Environmental and nutritional effects on beef tenderness in Texas

Effects of three environments and their native or rye pasture forage systems on subsequent carcass characteristics, composition, and meat palatability were examined following grain feeding. Calves (F2 Angus x Bos indicus) with similar pre-weaning management and genetics were randomly assigned to one of eight treatments (McGregor-Calf; McGregor-Low; Uvalde-Low; Uvalde-High; Overton-Low, Rotational; Overton-Low, Continuous; Overton-High, Rotational; and Overton-High, Continuous). After approximately five months of grazing, cattle in forage groups were transported to McGregor and finished on a corn-milo ration to visually assessed 10 mm fat thickness. Longissimus muscle sections were randomly assigned into aging periods of 1, 7, 14, 21, 28, or 35 days and two-2.54 cm steaks were removed per section for Warner-Bratzler shear force and trained sensory analysis. After grazing, steers fed diets designed to produce higher rates of gain weighed more than steers at the same location fed diets designed for lower rates of gain. Within the high nutrition treatments, Uvalde-High steers had a lower average daily gain than steers in the Overton-High treatments. Differences in environment and nutritional treatments induced differences in rate of gain during the stocker phase. All forage treatment groups increased average daily gain in the feedlot phase. After the feedlot phase, steers varied in live weight due to treatment. Nutritional/environmental treatments affected ribeye area; kidney, pelvic and heart fat; and hot carcass weight, but not final yield grade. Treatment had minimal affects on quality grade characteristics. There was no affect by treatment on Warner-Bratzler shear force or sensory panel perception of myofibrillar tenderness, connective tissue amount, overall tenderness, or overall flavor intensity. Storage day affected shear force; steaks were toughest at 1 and 7 days of storage with shear force values decreasing after 7 days of storage. There was no interaction between environmental/nutritional treatment and storage day. Juiciness and cook loss were affected by treatment (P=0.0001). Overall, environment and nutrition had little impact on eating quality. Despite differences in average daily gain, it appears that steers may be fed forage diets followed by grain feeding to a similar fat thickness endpoint and produce steaks that are similar in palatability to grain-fed steers.

O Brasil dedica-se em grande parte à criação de animais Bos indicus, principalmente da raça Nelore e seus cruzamentos, já que são animais resistentes a doenças e adaptados ao clima tropical. Entretanto, em relação à característica de maciez da carne, restrições têm sido atribuídas a este tipo de animal. Particularmente neste aspecto, existe grande interesse pela seleção de animais cuja genética seja favorável à carne mais macia e com redução na variação da maciez da carne. Durante o período de maturação da carne, a proteólise é o fator que mais contribui com o aumento da maciez. As proteases neutras ativadas por íons de cálcio, denominadas calpaínas, são parcialmente responsáveis pela proteólise post mortem, conduzindo ao aumento progressivo da maciez da carne. Entretanto, existe dificuldade em obter dados fenotípicos relacionados à maciez. Assim, a Seleção Assistida por Marcadores (MAS) pode ter grande impacto para melhorar estas características de difíc il mensuração. Marcadores do gene da calpaína (CAPN) e da calpastatina (CAST), denominados SNPs (Single Nucleotide Polymorphisms), já foram analisados e polimorfismos foram associados favoravelmente com maciez da carne bovina. Entretanto, pouco se conhece sobre a expressão gênica da calpaína e da calpastatina, particularmente em bovinos castrados da raça Nelore com diferentes combinações genotípicas para os marcadores moleculares SNP associados a essas proteases. Assim, este estudo teve como objetivo avaliar a expressão gênica da calpaína e da calpastatina em animais castrados da raça Nelore, com genótipos contrastantes identificados previamente por marcadores SNPs para um, sete e 14 dias de maturação. Foram genotipados 16 animais castrados da raça Nelore e dois marcadores foram utilizados, sendo um associado ao gene da µ-calpaína e outro associado à calpastatina (CAPN4751 e UOGCAST). Através da eletroforese bidimensional (2DE) as intensidades de expressão dos spots foram determinadas. As análises estatística s foram realizadas com auxílio do programa Statistical Analysis System (SAS), utilizando o procedimento PROC MIXED. A proteólise post mortem entre os tempos determinados de maturação foi avaliada e foram observados que 41 spots apresentaram efeitos significativos entre os tempos de maturação. Seis spots explicaram 57% da variabilidade da maciez aos 14 dias de maturação. Com o objetivo de verificar a existência de possíveis diferenças de expressão gênica entre amostras com genótipos distintos, avaliações proteômicas foram realizadas visando avaliar as diferenças no perfil proteico e foram baseadas no estudo comparativo (match) dos géis inter e intra combinações genotípicas. Dois spots explicaram 69% da variabilidade da maciez aos sete dias de maturação. Para o período de 14 dias de maturação 19 spots apresentaram efeito principal significativo para a fonte de variação de interação CAPN4751(UOGCAST) e, neste caso, a análise de regressão apresentou um spot que explica 70% da variabilidade da maciez aos 14 dias de maturação. / Brazil is largely devoted to the breeding of Bos indicus, especially Nellore and their intersections, because these animals are resist ant to diseases and adapted to tropical climate. However, in relation to beef tenderness, restrictions have been assigned to this breed. Particularly in this respect, there is great interest in the genetic selection of animals with more tender beef and reduced variation in meat tenderness. During the aging of the meat, proteolysis is the major factor that contributes to the increased tenderness. The neutral proteases activated by calcium ions, called calpains, are partially responsible for postmortem proteolysis, leading to a progressive increase in meat tenderness. However, there is difficulty in obtaining phenotypic data related to tenderness, in this way, Marker Assisted Selection (MAS) may have great impact to improve these characteristics. Gene markers of calpain (Capn) and calpastatin (CAST), called SNPs (single nucleotide polymorphisms) have been studied and polymorphisms were associated positively with beef tenderness. However, not much is known about the gene expression of calpain and calpastatin, particularly in Nellore steers with different genotype combinations for the SNP markers associated with these proteases. For the period of 14 days of aging 19 spots showed significant main effect for interaction CAPN4751(UOGCAST), in this case regression analysis showed a spot that explains 70% of the variability of tenderness to 14 days of aging.

Collagen Solubility and Calcium Concentration and Their Effects on Tenderness in the M. longissimus lumborum

Strip steaks from the McGregor genome project were used to evaluate the effects of sarcomere length, myofibrillar fragmentation index, 3 h postmortem pH, 24 h postmortem pH, marbling, electrical stimulation (ES), sarcoplasmic free calcium concentration, and collagen characteristics on tenderness as measured by Warner-Bratzler shear force (WBS). The WBS values were measured prior to this project so the animals were able to be separated into “tender” and “tough” groups using a WBS value of 30 N as the separating point, steaks with a WBS value less than 30 N being “tender” and the others being “tough”. It was found that ES sides had lower WBS values, however, “tough” steaks showed a greater response to ES than “tender” steaks. ES sides also had higher sarcoplasmic free calcium concentration and lower 3 h postmortem pH. Tenderness is best predicted by treatment (ES versus NON-ES), however, there is some efficacy in using total collagen and collagen solubility in conjunction with treatment.

Genetický polymorfismus vybraných kódujících lokusů ve vztahu k technologickým vlastnostem masa / Genetic polymorphism of selected loci in relationship to technological traits of meat

Within the framework of this diploma, a genotypization of 162 beef cattle of the ČESTR commercial breed from was performed in order to analyze the potential influence of gene coding calpain I (CAPNI) on the resulting beef tenderness. CAPN1 present on the BTA29 chromosome, was selected in previous studies as a candidate gen for QTL. Polymorphism of CAPNI gene was studied using PCR/RFLP method and CAPN530 marker. Genotype identification resulted from this procedure. A 341 bp long fragment was present in homozygotes AA, two fragments of 195 and 146 bp were present in homozygotes GG, and three fragments of 341, 195 and 146 bp were detected in heterozygotes AG. The genotypization output was subsequently statistically evaluated. 11 homozygotes AA, 62 homozygotes GG and 89 heterozygotes AG were detected in the analyzed beef cattle population. More frequent G allele occurred in the set with the frequency of 0,657 and A allele with the frequency of 0,343. Warner-Bratzler shear test was employed to determine beef tenderness based on the shear force attribute. Samples of raw and grilled beef aging 1, 14 and 28 days after the slaughter were analyzed. Statistical methods were used to evaluate relationship between genotype and detected amount of shear force. In case of raw beef, only the samples aging 1 day after the slaughter showed significant difference in the shear force. The most positive results for the given parameter were achieved in case of AG genotype. Samples aging 14 and 28 days after the slaughter showed no difference in shear force and related beef tenderness, indicating no genotype influence. In case of grilled beef, no significant difference in sheer force indicating possible genotype influence was detected at any day of analysis. Genotype influence on grilled beef tenderness within the test animal population is minimal and statistically inconclusive.

The influence of growth-promoting technologies on the biological structures responsible for cooked meat tenderness

Master of Science / Department of Animal Sciences and Industry / John Michael Gonzalez / The objective of this body of work was to examine effects of growth-promoting technologies (GP) on Longissimus lumborum meat tenderness, focusing on alterations of muscle fiber cross-sectional area (CSA) and collagen solubility. Two studies were conducted and analyzed as randomized complete block designs with repeated measures with GP and day of postmortem aging (DOA) as main effects. Treatments consisted of: a control (CON), implant only (IMP), and implant and [beta]-adrenergic agonist (COMBO). The [beta]-adrenergic agonist utilized for the first was zilpaterol hydrochloride, while the second study examined ractopamine hydrochloride. Objective tenderness of strip loin steaks was measured through Warner-Bratzler shear force (WBSF) after 2 (study 2) or 3 (study 1), 7, 14, 21, or 35 d of postmortem aging. Muscle fiber CSA and collagen solubility were analyzed via immunohistochemistry and hydroxyproline content, respectively. For the first study there was a treatment × DOA interaction (P < 0.01) for WBSF. Compared to CON steaks, IMP steaks had greater (P = 0.01) WBSF on d 3, but were similar (P = 0.21) by d 14. The COMBO steaks remained less tender at all-time points (P < 0.04) except d 21 (P = 0.13) when compared to the CON. Growth-promoting treatment increased the CSA of all three muscle fiber types (P < 0.01), but had no effect on collagen solubility measures (P > 0.21). The second study observed no treatment × DOA interaction (P = 0.54) for WBSF, but GP increased (P < 0.01) WBSF across all DOA. Growth-promoting treatment tended to increase the CSA of type I and IIX fibers (P < 0.10), and increased (P < 0.01) type IIA fiber CSA. In agreement with the first study, there was no treatment × DOA interaction or treatment effect on collagen solubility (P > 0.75). The addition of GP to feedlot heifer production increased WBSF of strip loin steaks and fiber CSA, but did not impact collagen characteristics.

Expressão gênica dos receptores de cortisol no músculo de bovinos Nelore e associação com características endócrinas, metabólicas e qualidade da carne / Gene expression of cortisol receptors in muscle of Nellore cattle and association with endocrine and metabolic characteristics and meat quality

O estresse provoca alterações significativas no metabolismo dos animais, provocando a liberação de hormônios glicocorticoides. Estas alterações do metabolismo têm efeito anabólico sobre o metabolismo proteico muscular, podendo afetar os processos bioquímicos de transformação do músculo em carne. O presente trabalho teve como objetivo geral (i) verificar as relações entre variáveis endócrinas e metabólicas associadas ao estresse e características indicadoras de qualidade da carne, em animais castrados e não-castrados; (ii) avaliar a expressão gênica dos receptores mineralocorticoide (MR) e glicocorticoide (GR) em variáveis endócrinas, metabólicas e relacionadas à qualidade da carne de bovinos Nelore castrados e não-castrados. Para tal, 130 animais foram abatidos entre os anos de 2009 e 2011. Amostras de sangue foram coletadas antes e depois do abate para mensuração das concentrações de ACTH e cortisol. Amostras do músculo Longissimus dorsi foram coletadas durante os abates para mensuração do glicogênio e lactato, bem como, para análises de expressão gênica (RT-qPCR). Para as análises de maciez, foram coletadas amostras maturadas por um, sete e 14 dias. Para expressão gênica foram determinados os genótipos dos animais para três marcadores relacionados ao MR (MR1_1, MR1_2 e MR1_3) e dois ao GR (GR2_1 e GR2_2), por meio de PCR em tempo real. Foi verificado que animais castrados apresentam pH 24 horas menores e carnes mais macias ao sétimo e 14º dias de maturação, bem como, concentrações de cortisol (in vivo e post mortem) e lactato significativamente superiores aos animais não-castrados. O marcador MR1_3 apresenta expressão gênica significativamente diferenciada. Os animais com genótipo GA apresentaram 57,27% mais transcritos quando comparados aos animais GG. A expressão gênica do MR e GR foi significativamente relacionada às concentrações de cortisol in vivo e post mortem, porém não influenciou as concentrações de ACTH (in vivo e post mortem), glicogênio e lactato. A expressão gênica do MR e GR não foi relacionada às características indicadoras da qualidade da carne. / The stress causes significant changes in the metabolism of the animals causing the release of glucocorticoid hormones. These metabolic changes have anabolic effect on muscle protein metabolism, affecting the biochemical processes of transformation of muscle on meat. This study aimed to (i) examine relationships between endocrine and metabolic variables associated with stress and meat quality characteristics in castrated and non-castrated animals, (ii) evaluate mineralocorticoid receptor (MR) and glucocorticoid receptor (GR) gene expression in endocrine and metabolic characteristics and related this to meat quality of Nellore castrated and non-castrated animals. To this end, 130 animals were slaughtered between the years 2009 and 2011. Blood samples were collected before and after slaughter to measure concentrations of ACTH and cortisol. Longissimus dorsi muscle samples were collected during slaughter for measurement of glycogen and lactate, as well for gene expression analyzes (RT-qPCR). For the shear force analyzes, samples were aged for one, seven and 14 days. For gene expression analysis, genotypes of three markers related to MR (MR1_1, MR1_2 and MR1_3), and the two related to GR (GR2_1 and GR2_2) were determined via real-time PCR. It was observed that castrated have lower pH value at 24 hours than non-castrated animals, and tender meat on the seventh and 14th day of aging, such as cortisol (in vivo and post mortem) and lactate concentrations significantly superior to non-castrated animals. Gene expression of MR1_3 was significantly different. Animals with GA genotype had 57.27% more transcripts than GG genotype. The gene expression of MR and GR was significantly related to cortisol concentrations in vivo and post mortem, but did not influence the concentrations of ACTH (in vivo and post mortem), glycogen and lactate. The MR and GR gene expression was not related to the meat quality characteristics.

To this end, 130 animals were slaughtered between the years 2009 and 2011. Blood samples were collected before and after slaughter to measure concentrations of ACTH and cortisol. Longissimus dorsi muscle samples were collected during slaughter for measurement of glycogen and lactate, as well for gene expression analyzes (RT-qPCR). For the shear force analyzes, samples were aged for one, seven and 14 days. For gene expression analysis, genotypes of three markers related to MR (MR1_1, MR1_2 and MR1_3), and the two related to GR (GR2_1 and GR2_2) were determined via real-time PCR. It was observed that castrated have lower pH value at 24 hours than non-castrated animals, and tender meat on the seventh and 14th day of aging, such as cortisol (in vivo and post mortem) and lactate concentrations significantly superior to non-castrated animals. Gene expression of MR1_3 was significantly different. Animals with GA genotype had 57.27% more transcripts than GG genotype. The gene expression of MR and GR was significantly related to cortisol concentrations in vivo and post mortem, but did not influence the concentrations of ACTH (in vivo and post mortem), glycogen and lactate. The MR and GR gene expression was not related to the meat quality characteristics.
