Vacinas formuladas com nanopartículas de quitosana têm sido utilizadas como sistema de entrega de antígenos pela via mucosa e imunopotenciador. O objetivo do presente estudo foi desenvolver e avaliar a eficácia de uma vacina inativada formulada com o genótipo BR-I do VBI encapsulado em nanopartículas de quitosana (VBI-CS) administrada por via mucosa em aves. Além disso, objetivou-se comparar a eficácia da vacina desenvolvida com a de uma vacina inativada convencional do mesmo vírus incorporada a um adjuvante oleoso (Montanide ISA 71) (VBI-O), administrada por via intramuscular. Foram avaliadas as respostas imunes humoral e celular (RIC) nos compartimentos de mucosa e sistêmico induzidas pelas vacinas VBI-CS e VBI-O quando associadas ou não a uma vacina viva atenuada da estirpe H120 do VBI. A proteção induzida pelos diferentes protocolos vacinais foi avaliada após o desafio com a estirpe homóloga virulenta do genótipo BR-I. A vacina VBI-CS usada sozinha induziu altos níveis de anticorpos IgA anti-VBI na mucosa e também a expressão dos genes de RIC (CD8β, Granzima A e IFNγ) na traqueia, enquanto que associada a uma vacina viva atenuada heteróloga, induziu maiores níveis de anticorpos IgG anti-VBI (mucosa e sistêmico) e RIC caracterizada pelo aumento da expressão dos genes CD8β, Granzima A e IFNγ, principalmente na traqueia. Vaccines formulated with chitosan nanoparticles have been used as an antigen delivery system in mucosal and immunopotentiator. The aim of this study was to develop and evaluate the efficacy of an inactivated vaccine formulated with IBV BR-I genotype encapsulated in chitosan nanoparticles (IBV-CS) administered by the mucosal route in chickens. In addition, this study aimed to compare the efficacy of the developed vaccine with a conventional inactivated vaccine of the same virus incorporated into oily adjuvant (Montanide ISA 71) (IBV-O) administered intramuscularly. The humoral and cellular (CMI) mediated immune responses in the mucosal and systemic compartments induced by IBV-CS and IBV-O vaccines were evaluated, when these vaccines were associated or not with a live attenuated H120 serotype vaccine. The protection induced by the different vaccine protocols was evaluated after the challenge with the virulent homologous strain of BR-I genotype.
