approach of Quality by Testing (QbT) limits the assurance of product quality to in-process and post-production testing. To overcome these limitations, a more proactive and systematic means to product development and optimisation is required. Quality by Design (QbD) is an example of such an approach which focuses on understanding the product and its manufacturing process and emphasises that quality should be built into the product and not merely tested. The study aims to optimise ethionamide tablets, an immediate release oral solid dosage form using QbD.

Stock Market Crashes & the Effect on Security Prices of the Pharmaceutical Industry

Class of 2006 Abstract / Objectives: To examine the influence stock market crashes have on pharmaceutical security prices. More specifically, the objectives are to quantify the stock market abnormal returns and volatility of domestically listed drug companies during periods of stock market crashes. Methods: An event study methodology was performed to determine the impact of stock market crashes on security prices of pharmaceutical firms. Pharmaceutical security price data was obtained from the Center for Research in Securities Prices (CRSP) database. Stock market crashes were identified and economic considerations regarding the nature of security price returns were reviewed including normality, autocorrelation, heteroscedasticity and cross sectional dependence. Results: The estimation period for the study ranged over a period of thirty-five months (-45 to -5) prior to the stock market crash. Mean estimates of the SIMM mean beta parameter ranged from 0.51-1.18 for all companies analyzed within the period of 1929-2001. Mean monthly abnormal returns ranged from 0.0039-0.0348 during the estimation period. Over a period of one, three, and five months the majority of the pharmaceutical industry failed to consistently produce above average abnormal returns. Conclusions: The purpose of this study was to investigate the performance of the pharmaceutical industry during ten stock market crashes to verify and or quantify current literature statements about the recession proof nature of the drug sector. The current investigation found that during the estimation periods surrounding the stock market crashes, the pharmaceutical industry did not outperform the average market return.

Cancer remains the second major cause of death in the world. Thus, there is a pressing need to identify potential synthetic route for the development of novel anticancer agents which will serve as lead compounds to effectively combat this life-threatening epidemic. Pyrrolo[2,1-c][1,4]benzodiazepines (PBDs) have sparked a great interest as lead compounds because of their cancerostatic and anti-infective properties. The twisted molecular structure of PBD analogs provides both helical and chiral elements. In an effort to expand novel PBDs that interact with the key exocyclic amino group of the DNA-guanine base, we hypothesized that construction of a fused cyclic active system, would likely serve as an electrophilic site when compared to traditional electrophilic C11-N10 imine group. To examine our theory, we report herein the synthesis and cell viability/cytotoxicity of a series of PBD analogs using NCI-60 cell lines screening. Thus, compounds 1–13 were synthesized and fully characterized. The selected PBDs were found to have marginal inhibition of growth, up to 30%, for certain cell lines.

<p>Heavy water moderated thermal nuclear fission reactors have a greater inherent neutron economy than light water or graphite moderated reactor designs. Consequently, such units, operating on a variety of fuel cycles, may play an ever-increasing role in meeting future global energy demands. This thesis explores, analytically, the operational advantages and challenges associated with the use of a low enriched uranium (LEU) fuel cycle in advanced reactors based on the CANDU heavy water moderated, pressure tube design concept. The flexibility afforded through the use of LEU fuel is applied to enhance the operational and safety characteristics of reactors utilizing this fuel cycle. An investigation of factors influencing coolant void reactivity is conducted. Design modifications are introduced to reduce the coolant void reactivity, while maintaining the continued capability of high power operation. An enrichment and element radius graded fuel bundle design is developed with a central graphite core, an inner ring of 14 fuel elements, and an outer ring of 21 fuel elements. Fuel and lattice design perturbations are investigated to examine the effect of lattice pitch variations, capability of radioisotope production, the use of burnable poisons, and light water coolant. Xenon override requirements with LEU fuel are addressed. The efficacy of using modified two group (M-2) neutron diffusion theory for LEU fuel management studies is investigated. A modelling strategy is developed for the simulation of reactivity devices and fuel lattice properties using the M-2 methodology with a fixed energy cut-off. Detailed fuel management studies are conducted to examine the operational intricacies of LEU fuelling. Improved checkerboard type fuelling strategies are developed. Finally, the CANDU - Spectral Shift Advanced Thermal Reactor (CANDU-SSATR) is introduced and characterized. This multi-spectrum high burnup advanced reactor design utilizing simplified fuel management strategies holds great promise for the future.</p> / Doctor of Philosophy (PhD)
