Vasopressin in preeclampsia

Preeclampsia is a devastating disorder of pregnancy characterized by high blood pressure, proteinuria, headache, renal glomerular endotheliosis, multi-organ system failure, and fetal and maternal demise. As reviewed in Chapter I, not much is known about the pathogenesis of preeclampsia, contributing to a lack of biomarkers and treatments for the disease. In Chapter II, we review arginine vasopressin, a circulating neuropeptide hormone with important fluid balance and cardiovascular actions. Vasopressin binds to numerous receptors throughout the body to elicit its effects and is associated with various disease states. In Chapter III, we discuss evidence for vasopressin as an etiology of preeclampsia. Specifically, that vasopressin secretion is elevated very early in pregnancies affected by preeclampsia and infusion of vasopressin into pregnant C57BL/6J mice causes physiological features similar to those seen in preeclampsia. In Chapter IV, we show that vasopressin administration during mouse pregnancy models specific subtypes of preeclampsia through time- and receptor-specific mechanisms. The role of angiotensin 1a receptors on vasopressin-producing cells in fluid homeostasis is shown in Chapter V and their role in metabolism is depicted in Chapter VI. Overall, we conclude that vasopressin is an important mediator of features of preeclampsia and that angiotensin 1a receptors on vasopressin-producing cells are important for normal fluid homeostasis.

NMDA receptor-dependent signalling pathways regulate arginine vasopressin expression in the paraventricular nucleus of the rat

Yes / The antidiuretic hormone arginine vasopressin (AVP) regulates water homeostasis, blood pressure and a range of stress responses. It is synthesized in the hypothalamus and released from the posterior pituitary into the general circulation upon a range of stimuli. While the mechanisms leading to AVP secretion have been widely investigated, the molecular mechanisms regulating AVP gene expression are mostly unclear. Here we investigated the neurotransmitters and signal transduction pathways that activate AVP gene expression in the paraventricular nucleus (PVN) of the rat using acute brain slices and quantitative real-time PCR. We show that stimulation with l-glutamate robustly induced AVP gene expression in acute hypothalamic brain slices containing the PVN. More specifically, we show that AVP transcription was stimulated by NMDA. Using pharmacological treatments, our data further reveal that the activation of ERK1/2 (PD184352), CaMKII (KN-62) and PI3K (LY294002; 740 Y-P) is involved in the NMDA-induced AVP gene expression in the PVN. Together, this study identifies NMDA-mediated cell signalling pathways that regulate AVP gene expression in the rat PVN. / Supported by a generous donation from Jonathan Feuer.

O núcleo hipotalâmico supraquiasmático (SCh), apresenta caracterização neuroquímica com duas subpopulações principais de células, a produtora de polipeptídeo intestinal vasoativo (VIP) e argenina vasopressina (AVP). As fibras IR AVP e VIP oriundas do SCh apresentam características morfológicas específicas que possibilitam o rastreamento a longas distâncias dentro do hipotálamo. No presente estudo buscamos mapear os terminais IR VIP e AVP nas áreas hipotalâmicas do primata Cebus apella utilizando a imuno-histoquímica, e fazer a identificação das áreas hipotalâmicas recipientes do SCh, utilizando o mapeamento da distribuição das fibras IR associado à análise morfológica destas duas substâncias neuroativas. As fibras IR VIP e AVP com características do SCh foram identificadas em porções anteriores como hipotálamo anterior, área pré-óptica, área hipotalâmica lateral, SPZV, até porções mais caudais, porção retroquiasmática, área tuberal. Os resultados indicam um padrão similar de distribuição de fibras IR VIP e AVP nas áreas do hipotálamo e também em áreas descritas como recipientes das projeções do SCh, quando comparado com outras espécies como roedores descritos na literatura. / The suprachiasmatic nucleus (SCN) of the hypothalamus, contains a variety of different neurons that tend to form two major subpopulations within the nucleus, the vasoactive intestinal peptide (VIP) and vasopressin (VP). The immunoreactive (IR) fibers derived from the VIP and VP IR cells of the SCN present morphological characteristics that allow their specific tracking in long distances within the hypothalamus. In the present investigation we aimed map VIP and VP IR terminals in hypothalamic areas of the primate Cebus apella using immunohistochemistry, and to do identification of hypothalamic recipient areas from SCN using the mapping distribution of fibers IR associated with morphological analysis of these two neuroactive substances. VIP and VP IR fibers with characteristics from SCN were identified in the rostral anterior hypothalamic area and medial preoptic area, laterally to the lateral hypothalamic area, and more caudally in SPZV and retrochiasmatic tuberal area. The results indicate that there is a similarity in the pattern of distribution of VIP and VP fibers in the hypothalamic areas and also in areas recipients from SCN projections when compared with nocturnal rodent species described in the literature.

Arginine vasopressin and adrenocorticotropin secretion in response to psychosocial stress is attenuated by ethanol in sons of alcohol-dependent fathers

Familial risk and environmental stress promote the development of alcohol dependence. We investigated whether a positive family history of alcoholism affects the neuroendocrine response to a standardized laboratory stress test in healthy subjects without alcohol use disorders. Twenty-four high-risk subjects with a paternal history of alcoholism (PHA) and 16 family history negative (FHN) controls were evaluated. Psychosocial stress was induced by having subjects deliver a 5-min speech and mental arithmetics in front of an audience on separate days, after drinking either placebo or ethanol (0.6 g/kg) in a randomized sequence. Adrenocorticotropin (ACTH) was measured in 10 plasma samples covering up to 75 min after the stress test. Plasma arginine vasopressin (AVP) was determined before the stressor, at the time of maximum ACTH secretion, and at 75 min after stress onset. The stress test induced a phasic increase in ACTH secretion. At the time of maximum ACTH, AVP was significantly increased in relation to baseline. Compared to placebo, alcohol administration significantly attenuated maximum ACTH concentration in PHA but not FHN subjects, and decreased AVP measured in the same samples in PHA but not FHN subjects. We conclude that activation of the hypothalamic–pituitary–adrenal system by psychosocial stress is accompanied by an increase in peripheral plasma AVP levels. Secretion of both ACTH and AVP suggest that alcohol attenuates the stress response selectively in PHA but not FHN subjects. This might imply some short-term positive alcohol effect in sons of alcoholics, but also constitute a mechanism by which their risk to develop alcohol use disorders is increased.

Etude des propriétés fonctionnelles de variants de polymorphisme du récepteur V1B de la vasopressine détectés dans les troubles affectifs / Functional properties of V1B vasopressin receptor polymorphism detected patients with affective disorders

La sécrétion de vasopressine (AVP) et de corticolibérine (CRH) déclenchent la sécrétion d'ACTH via leurs récepteurs V1B et CRF1, puis celle des catécholamines et des corticostéroïdes, résultant dans les effets physiologiques induits par le stress. Trois mutations résultant de variants non-synonymes affectant le récepteur V1B (K65N, R364H et G191R) ont été associées chez l'homme à des troubles psychiatriques tels que la phobie, l'hyperactivité et le comportement suicidaire. Nous avons étudiés les propriétés pharmacologiques de ces variants V1B qui pourraient être la cause de ces pathologies. Après construction de chacun des plasmides codant pour chaque variant, nous les avons exprimés dans des cellules HEK293 et avons établi les caractéristiques de chaque récepteur telles que son affinité de liaison, ses couplages à la PLC, à la MAP kinase et la mobilisation du Ca2+. Nous avons observé que le variant R191 avait un couplage à la PLC et à la MAP kinases fortement augmenté (+50%), alors que les autres variants montraient un baisse de ces couplages. Les différences n'étant pas dûes à des constantes d'activation (Kact) pour l'AVP différents. L'association avec les β-arrestines n'est pas équivalente pour tous les variants, le R191 montrant une forte association aux β-arrestines 1 et 2 alors que le variant N65 ne s'associe plus. Ces données indiquent que le déficit en ERK-phosphorylée peut être dû à la fois à une baisse de la production d'Inositol-P et de DAG via la voie Gq et à une baisse de signalisation directe via les β-arrestines. Nous avons également examiné les propriétés d'internalisation de ces variants en utilisant des lignées cellulaires stables exprimant chacun des récepteurs couplés à une EGFP. Nous avons observé, par microscopie confocale, que la cinétique et le taux d'internalisation étaient similaires pour tous les variants. Par contre, la voie du traffic intracellulaire est modifiée pour certains. Les variants possédant la mutation C terminale H364 (H364 et N65/H364) sont préférentiellement adressés à la voie de dégradation lysosomiale alors que les autres utilisent la voie classique de recyclage endosomial comme le récepteur sauvage, ainsi que le révèlent les marquages spécifiques de ces compartiments. En conclusion, les résultats obtenus durant cette thèse révèlent que les variants du récepteurs V1B trouvés chez des patients atteints de troubles affectifs ont des propriétés pharmacologiques de couplage et de trafic intracellulaire modifiées. Ces mutations pourraient participer à une plus grande vulnérabilité des patients au stress. Ainsi, elles pourraient être recherchées, à titre de marqueurs génétiques, chez des patients considérés comme à risque.Cette thèse translationnelle a été financée par un contrat doctoral CHRU/UM1. / The release of vasopressin (AVP) and corticoliberin (CRH) triggers the release of ACTH by the mean of V1B and CRF1 receptors, releasing catecholamines and corticosteroids and regulating stress. Genetic modifications of either element may produce severe consequences. Three non-synonymous variants of the V1B receptor gene were found in patients with phobia, hyperactivity or suicidal conducts (respectively K65N, R364H and G191R). To explore the pharmacological properties of these V1B variants, we generated and expressed in HEK293 cells cDNA constructs of each variant. We evaluated ligand affinity, PLC coupling, MAP kinase activation, Ca2+ mobilization. The capacity of R191 variant to accumulate inositol phosphates and to activate MAP kinase under AVP activation was higher as compared to N65 and H364 variants. However, all variants exhibited the same Kact for AVP. V1B variants were not similar in β-arrestin interaction either, with N65 variant impaired for this function, indicating that the deficit in ERK phosphorylation measured previously may result of 2 additional mechanisms, one due to a lower IP/DAG production with impaired coupling to Gq, and the other due to a decrease in β-arrestin interaction. Finally, we compared internalization properties using stable cell lines expressing EGFP–tagged V1B variants and confocal microscopy. First, we observed no difference neither in the internalization kinetics nor in amplitude (70% of total receptors internalized). However, H364 variant showed a different trafficking as compared to K65/G191/R364 (“wt”) and N65, with less cytoplasmic recycling endosomes and more lysosomial addressing as revealed by Lamp1 co-labelling.Altogether, these data reveal that V1B receptor variants in phobic and suicidal patients display modified coupling and/or desensitization processes.
