Thank you. / The present study was undertaken to determine the attitudes of a selected group of educated women toward cancer of the breast. The pur'pose of this study was to compare the attitudes of four groups of educated women, all of whom have a basic knowledge of cancer as a diseaseo Three important components were perceived as relevant in forming an attitude toward cancer of the breast. These were: 1) Feelings toward the mutilation involved; 2) Feelings toward the curability; 3) Feelings involved in a change of the self concept. The investigators were interested in comparing the attitudes of a group of women with nursing education and a group of women without this specialized education. The second concern was t he comparison of attitudes as manifested by a younger age group and an older age group. Based on the principle that one cannot necessarily determine action from an attitude alone, another concern was a correlation of a woman's attitude toward cancer of the breast and willingness to undertake health action. / 2999-01-01

Yes / Breast cancer is a leading cause of cancer-related deaths. Anemia is common in breast cancer patients and can be treated with blood transfusions or with recombinant erythropoietin (EPO) to stimulate red blood cell production. Clinical studies have indicated decreased survival in some groups of cancer patients treated with EPO. Numerous tumor cells express the EPO receptor (EPOR), posing a risk that EPO treatment would enhance tumor growth, but the mechanisms involved in breast tumor progression are poorly understood. Here, we have examined the functional role of the EPO-EPOR axis in preclinical models of breast cancer. EPO induced the activation of PI3K/AKT and MAPK pathways in human breast cancer cell lines. EPOR knockdown abrogated human tumor cell growth, induced apoptosis through Bim, reduced invasiveness, and caused downregulation of MYC expression. EPO-induced MYC expression is mediated through the PI3K/AKT and MAPK pathways, and overexpression of MYC partially rescued loss of cell proliferation caused by EPOR downregulation. In a xenotransplantation model, designed to simulate recombinant EPO therapy in breast cancer patients, knockdown of EPOR markedly reduced tumor growth. Thus, our experiments in vitro and in vivo demonstrate that functional EPOR signaling is essential for the tumor-promoting effects of EPO and underline the importance of the EPO-EPOR axis in breast tumor progression. / Cancer Research UK - C10141/A9977 (TRL, MET, KKC). Particle Manipulation Using Electric Field Gradients in Microdevices

Electrokinetics is a family of effects that induces motion of a liquid or a particle within a liquid in response to an external electric field. Using the intrinsic electrical properties of bacteria and of breast cancer cells, electrokinetics can be used to manipulate these particles for two different types of applications: tissue engineering and breast cancer detection. The first application studied the effects of electric fields on bacteria cells as well as calcium ions to potentially create a meniscus scaffold with hydroxyapatite ends for anchoring. In response to the electric field, calcium ions were able to deposit locally and simultaneously with cellulose growth. Bacteria cells were also studied to determine their response under an AC field. At low frequencies, bacteria demonstrated controlled movement caused by electroosmosis and dielectrophoresis with a net motion caused by a dielectrophoretic force. In the second application, the separation capabilities of different stages of breast cancer cells from the same cell line were tested using contactless dielectrophoretic (cDEP) devices. The electric field gradients in cDEP devices were altered to optimize selectivity and to determine an estimated membrane capacitance for each. From the results, the membrane capacitance of the early to intermediate stages proved to be very similar; however, late stage breast cancer cells have potential in being separated from early and intermediate stages. / Master of Science

Investigating The Role Of Chaperonin Containing TCP-1 In Extracellular Vesicles Isolated From Breast And Prostate Cancer Cell Lines

Extracellular vesicles have been shown to be an important factor in cancer progression through cell-to-cell communication of proteins, lipids, and nucleic acids. There are certain proteins that are seen commonly in these vesicles including the multi-subunit protein Chaperonin Containing TCP-1 (CCT) that is frequently upregulated in cancers as they progress in stage/severity. New literature suggests that the silencing of CCT leads to the dysregulation of EV secretion through alterations in the cellular metabolism of lipids. To date, no work has been done on cells that have an increased amount of CCT, which is what this work aims to explore. Exogenously expressing one of the CCT subunits, CCT2, caused an increase in CCT2 protein and RNA content in EVs. There was also a significant increase in the number of EVs smaller than 100nm in size. While the biological role of these EVs is yet to be determined, preliminary data suggests that EVs less than 100nm are able to increase total CCT2 protein levels in control cells when compared with conditioned media containing EVs of all sizes. This investigation aims to characterize these EVs in hopes to translate the findings clinically to detect EVs from cancer patients as a noninvasive diagnostic tool.

Cytotoxic Activity of Sphingosine-1-Phosphate against Human Triple-negative/ Basal-like Breast Cancer

Breast cancer is one of the most common malignancy diagnosed in women and is the primary cause of cancer-related deaths in women worldwide. It is a heterogeneous group of diseases that have a different response, prognosis, and clinical outcomes. Estrogen, progesterone and HER2 negative breast cancer, known as triple negative breast cancer (TNBC), does not respond to hormonal therapy. Basal-like breast cancer (BLBC) has shorter overall survival rate among other subtypes. Tumors sharing both TNBC and BLBC are considered less responsive to currently available treatment. Chemoresistance to treatment has been a challenge in cancer biology and force investigation toward developing new targeted therapies, which selectively target specific subtypes. Sphingolipid metabolites have an important physiological role in determining cell fate. Sphingolipid metabolites, ceramide, sphingosine, and sphingosine-1-phosphate (S1P), are implicated in cancer. S1P exerts its functions via extracellular and intracellular targets. S1P synthesized inside the cell is exported outside and binds to G-protein coupled receptors, the sphingosine-1-phosphate receptors 1-5 (S1PR1-5). Although the intracellular function is not well defined, its suggested intracellular S1P promotes cell apoptosis. The S1P pathway has received great attention recently due its function in cell survival and death. This effect was reported to be concentration dependent. In this research, I focused on S1P effect on nine TNBC/BLBC cell lines. I examined the in-vitro effects of S1P on apoptosis, proliferation, and cytotoxicity in triple negative/ basal-like breast cancer cell lines. Moreover, I studied the co-administration of S1P with currently used chemotherapeutic agents in these cell lines. Data show that S1P can selectively induce cell death in TNBC/BLBC cell lines at a specific concentration. In this research, I found that the mechanism of cell death following treatment with different S1P concentrations was mainly due to apoptosis. Results show that S1P leads to cell shrinkage, rounding and detachment in the nine TNBC/BLBC cell lines. S1P combination with doxorubicin and docetaxel at different concentrations shows no beneficial effect of the combination compared to the chemotherapeuitc agent alone. In some cell lines, the combination showed a protective effect. Further studies are required to determine the mechanism by which S1P induces cell apoptosis, inhibits cell growth, and demonstrates lack of responsiveness in combination studies.

Phytoestrogenic extracts of Cyclopia modulate molecular targets involved in the prevention and treatment of breast cancer

Thesis (PhD)--Stellenbosch University, 2014. / ENGLISH ABSTRACT: Phytoestrogen containing extracts of Cyclopia, an indigenous South African fynbos plant used to prepare honeybush tea, may serve as a source of new estrogen analogues. It would be of great benefit if these new analogues would not only prevent the development and progression of breast cancer which, globally, is responsible for the highest number of cancer associated deaths among females, but also have a reduced side-effect profile when compared to current treatments and, in addition, also alleviate menopause associated symptoms. In this study three extracts, P104, SM6Met, and cup-of-tea, from two species of Cyclopia, C. genistoides and C. subternata, were evaluated for their potential to modulate molecular targets involved in prevention and treatment of breast cancer. We show that the phytoestrogenic extracts of Cyclopia antagonise estrogen-induced cell proliferation both in vitro as well as in vivo. Furthermore, our study presents various molecular mechanisms whereby the Cyclopia extracts may be eliciting this effect. Importantly, we show, for the first time, that the Cyclopia extracts behave as ERα antagonists and ERβ agonists which, with respect to the known role of the ER subtypes in breast cancer, where the ERα subtype is associated with the stimulation of cell proliferation and the occurrence of breast cancer, while ERβ ameliorates the action of ERα in breast cancer and could act as an inhibitor of breast cancer development, may be beneficial for the prevention or treatment of breast cancer. In addition, we also show that the extracts of Cyclopia behave as selective estrogen receptor degraders by down-regulating ERα protein levels while stabilising ERβ protein levels, which not only provides a possible molecular explanation for the observed ERα antagonism and ERβ agonism, but, in addition, may be beneficial as higher ERα levels are associated with malignant breast cancer tumours, while higher ERβ levels are associated with benign tumours. Furthermore, we show that the Cyclopia extracts affect the nuclear localization and distribution of both ER subtypes in a manner that provides an additional molecular explanation for the observed ERα antagonism and ERβ agonism. Investigation of the molecular processes involved in the promotion and progression of breast cancer, such as the distribution of cells between the phases of the cell cycle, cancer cell invasion, and the regulation of genes governing these processes provides evidence that the Cyclopia extracts are not as proliferative as estrogen. In addition, Cyclopia extracts display anti-inflammatory properties, which may be beneficial as inflammation is an enabling characteristic in cancer development and progression. Furthermore, this study, for the first time, shows that the phytoestrogenic extracts of Cyclopia are absorbed, are not toxic, and display biological ERα antagonist activity in vivo by retarding uterine growth. Thus, we propose that the Cyclopia extracts act as selective estrogen receptor subtype modulators with potential to be developed as a nutraceutical for the treatment or prevention of breast cancer. / AFRIKAANSE OPSOMMING: Fitoëstrogeen-bevattende ekstrakte van Cyclopia, ‘n inheemse Suid Afrikaanse fynbosplant wat gebruik word vir die voorbereiding van heuningbostee, mag as ‘n bron van nuwe estrogeen-analoë dien. Verdere ondersoek van die molekulêre prosesse betrokke by die promosie en progressie van borskanker, soos die verspreiding van selle tussen die fases van die selsiklus, die beweging van kankerselle na omliggende weefsels, en die regulering van gene wat hierdie prosesse beheer, verskaf bewyse dat die Cyclopia-ekstrakte nie so proliferatief is soos estrogeen nie. Die ekstrakte van Cyclopia vertoon boonop ook anti-inflamatoriese eienskappe, wat voordelig mag wees aangesien inflammasie ‘n bydraende eienskap in kankerontwikkeling en -progressie is. Verder wys hierdie studie vir die eerste keer dat die fitoëstrogeniese ekstrakte van Cyclopia geabsorbeer word, nie toksies is nie, en dat hulle biologiese ERα-antagonis aktiwiteit vertoon deurdat hulle uterus-groei vertraag in vivo.
