Dietary accumulation of hexabromocyclododecane diastereoisomers in juvenile rainbow trout (Oncorhynchus mykiss): bioaccumulation/depuration parameters and evidence of bioisomerization

The major objectives of this research were to examine the bioaccumulation parameters [depuration rates (kd), half life (t1/2) and biomagnification factor (BMF)] of individual isomers of hexabromocyclododecane (HBCD, C12H18Br6) in fish and to test the hypothesis of in vivo bioisomerization. This was done by exposing three groups of juvenile rainbow trout (Oncorhynchus Mykiss) to food fortified with known concentrations of an individual diastereoisomer (α, β, γ) for 56 days (uptake phase) followed by 112 days (depuration phase) of unfortified food. A fourth group of fish were exposed to unfortified food for the duration of the experiment. Fish (n=4) from all four aquaria were sacrificed on days 0, 7, 14, 56, 63, 70, 112 and 168 and muscle tissue was extracted and analyzed for diastereoisomer concentrations by high performance liquid chromatography tandem mass spectrometry (LC/MS/MS). Bioaccumulation of the γ diastereoisomer was linear during the uptake phase while the α and β diastereoisomers were found to increase exponentially with respective doubling times of 14.1 and 20.5 days. Both the β and γ diastereoisomers followed first order depuration kinetics with calculated t1/2’s of 94 ± 25 and 84 ± 51 (± 1 × standard error) days, respectively. The BMF for the α diastereoisomer (BMF = 4.1) was one and a half times greater than the β-diastereoisomer (BMF = 2.6) and about one fifth larger than the γ-diastereoisomer (BMF = 3.6). The large BMF for the α diastereoisomer is consistent with this diastereoisomer dominating higher trophic level organisms in wildlife. Although the BMF of the β diastereoisomer suggests that it will biomagnify, because it is present in small quantities in commercial mixtures it is rarely detected in environmental samples. Results from these studies also provide evidence of bioisomerization of the β and γ diastereoisomers. Most importantly, the α diastereoisomer which was recalcitrant to bioisomerization by juvenile rainbow trout in this study and known to be the dominant diastereosiomer in fish, was bioformed from both the β and γ diastereoisomers. To our knowledge, this is the first report of bioisomerization of a halogenated organic pollutant in biota.

Paleoecological and Carbon Accumulation Dynamics of a Fen Peatland in the Hudson Bay Lowlands, Northern Ontario, from the Mid-Holocene to Present

Pollen assemblages, peat humification and carbon:nitrogen stratigraphy were examined at high resolution in a core from a fen peatland in the Hudson Bay Lowlands, Northern Ontario, to interpret the factors that drive long-term peatland dynamics. Subtle changes in the vegetation community are evident over the record, suggesting both allogenic and autogenic influences, but a fen community appears to have been resilient to external perturbations including isostatic rebound and hydroclimatic changes between 6400 and 100 years BP. Paleoclimatic reconstructions from the fossil pollen assemblages indicate that precipitation increased 3000 years BP at the end of the Holocene Thermal Maximum, and that carbon accumulation in the fen was controlled more by effective surface moisture (precipitation) than by temperature. The pollen record suggests changes over the past century, including increases in shrub Betula, Alnus, Ambrosia, and Cyperaceae and a decrease in Sphagnum spores, consistent with the observed Pan-Arctic shrub increase.

Cette recherche est une invitation à repenser ce(ux) qui constituent l’humain et de manière plus large le vivant en s’intéressant à la manière dont le non-humain a priori inanimé, ici la matière plastique, dans son couplage avec l’humain, devient force d’animation. Je pose la question suivante : comment est-ce que l’introduction du medium plastique reconfigure le tissu de relations composant le territoire et ainsi un mode d’existence humaine ? À l’occasion d’un terrain multi-ligne qui a eu lieu entre Ottawa et Montréal, j’ai suivi la circulation du pétrole ainsi que sa signification dans sa concrétion plastique. Je me suis mise à l’écoute des bruits émergeant d’une f(r)iction pétromoderne et suis moi-même entrée dans un devenir-visqueux en interférant avec le milieu pour le révéler. Je suggère dans ce texte que le régime plastique est un régime de transparence, d’abstraction, qui se soustrait aux regards et reconfigure la response-abilité à la matière.

Potential Impacts of Timber Harvesting, Climate, and Conservation on Sediment Accumulation and Dispersal in the South Slough National Estuarine Reserve, Oregon

Accurate sediment flux histories are critical data for deciphering the relative importance of climate and land use factors such as logging and road construction on sediment production and deposition. We use 210Pb activities derived from sediment cores taken on the tidal flats of the South Slough of the Coos Bay estuary to establish temporal variations in sediment accumulation rates. We determined that average deposition varied between 0.4 and 0.81 cm/yr based on two ~80 cm sediment cores. Sedimentation accumulation rates approached 2.1 cm/yr during the 1960s when a rainfall event of extreme intensity coincided with vigorous timber activity. Following this peak, a >40% reduction in peak lumber harvests in the latter part of the 20th century was accompanied by a decrease in sedimentation rates. Mean monthly rainfall during the same time period remained seasonably constant, indicating that land use is likely the key factor governing variations in sediment accumulation.

Identification of Proteins Involved in Salinity Tolerance in Salicornia bigelovii

With a global growing demand in food production, agricultural output must increase accordingly. An increased use of saline soils and brackish water would contribute to the required increase in world food production. Abiotic stresses, such as salinity and drought, are also major limiters of crop growth globally - most crops are relatively salt sensitive and are significantly affected when exposed to salt in the range of 50 to 200 mM NaCl. Genomic resources from plants that naturally thrive in highly saline environments have the potential to be valuable in the generation of salt tolerant crops; however, these resources have been largely unexplored. Salicornia bigelovii is a plant native to Mexico and the United States that grows in salt marshes and coastal regions. It can thrive in environments with salt concentrations higher than seawater. In contrast to most crops, S. bigelovii is able to accumulate very high concentrations (in the order of 1.5 M) of Na+ and Cl- in its photosynthetically active succulent shoots. Part of this tolerance is likely to include the storage of Na+ in the vacuoles of the shoots, making S. bigelovii a good model for understanding mechanisms of Na+ compartmentalization in the vacuoles and a good resource for gene discovery. In this research project, phenotypic, genomic, transcriptomic, and proteomic approaches have been used for the identification of candidate genes involved in salinity tolerance in S. bigelovii. The genomes and transcriptomes of three Salicornia species have been sequenced. This information has been used to support the characterization of the salt-induced transcriptome of S. bigelovii shoots and the salt-induced proteome of various organellar membrane enriched fractions from S. bigelovii shoots, which led to the creation of organellar membrane proteomes. Yeast spot assays at different salt concentrations revealed several proteins increasing or decreasing yeast salt tolerance. This work aims to create the basis for Salicornia research by providing a genome, transcriptomes, and organellar proteomes, contributing to salinity tolerance research overall. We identified a set of candidate genes for salinity tolerance with the aim of shedding some light on the mechanisms by which this plant thrives in highly saline environments.
