Fahrenheit 451, de Ray Bradbury e de François Truffaut : da alienação pós-moderna à oralidade homérica

Este trabalho analisa o Fahrenheit 451 de Ray Bradbury e o de François Truffaut, em sua proposição de utilizar a oralidade (discurso oral) como forma de manutenção do literário e resistência à imposição e à censura ideológica. Efetua também uma análise comparatista das Utopias Negativas do século XX (obras Distópicas). Ao longo deste projeto, é feita uma análise do surgimento da escrita (alfabético-fonética), e sua estreita relação com o discurso oral. Tenta-se reproduzir a trajetória traçada pelo discurso oral, passando pelo desenvolvimento da tecnologia escrita, assim como a produção cultural tanto no meio oral quanto no escrito, junto com suas conseqüências e influências sobre pensamento humano. Da oralidade dos poemas homéricos à oralidade advinda com o desenvolvimento de aparelhos eletrônicos como o telefone, que, em plena modernidade, voltam a valorizar o discurso oral. Neste projeto a oralidade é vista como algo cíclico. / This thesis analyses Ray Bradbury’s and François Truffaut’s Fahrenheit 451, in their proposal of using the oral discourse as a way of maintenance of the literacy as well as resistance to ideological imposition and censorship. It also brings a comparative analysis of the Negative Utopias of the twentieth century (Distopic Literature), along with the rise of writing (alphabetic-phonetic writing), and its close relation to oral discourse. It tries to reproduce the path trailed by oral discourse, passing through the development of writing technology, as well as the cultural production both in the oral and in the writing media, along with its consequences and influences to human thought. From Homer’s oral poems to the oral discourse emerged with the development of electronic devices such as the telephone, which enhance oral discourse in modern times. In this project oral discourse is seen as something cyclical.

This project discusses the ways in which Ray Bradbury’s novel Fahrenheit 451 functions as an indictment of media culture. While many analyses of the novel focus on the text’s sweeping themes of literary censorship, this study instead centers on Bradbury’s depiction of media—particularly television—culture and the ways in which Bradbury feared it could be harmful. Although Bradbury wrote about a future society a century beyond his own, his novel serves as a remarkable reflection of his contemporaneous culture’s media consumption and gendered divisions; this thesis discusses Bradbury’s novel alongside such forces, considering the effects such influences may have had on his work.

Effet de l'acide valproïque sur l'hématopoïèse : rôle du réseau de régulation "microARN/ facteurs de transcription" / Effect of valproic acid on hematopoiesis : role of regulatory network “microRNA / Transcription factor”

L’acide valproïque (VPA) est un inhibiteur des histones désacétylases (HDACi), qui présente des propriétés anti-tumorales nsur différents types de cancers. Son utilisation depuis plusieurs décennies comme médicament antiépileptique a révélé des effets secondaires, notamment sur le système hématopoïétique. Dans la présente étude, nous nous sommes intéressés à l’effet du VPA sur les réseaux microARN (miR)/Facteurs de transcription (FT) spécifiquement impliqués dans la régulation des voies de différenciation érythro-mégacaryocytaires. Nous montrons que le VPA est capable d’inhiber la différenciation érythroïde dans les cellules érythroleucémiques humaines TF1 et K562 et dans les cellules souches hématopoïétiques (CSH) CD34+, stimulées par l’érythropoïétine recombinante (Epo) ou par l’aclacinomycine A. Cette inhibition se traduit par une diminution de l’expression de la glycophorine A, de la γ-globine, des miR-144/451 et du FT GATA-1. L’inhibition du pré-miR-144 suggère que le VPA est capable de réguler l’expression du gène miR-144/451 au niveau transcriptionnel, via GATA-1. Dans les cellules Epo/CD34+, le VPA induit l’augmentation du FT PU.1 en accord avec l’inhibition du miR-155 et favorise son interaction avec GATA-1 pour inhiber son activité. L’utilisation d’un analogue du VPA, sans activité HDACi (Valpromide) et d’un inhibiteur d’HDAC de classe I, le MS-275, a montré que l’activité HDACi du VPA n’est pas requise pour l’inhibition de la différenciation érythroïde. Le VPA affecte également la voie mégacaryocytaire issue d’un progéniteur commun aux cellules érythroïdes. Dans la lignée mégacaryoblastique Meg-01, le VPA induit des modifications morphologiques du type mégacaryocytaire, une augmentation du marqueur CD61, du FT GATA-2 et du miR-27a. En revanche, l’expression du FT GATA-1 et des miR-144/451 diminuent. L’augmentation du miR-27a coïncide avec la diminution de l’expression de l’ARNm du FT RUNX1, en accord avec l’induction de la voie mégacaryocytaire. En conclusion, le VPA est capable de moduler le programme de différenciation érythro-mégacaryocytaire, à travers un micro réseau de régulation miR/FT / Valproic acid (VPA), a histone deacetylase inhibitor (HDACi), exhibits anti-cancer properties against several tumor types. Its use as an anti-epileptic drug for several decades reveled side effects at the hematological level. In this study, we analyzed the effect of VPA on an erythro-megakaryocyte-specific miR/transcription factors network. VPA inhibited erythroid differentiation in the erythroleukemia cell lines TF1 and K562 as well as in CD34+/hematopoietic stem cells (HSCs), induced by the recombinant erythropoietin (Epo) or aclacinomycin. This inhibition was characterized by glycophorin-A, γ-globin and GATA-1/miR-144/451 down-regulation. Inhibition of pre-miR-144 expression suggested that VPA regulates transcription of the miR-144/451 gene through GATA-1. In Epo-stimulated HSCs, VPA induced PU.1 expression in correlation with miR-155 inhibition and promoted GATA-1/PU.1 interaction. The use of valpromide, a VPA analogue without HDACi activity and the class-I HDACi MS-275, showed that HDAC inhibition by VPA was not required for its inhibitory activity on erythropoiesis. VPA also induced megakaryocyte features in Meg-01 cells, at both cellular and molecular levels. Notably, CD61, GATA-2 and miR-27a were over-expressed. RUNX1 mRNA expression and GATA-1/miR-144/451 axis decreased in accordance with megakaryocyte differentiation. In conclusion, VPA is able to modulate erythro-megakaryocytic differentiation program, through a regulatory micro-network involving miRs and TFs

Probability Learning In Normal And Parkinson Subjects: The Effect Of Reward, Context, And Uncertainty

In this thesis, the learning of probabilistic relationships between stimulus-action pairs is investigated under the probability learning paradigm. The effect of reward is investigated in the first three experiments. Additionally, the effect of context and uncertainty is investigated in the second and third experiments, respectively. The fourth experiment is the replication of the second experiment with a group of Parkinson patients where the effect of dopamine medication on probability learning is studied. In Experiment 1, we replicate the classical probability learning task by comparing monetary and non-monetary reward feedback. Probability learning behavior is observed in both monetary and non-monetary rewarding feedback conditions. However, no significant difference between the monetary and non-monetary feedback conditions is observed. In Experiment 2, a variation of the probability learning task which includes irrelevant contextual information is applied. Probability learning behavior is observed, and a significant effect is found between monetary and non-monetary feedback conditions. In Experiment 3 / a probability learning task similar to that in Experiment 2 is applied, however, in this experiment, stimulus included relevant contextual information. As expected, due to the utilization of the relevant contextual information from the start of the experiment, no significant effect is found for probability learning behavior. The effect of uncertainty observed in this experiment is a replication of the reports in literature. Experiment 4 is identical to Experiment 2 / except that the subject population is a group of dopamine medicated Parkinson patients and a group of age matched controls. This experiment is introduced to test the suggestions in the literature regarding the enhancement effect of dopamine medication in probability learning based on positive feedback conditions. In Experiment 4, probability learning behavior is observed in both groups, but the difference in learning performance between Parkinson patients and controls was not significant, probably due to the low number of subject recruited in the experiment. In addition to these investigations, learning mechanisms are also examined in Experiments 1 and 4. Our results indicate that subjects initially search for patterns which lead to probability learning. At the end of Experiments 1 and 4, upon learning the winning frequencies, subjects change their behavior and demonstrate maximization behavior, which makes them prefer continuously one option over the other.
