Orientador: Luiz Claudio Di Stasi / Resumo: O termo Doença Inflamatória Intestinal (DII) compreende duas doenças, a Retocolite Ulcerativa (RCU) e a Doença de Crohn (DC), caracterizadas por inflamação crônica que compromete o trato gastrointestinal. Estudos mostram que o estresse oxidativo é um dos fatores determinantes para o desenvolvimento da DII. A terapêutica atual não proporciona cura permanente, demonstrando a necessidade de novas estratégias terapêuticas. A espécie Bertholletia excelsa (Lecythidaceae), popularmente conhecida como castanha-do-pará, é uma árvore nativa da Floresta Amazônica; de seu fruto retira-se a castanha, semente oleaginosa com alto teor de ácidos graxos, selênio e outros componentes como vitaminas e minerais, que podem apresentar atividade farmacológica associada a efeitos benéficos. Devido à sua composição, o objetivo do presente estudo foi avaliar a atividade anti-inflamatória da castanha-do-pará como potencial terapia adjuvante para o tratamento da DII. Ratos Wistar receberam por via oral cinco doses do óleo (18,75; 37,5; 75; 150 e 300 mg/kg) durante 5 dias consecutivos. Após o tratamento, foi feita indução do processo inflamatório por meio da administração via retal de 0,25 mL de solução contendo 10 mg de TNBS em etanol a 50% (v/v). Após 72h da indução do processo inflamatório, os animais foram mortos e os cólons retirados para análises macroscópicas (tamanho e escore da lesão, relação peso-comprimento colônico) e bioquímicas tais como atividade das enzimas mieloperoxidase (MPO) e fosfata... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Inflammatory Bowel Disease (IBD) is a term that comprises two disorders, Ulcerative Colitis (UC) and Crohn’s Disease (DC), which are chronic inflammation that compromise the gastrointestinal tract. Studies show that the oxidative stress is one of the many factors involved in DII pathogenesis. The pharmacological treatment doesn’t bring cure, highlighting the necessity of new therapeutical strategies. The species Bertholletia excelsa Bonpl. (Lecythidaceae), popularly known as Brazil nut, is a native tree from the amazon rainforest. From its fruit is removed a oilseed with high contents of fatty acids, selenium and another components such vitamins and minerals, that can produce pharmacological activity associated with beneficial effects. Due to its composition, the objective of this project was to evaluate the anti-infammatory activity of the Brazil nut oil as a potential adjuvant therapy for the treatment of IBD. Male wistar rats received orally five dosis of oil (18,75; 37,5; 75; 150 e 300 mg/kg) through five consecutive days. After treatment the inflammatory process was induced by rectally administration of 0,25 mL of a solution containing 10 mg of TNBS diluted in ethanol 50% (v/v).
