This thesis reports the effect of the non-toxic cyanobacterium Oscillatoria agardhii CYA29 and its spent medium on the cell-bound toxin levels in Microcystis aeruginosa PCC7820 and Nodularia sp. PCC7804. Microcystin levels were elevated when O. agardhii or its spent medium were added to cultures of M. aeruginosa PCC7820. This effect was also observed for two nodularin variants in Nodularia PCC7804. However, growth of M. aeruginosa in its own spent medium did not lead to elevated microcystin levels. Some Gram-negative bacteria use quorum sensing, the determination of population density by pheromones, to regulate the expression of traits in a density dependent manner. The presence of the bacterial pheromones, acyl-homoserine lactones (AHLs), was investigated in cyanobacteria and a simple and fast synthesis for AHLs employed. No AHLs have been found in the cyanobacterial species tested, neither by Chromobacterium violaceum CV026 nor by Escherichia coli HB101 pUCD607 bioassay. No changes in dry weight or microcystin concentration were observed in M. aeruginosa PCC7820 grown in the presence of the AHL N-hexanoyl-DL-homoserine lactone. The activity of the spent medium of O. agardhii CYA29 was retained when it was heated up to 100°C for 20 min, frozen or freeze-dried. Molecular weight cut-off filtration showed the active compound to be less than 1 kD in size. The active component could not be extracted by dichloromethane or methanol, and activity was lost upon acidification. A small peptide of five amino acid moieties was isolated from an active fraction of the spent medium, four of which have been identified by 1H NMR to be serine, glycine, alanine, and the modified serine(thiazole). To my knowledge this is the first report of serine(thiazole) in natural products and the first thiazole containing peptide from O. agardhii.

MHC, parasite burden and heterozygosity in the blue shark (Prionace glauca, L.1758)McMillan, Heather Anne January 2013 (has links)
The blue shark (Prionace glauca) is a highly migratory pelagic elasmobranch that inhabits ocean basins globally. As a result, this shark is exposed to intensive ocean exploitation by commercial target fisheries, by-catch and for recreational pursuits globally. This top predator is therefore at high risk of becoming overfished. Advances to current knowledge of genetic population structure and diversity of this species would provide vital information required to initiate co-operative management approaches. In this study, the major histocompatibility complex (MHC) class IIa and IIβ genes were successfully isolated and characterised from blue sharks. Phylogenetic trees of the class II genes showed three major clades; one of teleost fish, one of tetrapods and one of sharks. The MHC class IIβ gene exon 2 primers successfully amplified partial sequences in blue sharks from several global locations. Analysis of sequences using denaturing gradient gel electrophoresis (DGGE) suggested the assay resolved different sequences up to one basepair, making the assay potentially very useful with further development. The class II genes presented in this study show conflicting evidence for the presence of more than one class II locus. To explore inheritance patterns of MHC exon 2 diversity, a single blue shark litter (mother + 19 pups) was cloned and sequenced, revealing evidence to suggest the possibility of more than one locus for class IIβ. Statistical analysis of parasite loads and diversities from blue shark spiral valves revealed no definitive population structure, supporting global and North Atlantic mtDNA and microsatellites genetic analyses presented here. The size (fork length) of sharks was found to be potentially influential when modelled with individual microsatellite heterozygosity and fork length. International co-operation will be required to prevent this species becoming extinct from global marine ecosystems. Reductions in numbers could lead to reduced genetic diversity, decreased immunity and ultimately an 'unhealthy' population.

Best Linear Unbiased Estimation Fusion with ConstraintsZhang, Keshu 19 December 2003 (has links)
Estimation fusion, or data fusion for estimation, is the problem of how to best utilize useful information contained in multiple data sets for the purpose of estimating an unknown quantity — a parameter or a process. Estimation fusion with constraints gives rise to challenging theoretical problems given the observations from multiple geometrically dispersed sensors: Under dimensionality constraints, how to preprocess data at each local sensor to achieve the best estimation accuracy at the fusion center? Under communication bandwidth constraints, how to quantize local sensor data to minimize the estimation error at the fusion center? Under constraints on storage, how to optimally update state estimates at the fusion center with out-of-sequence measurements? Under constraints on storage, how to apply the out-of-sequence measurements (OOSM) update algorithm to multi-sensor multi-target tracking in clutter? The present work is devoted to the above topics by applying the best linear unbiased estimation (BLUE) fusion. We propose optimal data compression by reducing sensor data from a higher dimension to a lower dimension with minimal or no performance loss at the fusion center. For single-sensor and some particular multiple-sensor systems, we obtain the explicit optimal compression rule. For a multisensor system with a general dimensionality requirement, we propose the Gauss-Seidel iterative algorithm to search for the optimal compression rule. Another way to accomplish sensor data compression is to find an optimal sensor quantizer. Using BLUE fusion rules, we develop optimal sensor data quantization schemes according to the bit rate constraints in communication between each sensor and the fusion center. For a dynamic system, how to perform the state estimation and sensor quantization update simultaneously is also established, along with a closed form of a recursion for a linear system with additive white Gaussian noise. A globally optimal OOSM update algorithm and a constrained optimal update algorithm are derived to solve one-lag as well as multi-lag OOSM update problems. In order to extend the OOSM update algorithms to multisensor multitarget tracking in clutter, we also study the performance of OOSM update associated with the Probabilistic Data Association (PDA) algorithm.

Frog abundance and diversity in urban and non-urban habitats in the upper Blue Mountains (New South Wales)Lane, Alan Gordon, University of Western Sydney, College of Health and Science, School of Natural Sciences January 2005 (has links)
This study was undertaken between July 24, 2003 and January 16, 2004 to investigate the influence of urban development upon the abundance and species diversity of frogs in the upper Blue Mountains of New South Wales, Australia. Five urban sites were paired with matched non-urban sites. Urban sites were located within or on the fringes of the towns of Katoomba and Blackheath and were subject to varying degrees of physical disturbance, as well as degradation and pollution by urban runoff and sewage. The non-urban sites were located within the Blue Mountains National Park and were effectively un-impacted by human activity. No adequate explanation emerged for the marked difference between the frog assemblages at the two types of habitat. It is speculated that the salts, detergents and other chemicals in urban wastewaters (roadway runoff, yard runoff and sewage) may provide the frogs at urban sites with some level of protection against disease, particularly chytridiomycosis. All indications from this and previous work are that the frog abundance and diversity in non-urban habitats in the upper Blue Mountains of New South Wales are showing the same trends in decline as observed in other montane regions of Australia. Urban habitats are important population reservoirs for the diversity of frog species absent from the non-urban habitats, but are vulnerable to progressive destruction from a variety of human impacts. Studies in the biology and reproductive characteristics of Pseudomugil signifer.Howe, Effie Helena Irene January 1995 (has links)
The aims of this study were firstly to observe the breeding behaviour and embryo ology. and then to identify factors affecting the reproductive biology of the Australian native pseudomtlgilid Pseudomugil signifer (pacific blue-eye) and the impact upon it of the presence of the exotic species Gambusia holbrooki (eastern gambusia). Six species of the genus Pseudomugil, and the related Scaturiginichthys venneilipinnis, are found on the Australian continent. The normal breeding behaviour, egg surface morphology and embryology of four species of Pseudomugil (P. signijer, P. genrudae, P. tenellus and P. mellis) were first investigated, using aquarium and microscopic (light and S.E.M.) studies. The four species were divided into two groups: P. signifer and P. mellis; and P. tenellus and P. gertrudae. The study provided further evidence for the view that the embryology of the genus Pseudomugil differs markedly from that of members of the family Melanotaeniidae, with which the pseudomugilids have previously been grouped. The seasonal pattern of gonadal function in P. signifer, both in the field and in aquariums, was then investigated for populations of P. signifer from the Sydney region. It was found that P. signifer bred over the spring and summer months, commencing breeding as the temperature and daylength increased, and declining in breeding activity as daylength and temperature declined. There was no substantial difference in the pattern of reproductive activity between wild and captive stocks of P. signifer in the populations used. The impact of the presence of the introduced G. holbrooki on P. signifer was then examined, first in open-air tank experiments , and then in the field. In the tank experiments the exotic species profoundly affected the breeding of the native species. When G. holbrooki were in the tanks P. signifer did not gain weight or grow in total length (except for females given supplementary feed); ovarian weight and fecundity was greatly reduced and the ovaries were morphologically undeveloped. No eggs from P. signifer were observed in tanks which also housed G. holbrooki. G. holbrooki were observed to actively hunt and eat young P. signifer and to nip the caudal fins of adult P. signifer. The results indicate clearly, that at least in a captive situation, the presence of the exotic species has a very deleterious effect on breeding and hence possible survival, of a native population. A pilot study conducted at the same time as the harvest of the second tank study did not reveal such drastic consequences. However, even in the less confined field situation, some evidence of an interrelation between water quality, numbers of P. signifer and numbers of G. holbrooki were seen in one disturbed site (Home bush Bay). These findings suggest that a newly designed field experiment based on data collected from the power analysis of the pilot study could clarify whether G. holbrooki adversely affects P. signifer in the wild. The information gained from these studies can be used in the management of P. signifer in the wild, and serve as a model of the possible effects upon other native species.

Incorporation of Organic Molecules in the Tunnels of the Sepiolite Clay MineralBlank, Katrin 13 September 2011 (has links)
Sepiolite is a clay mineral, a complex magnesium silicate, a typical formula for which is (OH2)4(OH)4Mg8Si12O30•8H2O. It is formed by blocks and cavities (tunnels) growing in the direction of the fibres. The tunnels, 3.7 x 10.6 Å in cross-section, are responsible for the high specific surface area and sorptive properties of sepiolite. The co-intercalation of 3-methyl cyclohex-2-en-1-one (MCH), the Douglas-Fir beetle anti-aggregation pheromone, with methanol, ethanol, acetone, or benzene into sepiolite tunnels was studied. The resulting nanohybrid materials were characterized by means of various techniques, such as multinuclear solid-state NMR spectroscopy, porosity studies and Thermal Gravimetric Analysis (TGA). This was done in the hope of obtaining slow and controlled release of MCH from the sepiolite tunnels. It was demonstrated by 13C MAS NMR (carbon-13 magic angle spinning nuclear magnetic resonance) that at room temperature there are two different MCH molecules: one MCH inside the tunnels and the other one outside the tunnels of the sepiolite. Heating nanohybrid materials at 60˚C for 20 hours removes the external MCH molecules from the sepiolite. 13C MAS NMR showed that by further heating nanohybrid materials at 120˚C for 20 hours, methanol, ethanol, or acetone peaks were greatly reduced; however, the benzene peak was not reduced. To better understand how benzene acts inside sepiolite, intercalation of d6-benzene, and co-intercalations of d6-benzene with MCH and d6-benzene with pyridine into sepiolite tunnels were carried out, and these samples were studied by the same techniques. Another technique was used in order to see whether the slow and controlled release of MCH from the sepiolite tunnels could be obtained: sepiolite-MCH nanohybrids were treated with 20 ml of 0.5 M HCl solution. It was found that when 1 gram of MCH-sepiolite sample was acid treated at room temperature, about 35% of intercalated MCH was removed from the sepiolite. The role of sepiolite clay was also studied in Maya-Blue representative structure sepiolite-indigo adduct. It is known that upon heating the sepiolite and indigo mixture, the stability that is present in Maya-Blue is achieved. It is still a mystery, however, how exactly indigo and sepiolite interact with each other.
