Vers l’identification des acteurs moléculaires responsables des propriétés mécaniques du bois de tension de peuplier / Toward identifying molecules responsible for the mechanical properties of poplar tension wood

Cette thèse visait à caractériser la composition en polysaccharides des fibres de bois de tension chez le peuplier et établir de possibles corrélations avec les mécanismes de création de la tension. A cette fin, nous avons étudié le bois de jeunes peupliers qui ont poussé sous conditions contrôlées. Nous avons pu déterminer la composition en polysaccharides des couches G isolées ainsi que la structure de ces polysaccharides. L’évolution de cette composition au long de la différentiation du bois de tension et du bois opposé a été évaluée à l’aide de 178 anticorps dirigés contre les principaux polysaccharides pariétales. La fonction de deux protéines à arabinogalactanes avec domaine fascicline-like a été également étudiée à l’aide des anticorps produits à cet effet. La génétique inverse a été également utilisée pour compléter l’étude de la fonction d’une de ces protéines dans la formation du bois de tension. Les résultats obtenus ont mis en évidence une évolution de la composition de la couche G au cours de la différenciation. Des différences entre la composition de la couche S2 des fibres du bois de tension et du bois opposé ont aussi été détectées. Une grande quantité de pectines de type RG-I a été décelé dans la couche G, molécules qui pourraient participer à la formation d’un gel. Le gonflement de ce gel serait responsable de la mise en tension des microfibrilles de cellulose. Cependant, nos travaux ne montrent aucune évidence pour la présence de xyloglucanes dans la couche G. Des indices en faveur de l’implication des FLAs dans la construction de la couche G ont été également trouvés dans cette étude. Ce travail de thèse ouvre des perspectives pour l’identification du déterminisme moléculaire à l’origine de la création de la tension dans le bois de tension. / This work aimed at characterizing the composition in polysaccharides of poplar tension wood fibres and a possible correlation with mechanisms for creating tension. Firstly, we isolated G-layers from tension wood formed in young poplars which grows in a greenhouse under controlled conditions and determine its polysaccharides composition and the structure of these polysaccharides. Therefore, we investigate the polysaccharide distribution during tension and opposite wood fibres differentiation by using 178 antibodies raised against the major wall polysaccharides. The distribution and function of two fasciclin-like arabinogalactan proteins were studied using two antibodies produced to this end and the function of one of the proteins were studied by reverse genetics. This work show an evolution in the polysaccharides composition of G-layer through its differentiation and also differences concerning S2 layer composition between tension and opposite wood fibres. No evidences for the presence of xyloglucans in the G-layer. However, this work shows the presence of a high quantity of RG-I type pectin which may be implicated in a gel-like structure formation which swelling could be responsible for tension creation in cellulose microfibrils. Our results suggest an implication of fasciclin-like arabinogalactan proteins in G-layer construction. Further, this work opens up new perspectives towards identification of molecular basis of tension creation in tension wood.

Há muitos agentes terapêuticos comercializados sob forma racêmica. Os enantiômeros podem apresentar diferenças significativas nos perfis farmacocinético e farmacodinâmico. O uso do enantiômero puro em formulações farmacêuticas pode resultar em melhor ajuste de dose e menos efeitos adversos. A atropina, um alcalóide natural da Atropa belladonna, é a mistura racêmica de l-hiosciamina e d-hiosciamina. Este fármaco é principalmente utilizado para dilatar a pupila e como um antiespasmódico. Para quantificar estes enantiômeros, foram desenvolvidos métodos analíticos utilizando a cromatografia líquida de alta eficiência (CLAE) com as fases estacionárias quirais Chiralcel-OD® e Chiral-AGP®. Para quantificar estes enantiômeros em soluções oftálmicas, foi realizada a validação com sucesso de um método por CLAE, empregando uma coluna Chiral-AGP®, a 20°C. A fase móvel foi uma solução tampão fosfato (contendo 10 mM de 1-octanosulfonato de sódio e 7,5 mM de trietilamina, ajustada para pH 7,0 com ácido fosfórico) e acetonitrila (99:1 v/v). A vazão foi de 0,6 mL/min, com detecção ultravioleta em 205 nm. No intervalo de concentração de 14,0 µg/mL a 26,0 µg/mL, o método é linear (r > 0,9999), exato (100,1% - 100,5%) e preciso (RSDsistema ≤ 0,6%; RSDintra-dia ≤ 1,1%; RSDentre-dias ≤ 0,9%). O método é específico e os testes de validação asseguram que as soluções de padrão e amostra são estáveis até 72 horas. O planejamento fatorial assegura a robustez com variação de ±10% nos componentes da fase móvel e 2°C na temperatura da coluna. / There are many therapeutic agents commercialized under racemic form. A New nutritional Approach for promoting Gut Health and Animal Performance

Il fattore antisecretivo (AF) è una proteina secreta nel plasma e nei tessuti dei mammiferi, che ha dimostrato di essere un potente inibitore dell’ipersecrezione intestinale e dell'infiammazione. Dopo il bando degli AGP, diversi approcci nutrizionali finalizzati all’ottimizzazione della fase di transizione dello svezzamento e alla riduzione delle malattie intestinali sono stati proposti, ma con scarsi risultati rispetto agli antibiotici. Le diete in grado di indurre la secrezione endogena di AF potrebbero essere una valida alternativa agli AGP. Si ritiene che AF contenuto nel colostro e nel latte delle scrofe possa essere un fattore di protezione contro la diarrea nei suinetti. E’ stato dimostrato che è possibile incrementare il livello di AF nel plasma attraverso la dieta, sia nell’uomo che negli animali, con l’ingestione di cereali sottoposti ad un particolare processo idro-termico (HPC). In questa tesi è stato studiato l’effetto dell’aggiunta di HPC alla dieta di suinetti svezzati, sulle performance e sullo stato infiammatorio dell’epitelio intestinale. La supplementazione della dieta con HPC ha migliorato ADG e FCR durante tutto il periodo sperimentale. I valori I-FABP, considerato un parametro del danneggiamento della mucosa intestinale, nel plasma dei suinetti sono risultati bassi in tutti i gruppi e non influenzati dalla dieta. L'attività antisecretiva di AF-16, il dominio attivo della proteina, è stata valutata mediate su colture cellulari IPEC-J2 trattate con tossina colerica (CT) in Ussing chamber. AF-16 non ha inibito l’incremento di Isc indotto da CT. E’ stato osservato un ridotto incremento di Isc durante la somministrazione contemporanea di AF-16 e CT. L'attività anti-infiammatoria di AF-16 è stata studiata in colture di macrofagi RAW 264,7, trattati con LPS, e su macrofagi alveolari di suino stimolati con PMA. AF-16 ha ridotto la produzione di NO nei macrofagi stimolati con LPS in funzione del dosaggio, con maggiori effetti a dosaggi più elevati. L’effetto antiinfiammatorio di AF-16 non è stata confermata dai ROS test, neanche usando dosi elevate di peptide. I risultati confermano l’efficacia dei cereali HPC come promotori della crescita nei suinetti. Ulteriori studi in vitro sono necessari per capire il meccanismo d’azione di AF. / The antisecretory factor (AF) is a protein secreted in plasma and other tissue fluids in mammalians which was shown to be a potent inhibitor of intestinal fluid secretion and inflammation. After the AGP ban, several nutritional approaches aimed to the optimization of the weaning transition and reduction of gut diseases have been proposed, but the success rates are really low compared to antimicrobials. AF-inducing diets appear to be suitable alternatives to AGP. Indeed AF content in sows’ colostrum and milk appears to be a protection factor against diarrhoea in suckling piglets. Increased AF level in plasma by dietary means, such as feeding hydro-thermally processed cereals (HPC), has been demonstrated in human and animals. We tested the effect of two different HPC level of inclusion in a wheat-barley based diet on weaned piglets growth performance, reared in an experimental farm. The results confirmed the efficacy of HPC as growth promoters in piglet nutrition: HPC supplementation improved ADG and FCR. In vitro test were made to study the antisecretory and anti-inflammatory properties of the AF protein and its mechanism of action by using AF-16, the active region of the protein. The Ussing chamber experiments performed on polarized IPEC-J2 cells confirmed the neuronal involvement in the antisecretory activity of AF. In fact, AF-16 did not inhibit CT-induced Isc. A slower rate of Isc increase was observed during the simultaneous administration with AF-16 and CT. High dosages of AF-16 were found to reduce the LPS-stimulated NO production in RAW264.7 cell, thus supporting the hypothesis of an anti-inflammatory action. On the contrary, no significant results were obtained on PMA-stimulated ROS generation in pig alveolar macrophages.
