Influência do resveratrol sobre metabolismo energético, estresse oxidativo e expressão SIRT1 no miocárdio de ratos diabéticos /

Orientador: Ana Angélica Henrique Fernandes / Banca: Amanda Martins Baviera / Banca: Carlos Alan Candido Dias Junior / Resumo: Estudos sugerem que o Diabetes mellitus (DM) promove alterações no metabolismo energético e estresse oxidativo, que constituem fatores importantes para desencadear distúrbios cardiovasculares. A utilização de antioxidantes tem despertado interesse na clínica devido a propriedades terapêuticas sobre as doenças cardiovasculares, podendo, desta forma, evitar a morbimortalidade em pacientes diabéticos. Neste sentido, propõe-se estudar o efeito do resveratrol (RSV - 3,5,4'-trihydroxystilbene) através de parâmetros bioquímicos séricos, metabolismo energético e estresse oxidativo no tecido cardíaco de ratos com diabetes mellitus tipo 1. Foram utilizados 32 ratos, raça wistar, com peso corporal ± 250g. Os animais foram distribuídos nos grupos experimentais (n=8): G1 (normais, controle); G2 (normais, tratados com resveratrol); G3 (diabéticos não tratados); G4 (diabéticos, tratados com resveratrol). O Diabetes mellitus experimental foi induzido através da administração intraperitoneal de estreptozotocina (STZ), em dose única, na concentração de 60 mg/kg de peso corporal. Os animais pertencentes aos grupos G2 e G4 receberam resveratrol, via gavagem, na concentração de 1,0 mg/kg/dia durante 30 dias. No final do período experimental (30 dias) foram avaliados parâmetros nutricionais e determinou-se a glicemia, triacilgliceróis (TG), colesterol total (CT), frações lipoproteicas e atividade da lactato desidrogenase (LDH). No tecido cardíaco foi determinada a concentração de proteínas totais, glicogênio, hidroperóxido de lipídio (HP), glutationa total, glutationa reduzida, glutationa peroxidase (GSH-Px) e superóxido dismutase (SOD), atividade das enzimas marcadoras do metabolismo energético, lactato desidrogenase (LDH), citrato sintase (CS) e β-hidroxiacil hidroxilase (OHADH), além da expressão da proteína situína (SIRT1) através da técnica Western Blotingt. The animals were divided into experimental groups (n = 8) : G1 (normal control), G2 (normal, treated with resveratrol), G3 (diabetic untreated) , G4 (diabetic treated with resveratrol) . The experimental Diabetes mellitus was induced by the intraperitoneal administration of streptozotocin (STZ) in a single dose, at a concentration of 60 mg / kg body weight. The animals in Groups G2 and G4 received RSV via gavage at 1.0 mg/kg/day for 30 days. At the end of the experimental period (30 days), nutritional parameters were evaluated and determined to glucose, triglycerides (TG), total cholesterol (TC), lipoprotein fractions and lactate dehydrogenase (LDH) activity. In cardiac tissue, concentration of total protein, glycogen, lipid hydroperoxide (HP), total glutathione, reduced glutathione, glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) was determined and activity of marker enzymes of energy metabolism, lactate dehydrogenase (LDH), citrate synthase (CS) and β-hydroxyacyl hydroxylase (OHADH), and the expression of situín protein (SIRT1) through Western Blotingt. The experimental design was completely randomized with 4 treatments and 8 replicates, according to analysis of variance (ANOVA).
