A caracterização no estado sólido de insumos farmacêuticos constitui uma parte muito importante no entendimento de suas propriedades físicas, químicas e farmacológicas. A partir da análise estrutural por difração de raios X em monocristal, pode-se identificar a conformação no estado sólido do fármaco, assim como sua densidade eletrônica. Estes estudos podem ser complementados com dados provenientes da Modelagem Molecular, que compreende um número de ferramentas e métodos computacionais e teóricos que têm como objetivos compreender e prever o comportamento de sistemas reais. Nesse contexto, visando compreender melhor as propriedades de estado sólido apresentadas pelo fármaco anti-filariose dietilcarbamazina (DEC), foi obtido um novo sal, o maleato de dietilcarbamazina (DEC maleato), cujas propriedades foram comparadas com as das estruturas já reportadas, DEC citrato e DEC pura. A DEC maleato foi caracterizada por difração de raios X em monocristal, espectroscopias Raman e Infravermelho e análise térmica. A DEC maleato cristaliza no grupo espacial triclínico PI com dois confôrmeros da molécula de DEC na unidade assimétrica, ambos exibindo caudas etílicas na conformação syn em relação ao anel piperazina, diferentemente do que ocorre na DEC citrato e na DEC pura, nas quais esses fragmentos moleculares apresentam uma conformação anti. A principal interação intermolecular entre o fármaco e o ácido maleico é do tipo N-H•••O, que caracteriza a formação do sal e, consequentemente, do par iônico (DEC)+(maleato)-. Além disso, uma rede complexa de interações intermoleculares não-clássicas do tipo C-H•••O estão presentes entre as moléculas de DEC, DEC-maleato e maleato-maleato, levando a um empacotamento cristalino na forma de um sanduíche, onde os confôrmeros da DEC acomodam-se em colunas intercaladas por bicamadas de íons maleato. Não foram observadas transições de fase estruturais em função da temperatura entre 100 K e temperatura ambiente. No entanto, devido à variação conformacional observada entre as moléculas de DEC, cálculos quânticos foram realizados na fase gasosa, otimizando as conformações moleculares tanto da molécula de DEC neutra quanto da carregada a fim de determinar as características de sua estrutura eletrônica utilizando o método da Teoria do Funcional de Densidade, com o funcional híbrido B3LYP e o conjunto de função de base 6-31++G. Através dos cálculos teóricos foram obtidas quatro novas conformações, uma para DEC neutra e três da carregada, para as quais analisou-se as energias de conformação, os espectros vibracionais simulados e por fim os mapas de potencial eletrostático e os orbitais de fronteira. / The solid state characterization of active pharmaceutical ingredients (API) constitutes an important part in understanding their physical, chemical and pharmacological properties. From the structural analysis by single crystal X-ray diffraction, the API conformation in the solid form, as well as its electronic density, can be identified. These studies can be supplemented with data from the Molecular Modeling, which includes a number of theoretical and computational tools used to understand and to predict the behavior in real systems. In this context, aiming to better comprehend the solid state properties exhibited by the anti-filarial drug diethylcarbamazine (DEC), a new salt was obtained, the diethylcarbamazine maleate (DEC maleate), and its properties were compared with the ones of the reported structures, DEC citrate and pure DEC. The DEC maleate was characterized by single crystal X-ray diffraction, infrared and Raman spectroscopy and thermal analysis. DEC maleate was found to crystallize in the triclinic space group PI with two very similar conformers of the DEC molecule in the asymmetric unit, both exhibiting the ethylic chains in conformation syn in relation to the piperazine ring, unlike what happens to DEC citrate and pure DEC, where these chains are anti related. The main intermolecular interaction between the API and the maleic acid is of the type NH•••O, characterizing the salt formation, and thus, the ionic pair (DEC)+(maleate)-. Moreover, a complex network of no-classical intermolecular interactions of the type CH•••O occur between DEC-DEC, DEC-maleate, and maleate-maleate molecules, leading to a sandwich like crystal packing, where DEC conformers are accommodated in columns intercalated by maleates bilayers. No phase transitions were observed for the molecule structure in function of temperature between 100 K and room temperature. However, due conformational variations observed among DEC molecules of the three structures, quantum calculations were performed in the gas phase, optimizing the molecular conformations of both, the neutral and the charged DEC molecules to determine the characteristics of the electronic structure using the method of Density Functional Theory with the B3LYP hybrid functional and basis set 6-31++G. new conformations it were found, for which geometrical characteristics, conformation energies, vibrational spectra simulation and finally the electrostatic potential maps and the frontier orbitals, were analyzed.

As moléculas Prodan (6-propionil-2-dimetilamino-naftaleno) e Laurdan (6- dodecanoil-2-dimetilamina-naftaleno) são muito usadas como sondas fluorescentes em bicamadas lipídicas e, mais raramente, em membranas celulares. Esta tese de doutorado teve por objetivo realizar um estudo dessas sondas em diferentes ambientes, para ampliar o entendimento de suas estruturas e características espectroscópicas em diferentes solventes e em bicamadas lipídicas. Este estudo foi feito utilizando duas abordagens, uma experimental e outra teórica. Com resultados teóricos e experimentais, mostramos que o cromóforo é o mesmo para as duas moléculas. Experimentalmente, foram medidos e comparados os espectros de absorção eletrônica em solventes de diferentes polaridades e em bicamadas lipídicas. Foi construído um modelo teórico para o estado fundamental do Prodan em diferentes solventes, onde verificamos que a geometria molecular é planar e a molécula sofre uma grande polarização em solventes de maior polaridade. Teoricamente, reproduzimos os espectros de absorção em todos os solventes, mostrando que eles são compostos por três transições eletrônicas. Verificamos, experimental e teoricamente, que o Prodan forma agregados quando colocado em solução aquosa, em todas as concentrações estudadas (0.9 a 20.0 uM). Adicionalmente, foram medidos e comparados os espectros de emissão fluorescente, e mostramos que eles apresentam duas bandas de emissão em todos os solventes estudados e em bicamada lipídicas. Essas bandas estão relacionadas com dois tempos de vida, evidenciando a presença de dois estados excitados para essas sondas. Porém, a natureza dos estados excitados em solventes homogêneos e em bicamadas lipídicas parece ser diferente. Uma metodologia de análise dos espectros de emissão fluorescente foi proposta. A comparação dos espectros de emissão do Prodan e do Laurdan em bicamadas lipídicas mostrou que as sondas monitoram micro-ambientes ligeiramente diferentes. Nossos resultados indicam que os dois fluoróforos posicionam-se próximos à superfície da bicamada lipídica, porém o Prodan parece estar mais exposto à água. Além disso, o Prodan apresenta uma pequena partição em água, quando a bicamada está na fase gel. / Prodan (6-propionyl-2-dimethylamino-naphthalene) and Laurdan (6-dodecanoyl-2- dimethylamino-naphthalene) molecules are frequently used as fluorescent probes in lipid bilayers and, more rarely, in cell membranes. The objective of this PhD thesis was to study these probes in several environments, to increase the understanding about its structures and spectroscopic characteristics in several solvents and lipid bilayers. This study was carried out using two approaches: experimental and theoretical. With experimental and theoretical results, we demonstrated that the two molecules have the same chromophore. Experimentally, electronic absorption spectra were measured and compared in solvents of several polarities and lipid bilayers, for both molecules. A theoretical model was constructed for Prodans fundamental state in several solvents, in which we found that its molecular geometry is planar and that the molecule undergoes a great deal of polarization in solvents of higher polarity. Theoretically, we emulated the absorption spectra in all solvents studied, showing that they are well described by three electronic transitions. We verified, experimental and theoretically, that Prodan aggregates in aqueous solution at all concentration ranges (0.9 a 20.0 uM). Additionally, the fluorescent emission spectra were measured and compared, and we demonstrated that they are composed by two emission bands in all solvents and lipid bilayers. These bands were related to two fluorescent lifetimes, evidencing the presence of two excitation states for these probes. However, the nature of the excited states in homogeneous solvents and in lipid bilayers seems to be different. A methodology to analyze the emission spectra was proposed. The comparison of Prodans and Laurdans emission spectra in lipid bilayers showed that the probes monitor a slightly different micro-environment. A Emodina (EMH) é uma das antraquinonas mais abundantes na natureza. Essa molécula vem sendo largamente usada como material de estudo científico por apresentar diversas atividades farmacológicas, tais como antiviral, antitumoral, antifungal, digestiva e outras. É conhecido que a Emodina em solução aquosa alcalina pode sofrer mais de um processo de desprotonação, apresentando-se na forma desprotonada, EM-, após a primeira desprotonação. Nesta tese de doutorado estudamos as propriedades estruturais e eletrônicas da molécula Emodina em meio solvente e em bicamadas lipídicas a fim de caracterizar as propriedades relacionadas à espectroscopia UV-Vis, à reatividade e à termodinâmica dessa molécula nesses ambientes. Realizamos cálculos quânticos com a Emodina em vácuo e em meio solvente, onde consideramos todos os possíveis sítios de desprotonação. Como resultados desses cálculos, identificamos os sítios da primeira, segunda e terceira desprotonação. Calculamos o pKa1 da Emodina em água e o pK*a1 em metanol através de simulações computacionais com o método Monte Carlo e cálculos quânticos, com o solvente descrito com o modelo contínuo polarizável. Nossos melhores valores para o pKa1 da Emodina determinados nesses solventes foram 8.4±0.5 e 10.3±1.5, que estão em boa concordância com os valores experimentais, (pKa1=8.0±0.2 e pK*a1=11.1±0.1) obtidos nesta tese para Emodina em água e metanol, respectivamente. Adicionalmente realizamos simulações com Dinâmica Molecular com as espécies EMH e EM- em bicamada lipídica de DMPC, para investigar a nível atômico as interações dessas espécies com a bicamada e determinar as posições preferenciais dessas espécies nesse ambiente anfifílico. Os resultados dessas simulações mostraram que as espécies EMH e EM- ficam inseridas na bicamada, na região polar dos lipídios, próximos aos gliceróis. Esses resultados corroboram as nossas medidas do espectro de absorção dessas espécies em bicamada lipídica, onde mostramos de forma qualitativa, que ambas as espécies ficam inseridas na bicamada, na região das cabeças polares dos lipídios. A análise das propriedades estruturais da bicamada na vizinhança das espécies da Emodina como área por lipídio e densidade eletrônica dos lipídios, mostrou que o efeito da EM- na estrutura da bicamada lipídica é maior do que o da EMH. Esses resultados corroboram as nossas medidas de DSC(Differential Scanning Calorimetry) das espécies da Emodina na bicamada. / Emodin (EMH) is one of the most abundant anthraquinone derivatives found in nature. This molecule has been used widely as research material, due to its biological and pharmacological activities such as antiviral, anticancer, antifungal, digestive and antibacterial activities. It is known that Emodin in alkaline aqueous solution can undergo more than one deprotonation, leading to the specie EM- in the first deprotonation process. In this PhD thesis, we studied the structural and electronic properties of this molecule in several solvents and lipid bilayers, in order to characterize the properties related to UV-Vis absorption spectroscopy, reactivity and thermodynamics of this molecule in these environments. Performing quantum mechanics (QM) calculations for all possible deprotonation sites and tautomeric isomers of Emodin in vacuum and in water, we identified the sites of the first, second and third deprotonations. We calculated the pKa1 of Emodin in water and pK*a1 in methanol with free energy perturbation method, implemented in the Monte Carlo simulation, and with QM calculations, where the solvent was treated as a polarizable continuum medium. Our best values for pKa1 of Emodin in these solvents were 8.4±0.5 and 10.3±1.5, which are in very good agreement with the experimental values obtained in this thesis pKa1=8.0±0.2 and pK*a1=11.1±0.1, for water and methanol, respectively. Additionally, we performed molecular dynamics simulations of both species in fully hydrated lipid bilayers of DMPC to investigate at atomic detail the molecular mechanism of the interaction of these species with lipid membrane and its preferred positions in this amphiphilic environment. As results of these simulations, we obtained that both species of Emodin have a strong tendency to insert into the lipid bilayer, remaining near the glycerol group of DMPC. These results corroborate our measured absorption spectra of these species in the bilayer, which qualitatively showed that both species are within the bilayer, inserted in the lipid headgroup region. Our results also show that the effect of EM- specie in the lipid bilayer structure is stronger than the EMH, which corroborate our DSC(Differential Scanning Calorimetry) measurements.

Os resultados dessas simulações mostraram que as espécies EMH e EM- ficam inseridas na bicamada, na região polar dos lipídios, próximos aos gliceróis. Esses resultados corroboram as nossas medidas do espectro de absorção dessas espécies em bicamada lipídica, onde mostramos de forma qualitativa, que ambas as espécies ficam inseridas na bicamada, na região das cabeças polares dos lipídios. A análise das propriedades estruturais da bicamada na vizinhança das espécies da Emodina como área por lipídio e densidade eletrônica dos lipídios, mostrou que o efeito da EM- na estrutura da bicamada lipídica é maior do que o da EMH. Esses resultados corroboram as nossas medidas de DSC(Differential Scanning Calorimetry) das espécies da Emodina na bicamada. / Emodin (EMH) is one of the most abundant anthraquinone derivatives found in nature. This molecule has been used widely as research material, due to its biological and pharmacological activities such as antiviral, anticancer, antifungal, digestive and antibacterial activities. It is known that Emodin in alkaline aqueous solution can undergo more than one deprotonation, leading to the specie EM- in the first deprotonation process. In this PhD thesis, we studied the structural and electronic properties of this molecule in several solvents and lipid bilayers, in order to characterize the properties related to UV-Vis absorption spectroscopy, reactivity and thermodynamics of this molecule in these environments. Performing quantum mechanics (QM) calculations for all possible deprotonation sites and tautomeric isomers of Emodin in vacuum and in water, we identified the sites of the first, second and third deprotonations. We calculated the pKa1 of Emodin in water and pK*a1 in methanol with free energy perturbation method, implemented in the Monte Carlo simulation, and with QM calculations, where the solvent was treated as a polarizable continuum medium. Our best values for pKa1 of Emodin in these solvents were 8.4±0.5 and 10.3±1.5, which are in very good agreement with the experimental values obtained in this thesis pKa1=8.0±0.2 and pK*a1=11.1±0.1, for water and methanol, respectively. Additionally, we performed molecular dynamics simulations of both species in fully hydrated lipid bilayers of DMPC to investigate at atomic detail the molecular mechanism of the interaction of these species with lipid membrane and its preferred positions in this amphiphilic environment. As results of these simulations, we obtained that both species of Emodin have a strong tendency to insert into the lipid bilayer, remaining near the glycerol group of DMPC. These results corroborate our measured absorption spectra of these species in the bilayer, which qualitatively showed that both species are within the bilayer, inserted in the lipid headgroup region. Our results also show that the effect of EM- specie in the lipid bilayer structure is stronger than the EMH, which corroborate our DSC(Differential Scanning Calorimetry) measurements.
