Characterization of Effects of Muc1 Expression on Epidermal Growth Factor Receptor Signaling in Breast Cancer

EGF receptors are key regulators of cell survival and growth in normal and transformed tissues. Ligand binding results in formation of homo/hetero dimers of these receptors, followed by activation of the kinase activity and subsequent tyrosine phosphorylation of many downstream molecules. The activation of these receptors is not only mediated by the binding of their cognate ligands, but by transactivaton by other molecules as well. Recent studies have identified an oncogenic glycoprotein MUC1 as a binding partner for EGFR and that MUC1 expression can potentiate EGFR-dependent signal transduction. After receptor activation, EGFR is typically downregulated via an endocytic pathway that results in receptor degradation or recycling. We report here that MUC1 expression inhibits the degradation of ligand-activated erbB1. In addition, MUC1 expression results in prolonged activation of Akt, but not ERK1,2 MAPKinase. The MUC1-mediated protection against degradation occurs with a decrease in EGF-stimulated ubiquitination of erbB1, and an increase in erbB1 recycling. We then utilized the WAP-TGFα transgenic mouse model of breast cancer and determined that a loss of Muc1 expression dramatically alters mammary tumor progression. While 100% of WAP-TGFα/Muc1^(+/+) mice form mammary gland tumors, only 37% of WAP-TGFα/Muc1^(-/-) form tumors. Furthermore, expression of cyclin D1 expression is significantly suppressed in tumors derived from WAPTGFα/Muc1^(-/-) animals, and loss of Muc1 expression resulted in a significant inhibition in the formation of hyperplastic lesions in the mammary gland. We also observed metastatic pulmonary adenocarcinoma (1/29) and perivascular lymphoma of unknown origin (28/29) in the WAP-TGFα transgenic mice but not in the WAP TGFα/Muc1^(-/-) animals. To determine the effects of Muc1 expression on metastasis in a model lacking perivascular lymphoma, we crossed MMTV-Wnt-1 and MMTV-MUC1 transgenic mice and evaluated interactions between Muc1 and EGFR. Although the MMTV-Wnt-1 mice are non-metastatic, a majority (6/10) of the bitransgenic MMTVWnt- 1/MMTV-MUC1 formed pulmonary metastases. Furthermore, overexpression of MUC1 increases the breast cancer cell invasion in vitro. The MUC1 induced increase in invasion is found to be EGF and EGFR-kinase dependent. Collectively, these data indicate that MUC1 expression contributes to many of the hallmarks of cancer and in addition, is an important modulator of EGFR-associated mammary tumor progression.

Cancer systems biology : is the devil in the glycolytic detail?

An approach to investigating cancer that has recently seen resurgence of interest is the “Warburg effect”. Otto Warburg originally described the altered metabolism of cancer cells and identified that they exhibit an increase in glucose uptake and lactate production. This up-regulation of glycolytic flux and glucose transport is now associated with 90% of cancers. In order to improve the overall understanding of the “Warburg effect” two forms of systems biology have been implemented - comparative in vitro analysis of kinetic activities and dynamic modelling. In this analysis, human breast cancer cell lines MCF-7, MDA-MB-231 and T47D and a non transformed breast cell line MCF-10A were used to identify key similarities and differences in kinetic activities across the glycolytic pathway. Additionally, activities of key glycolytic enzymes hexokinase, pyruvate kinase and lactate dehydrogenase were compared under hypoxic conditions to further understand regulation of cancer cells. The most prominent feature that arose from comparing the kinetic activities of the three malignant and one non-malignant cell line is that each cell line has its own specific set of activities for glycolysis. This indicates that there are differences in regulation across the glycolytic pathway for each of these cell lines. This is of specific interest in the search for therapeutic targets. Further, we determined that despite the prominence of oncogenic HIF signalling activities of hexokinase, pyruvate kinase and lactate dehydrogenase were further modulated by growth under hypoxic conditions. Despite the lack of obvious distinct kinetic differences between the non-cancerous and cancerous cells lines some discernible differences are apparent when modelled in silico.

Development Of Bio-Photonic Sensor Based On Laser-Induced Fluorescence

Laser-induced fluorescence (LIF) has been shown to be potentially useful for identifying microorganisms in real time. It is a selective and sensitive technique because the excitation is performed at one wavelength while the emission is monitored at longer wavelengths so that background from the excitation source can be eliminated. This specialized optical property of LIF can be applied to development of an optical sensor capable of quickly, non-invasively, and quantitatively probing complex biochemical transformations in microorganisms. Various bio-photonic optical fiber sensors based on laser-induced fluorescence (LIF) spectroscopy were developed as diagnostic tools for microorganisms. In the first phase, the enhancement of the sensitivity and selectivity of the optical sensor system focused on diagnosis of human breast cancer cell lines and Azotobacter vinelandii (an aerobic soil-dwelling organism). Autoluorescence spectra from human breast cancer cell lines and Azotobacter vinelandii corresponding to different growth environments were investigated. Then, the study has expanded to include the use of gold nanoparticles for specific DNA detection. The use of gold nanoparticales opens a door into construction of a compact, highly specific, inexpensive and userriendly optical fiber senor for specific DNA detection. An optical fiber laser-induced fluorescence (LIF) sensor based has been developed to detect single-strand (ss) DNA hybridization at the femtomolar level. Effects of various experimental parameters and configuration were investigated in order to optimize sensor performance and miniaturize sensor size.

Cytochrome P450 mRNA profile in human breast cancer cell lines

Cytochrome P450 enzymes (P450s) are involved in cancer development and treatment due to their roles in the oxidative metabolism of various endogenous (e.g. oestrogen) and exogenous (e.g. tamoxifen) compounds. It is well-known that intermediate P450 metabolites derived from oestrogen metabolism are associated with breast carcinogenesis. The main aim of this project was to profile the cytochrome P450 and P450-regulatory nuclear receptor mRNAs in a series of breast cancer cell lines (BCCs) and compare this profile with normal breast cells. This study used the qualitative reverse transcriptasepolymerase chain reaction (RT-PCR) to detect mRNA expression of target genes. Results showed CYP1B1, CYP2D6, CYP2J2, CYP2R1, CYP2U1 and CYP4X1 mRNA to be present in all cell lines. CYP2A6, CYP2C8, CYP2C18, CYP2F1 and CYP4Z1 mRNA were expressed in oestrogen receptor (ER)-positiveCaucasian and ER-negative Afro- Caribbean BCCs. Although no differences in P450 mRNA were observed between the different ethnic groups, these preliminary findings suggest potential similarities in the ERpositive Caucasian and ER-negative Afro-Caribbean BCCs which warrant further investigation The CYP4Z1 PCR product was identified as two distinct bands. Specific primer sets were used to demonstrate potential intron retention in CYP4Z1. Using established in vitro models for the study of regulatory mechanisms of CYP4Z1, T47D and ZR-75-1 breast cancer cell lines were used to determine the appropriate nuclear receptors (i.e. progesterone receptor, glucocorticoid receptor or peroxisome proliferator-activated receptor alpha ). These findings suggest that there may be an alternative receptor mechanism involved in CYP4Z1 mRNA induction in these cells. In conjunction, pre-treatment of these two cell lines with the RNA synthesis inhibitor actinomycin D followed by the agonists showed a significant reduction (p < 0.05) of CYP4Z1 mRNA levels and inhibited CYP4Z1 induction by either progesterone, dexamethasone or pirinixic acid, indicating that these agonists have effects on CYP4Z1 mRNA transcription or stability. In contrast, cycloheximide differentially affected the level of CYP4Z1 mRNA induction by these agonists. Taken together, these results suggest that CYP4Z1 mRNA induction in T47D and ZR-75-1 is mediated through differential cell type specific regulatory mechanisms and there is evidence for differential regulation of the splice variants.

L'objectif de cette thèse est d'étudier la sensibilité des lignes cellulaires du cancer du sein BTCL aux agents stabilisants des microtubules (taxanes et ixabépilone) afin de: 1 - identifier la pharmaco-génomique prédictif de la réponse (résistance / sensibilité) comme une signature moleculaire, et de valider cette signature sur d'autres études dont les données génomiques sont disponibles en ligne, donc mis l'expression des gènes prédictifs de GES pour Tax- sensibilité (333 gènes ) et Ixa-sensibilité (79 gènes) ont été définis, et les Taxanes prédicateurs GES a considérablement prédit Pac-sensibilité dans BTCL, et pathologiques réponse complète à base de Pac-chimiothérapie néoadjuvante chez les patients du cancer du sein. 2 - étudier le rôle des cellules souches du cancer (ALDH +) sur la réponse thérapeutique aux Taxanes et donc, Nous identifions quatre lignes BTCL qui présentent un enrichissement significative dans le pourcentage et le nombre absolu de ALDELFUOR cellules positives dans chacun de ces quatre BTCLs après 5 jours de traitement par le paclitaxel, en contraste avec les résultats précédents, nous avons constaté que dans ces autres 3 BTCLs le phénomène est inversé avec la diminution significative du pourcentage et le nombre absolu de cellules positives ALDELFUOR trouve dans chacun de ces trois BTCLs après 5 jours du traitement par le paclitaxel. Une signature moléculaire de SCC résistant / sensible de 243 pb avec 179 gènes dont 152 gènes sont régulés à la hausse et 27 gènes régulés à la baisse au CSC résistantes au paclitaxel, une sorte prédicteurs génomiques pour Tax - sensibilité au CSC résistantes au paclitaxel peut être dérivée à partir BTCL et peut être utile pour mieux comprendre les mécanismes de résistance aux taxanes et de l'implication de la CSC dans cette résistance, afin de mieux sélectionner des traitements cytotoxiques chez les patients du cancer du sein et l'identification des d'autres marqueurs potentiels de thérapies ciblées dans l'avenir. 3 - Nous avons testé l'impact de l'altération des paramètres génomiques et protéiques ou les mutations de certains gènes comme tau (MAPT), K-alpha tubuline (TUB A1B) tubuline alpha-6 (A1C TUB) tubuline beta 3 (TUBB3) et stathmine (STMN1), malheureusement nous n'avions jamais identifier une mutation pour être corrélée à la réponse des BTCL aux Taxanes. 4 - Nous essayons d'étudier au niveau de protéines par immunohistochimie sur le tissu de micro-array et cyto-micro-array pour certains paramètres qui ont été déjà prouvé (in vitro) pour être corrélée à la réponse aux Taxanes, (cette partie est en fait en cours). / The aim of this thesis is to study the sensitivity of breast cancer cell lines BTCL to microtubule-stabilizing agents (Taxanes and ixabepilone) in order to:1- identify pharmaco-genomic predictor of response (resistance /sensitivity) as a molecular signature, and to validate this signature on others studies of which the genomic data are available on line, so gene expression set GES predictors for Tax-sensitivity (333 genes) and Ixa-sensitivity (79 genes) were defined, and the Taxanes GES predictors has significantly predicted Pac-sensitivity in BTCL, and pathological complete response to Pac-based neo-adjuvant chemotherapy in BC patients.2- study the role of cancer stem cell (ALDH+) on the therapeutic response to Taxanes and their we identify 4 BTCLs which present a significant enrichment in the percentage and the absolute numbers of ALDELFUOR-positive cells found in each of these 4 BTCLs after 5 days of treatment by Paclitaxel , In contrast to the previous results we found that in others 3 BTCLs these phenomenon is inversed with the significant decrease of the percentage, and the absolute numbers of ALDELFUOR-positive cells found in each of these 3 BTCLs after 5 days of treatment by Paclitaxel.A molecular signature of CSC resistant /sensitive of 243 pb with 179 genes of which 152 genes are up- regulated and 27 genes down-regulated in CSC resistant to Paclitaxel, so a genomic predictors for Tax-sensitivity in CSC resistant to Paclitaxel can be derived from BTCL and may be helpful for better understanding the mechanisms of resistance to Taxanes and the implication of CSC in this resistance in order to better select of cytotoxic treatment in breast cancer patients and identification of others potential markers for targeted therapies in the future .3- we tested the impact of the alteration of genomic and proteic parameters or the mutations of some genes like tau (MAPT),Tubulin K- ALPHA (TUB A1B) Tubulin alpha-6 (TUB A1C) Tubulin beta 3 (TUBB3) and Stathmin (STMN1), unfortunately we did'nt identify a mutations to be correlated to BTCL response to Taxanes .4- we try to study at the level of proteins by immunohistochemistry on the tissue micro- array and cyto-micro-array for some parameters which have been already proved (in vitro) to be correlated with response to Taxanes , ( this part is actually ongoing).

La 7β-hydroxy-épiandrostérone dans des modèles in vitro de cancer du sein : effets anti-estrogéniques et rôle des récepteurs des estrogènes / The DHEA metabolite 7β-hydroxy-epiandrosterone exerts anti-estrogenic effects on breast cancer cell lines

La 7β-hydroxy-épiandrostérone, stéroïde endogène dérivant de la DHEA, présente des propriétés anti-inflammatoires. En effet, elle module la voie des prostaglandines (PGs) en inhibant la production de la PGE2 pro-inflammatoires et en augmentant la production de la 15-Deoxy-∆12,14-PGJ2 cyto-protectrice in vivo et in vitro. Les faibles doses de 7β-hydroxy-épiandrostérone (1nM, 10nM, 100nM) pour lesquelles ces effets sont observés, suggèrent une liaison à un récepteur spécifique. L’inflammation et la production des PGs jouent un rôle important dans le développement et la prolifération des tumeurs mammaires estrogéno-dépendantes. Le 17β-estradiol (E2), en se fixant sur les récepteurs des estrogènes (REs), induit la production de PGE2 et la prolifération cellulaire dans ces cellules tumorales. De ce fait, notre objectif était de tester les effets de la 7β-hydroxy-épiandrostérone sur la prolifération (comptage avec exclusion au bleu trypan), le cycle cellulaire et l’apoptose (cytométrie de flux) dans les lignées cellulaires de cancer du sein MCF-7 (REα+, REβ+, GPR30+) et MDA-MB-231 (REα-, REβ+, GPR30+) et d’identifier une(des) cible(s) potentielle(s) dans ces cellules (transactivation) et dans des cellules négatives pour les REs nucléaires SKBr3 (GPR30+) (études de prolifération). Cette étude a montré que la 7β-hydroxy-épiandrostérone exerce des effets anti-estrogéniques dans les cellules MCF-7 et MDA-MB-231 associés à une inhibition de la prolifération et un arrêt du cycle cellulaire. Les études de transactivation et de prolifération avec les agonistes spécifiques des REs ont montré une interaction avec le REβ. De plus, les résultats des études de proliférations sur les trois lignées cellulaires suggèrent que la 7β-hydroxy-épiandrostérone pourrait également interagir avec le GPR30. Ces résultats indiquent que ce stéroïde androgène agit comme un anti-estrogène. De plus, c’est la première fois qu’un stéroïde androgène à faible dose montre une action anti-proliférative dans des lignées de cancers du sein. Des études ultérieures restent à réaliser afin de mieux comprendre ces effets observés. / 7β-hydroxy-epiandrosterone, an endogenous androgenic derivative of DHEA, has previously been shown to exert anti-inflammatory action in vitro and in vivo via a shift from prostaglandin E2 (PGE2) to 15-deoxy-∆12,14-PGJ2 production. This modulation in prostaglandin production was obtained with low concentrations of 7β-hydroxy-epiandrosterone (1–100 nM) and suggested that it might act through a specific receptor. Inflammation and prostaglandin synthesis is important in the development and survival of estrogen-dependent mammary cancers. Estrogen induced PGE2 production and cell proliferation via its binding to estrogen receptors (ERs) in these tumors. Our objective was to test the effects of 7β-hydroxy-epiandrosterone on the proliferation (by counting with trypan blue exclusion), cell cycle and cell apoptosis (by flow cytometry) of breast cancer cell lines MCF-7 (ERα+, ERβ +, G-protein coupled receptor 30 : GPR30+) and MDA-MB-231 (ERα-, ERβ +, GPR30+) and to identify a potential target of this steroid in these cell lineages (by transactivations) and in the nuclear ER-negative SKBr3 cells (GPR30+) (by proliferation assays). 7β-hydroxy-epiandrosterone exerted anti-estrogenic effects in MCF-7 and MDA-MB-231 cells associated with cell proliferation inhibition and cell cycle arrest. Moreover, transactivation and proliferation with ER agonists assays indicated that 7β-hydroxy-epiandrosterone interacted with ERβ. Data from proliferation assays on the MCF-7, MDA-MB-231 and SKBr3 cell lines suggested that 7β-hydroxy-epiandrosterone may also act through the membrane GPR30 receptor.These results support that this androgenic steroid acts as an anti-estrogenic compound. Moreover, this is the first evidence that low doses of androgenic steroid exert antiproliferative effects in these mammary cancer cells. Further investigations are needed to improve understanding of the observed actions of endogenous 7β-hydroxy-epiandrosterone.

A biodiversidade de plantas brasileiras é uma fonte muito rica de moléculas bioativas, dentro da proposta da busca por novas drogas antitumorais, avaliamos neste estudo o potencial antiproliferativo do composto 7-epi-clusianona. Foram utilizadas duas linhagens celulares derivadas de tumor de mama humana, Hs 578T e MCF-7, cultivadas em monocamada e como esferoides. O IC50 após 48 horas de tratamento das células é de 20 &mu;M para Hs 578T e 6 &mu;M para MCF-7. A análise do ciclo celular mostrou que o composto é capaz de reter as células em fase G1/G0 em ambas as linhagens em 2D, mas não em 3D. O composto é capaz de induzir as células a senescência celular, como mostrado pelo ensaio de detecção de &beta;-galactosidase. Esses dados indicam que o composto 7-epi-clusianona é uma molécula promissora, que demonstrou potencial antitumoral em células de tumor de mama. A cultura tridimensional se mostrou mais resistente ao tratamento com 7-epi-clusianona, portanto estudos mais abrangentes são necessários para melhor entendimento dos efeitos do composto sobre esse tipo de cultura. / Brazilian flora is considered one of the most diverse in the world and natural products are some of the important sources of new antitumoral compounds. The aim of this study was to evaluate the antiproliferative potential of 7-epi-clusianone. Two cell lines derived from human breast tumor were used, Hs 578T and MCF-7, cultured in monolayer and as spheroids. The IC50 after 48 hours of treatment is 20 &mu;M to Hs 578T cells and 6 &mu;M to MCF-7 cells.
