Análise da expressão de MMP-2 e MMP-9 na pele de cães com Leishmaniose visceral

A pele é o primeiro órgão infectado pelo parasito na LVC. As enzimas metaloproteinases da matriz (MMPs) atuam na degradação da matriz extracelular (MEC) e na modulação da resposta inflamatória, frente a vários tipos de injúrias. O objetivo deste estudo foi avaliar a expressão de MMP-2 e MMP-9, por meio da técnica de imuno-histoquímica e zimografia na pele de cães naturalmente acometidos por LVC e comparar estes resultados com a imunodetecção das citocinas TNF- e TGF- e do protozoário Leishmania, com as possíveis alterações da matriz extracelular dermal. A alteração da composição de fibras colágenas da derme foi avaliada pela coloração PicroSirius. O método foi usado para diferenciar fibras colágenas dos tipos I e III, nas regiões do focinho, orelha e abdômen. A carga parasitária, intensidade de inflamação e a produção de MMP 2 e 9 foram maiores nas regiões cutâneas da orelha e focinho. Os macrófagos, linfócitos e plasmócitos predominaram no infiltrado inflamatório da derme superficial, formando granulomas, associados a degradação do colágeno maduro (tipo I) e a discreta deposição de colágeno jovem (tipo III), com maior intensidade nos cães com elevada carga parasitária cutânea. O TGF-β foi detectado nos três grupos de cães infectados (assintomático, oligossintomático e sintomático), sem diferenças significativas entre eles. O TNF-α não foi detectado na maioria dos animais infectados e no grupo controle. Conclui-se que quanto maior a carga parasitária e a intensidade de inflamação na pele, maior a degradação do colágeno maduro, produção de MMPs 2 e 9 e produção de TGF-β. A MMP-9 ativa poderia ser utilizada... / Dogs infected by Leishmania (Leishmania) chagasi may or may not show clinical signs of the zoonosis and they are the main source of infection for man. The skin is the first organ infected by the parasite in the LVC. The enzyme matrix metalloproteinases (MMPs) act on the degradation of extracellular matrix (ECM) and in the inflammatory response modulation against many kinds of injuries. The aim of this study was to evaluate the expression of MMP-2 and MMP-9 by immuno-histochemistry and zymography in the skin of dogs naturally affected by LVC and the immunodetection of TNF- and TGF- and of Leishmania and compare these results with the possible alterations of dermal extracellular matrix. The method was used to differentiate collagen fibers in the dermis was evaluated by Picrosirius red staining to differentiate collagen types I and III, in regions of the nose, ear and abdomen. The parasite load, intensity of inflammation, increased production of MMP 2 and 9 were higher in regions of the ear and muzzle. Macrophages, lymphocytes and plasma cells predominated in the inflammatory infiltrate of the superficial dermis, forming granulomas, associated with degradation of mature collagen (type I) and the discrete deposition of the young collagen (type III), with greater intensity in dogs with high parasite load skin. The TGF-β was detected in all groups of infected dogs (asymptomatic, oligosymptomatic and symptomatic), no significant differences among them. The TNF-α was not detected in most infected animals and the control group. It follows that the greater is the intensity of parasitic load and inflammation of the skin, the greater is the degradation of mature collagen, the production of MMPs and 2 and 9, production of TGF-β. Níveis de IgG anti-Leishmania e perfil de citocinas em cães machos e fêmeas assintomáticos naturalmente infectados por Leishmania (L.) Chagasi

Na (LV) estudos em modelos experimentais, têm mostrado diferenças na resposta imunológica entre macho e fêmea. Na (L VC) tais estudos, não foram realizados. Este trabalho investigou em cães assintomáticos machos e fêmeas, naturalmente infectados por Leishmania (L.) chagasi, os níveis séricos de IgG contra antígenos totais de Leishmania, o nível de IL-10 e IFN-y, no sobrenadante do extrato do baço e fígado, e a citocina regulatória TGF-j3 no sobrenadante do extrato do baço e fígado, e sua produção natural no sobrenadante de cultura de células esplênicas. Os níveis de anticorpos anti-L. (L.) chagasi da classe IgG nos cães assintomáticos machos e fêmeas, não apresentaram diferença significativa entre o sexo. O nível de IL-10 mostrou-se elevado, no sobrenadante do extrato do fígado em cães infectados machos e fêmeas assintomáticos e apresentou dominância marcante em relação às outras citocinas. O nível do TGF-j3 mostrou-se aumentado no extrato do baço. O nível de IFN-y no baço dos grupos avaliados foi quantitativamente menor que as citocinas IL-10 e TGF-B. diferindo do observado no fígado onde foi observado uma baixa produção de TGF-j3 em relação ao IFN-y. No baço, o IFN-y apresentou diferença significativa em fêmeas assintomáticas quando comparadas com os machos assintomáticos. A observação da predominância das citocinas TGF-j3 no baço, e IL-10 no baço e fígado, em modelo ex vivo, sugere uma polarização da resposta imunológica para o padrão Th2 em cães naturalmente infectados por Leishmania (L.) chagasi. / The dog is the main reservoir of Leíshmanía (Leíshmanía) chagasí, the parasite responsible for Visceral Leishmaniasis (VL) in the Americas. Experimental studies on VL have shown differences in the immune response of males and females. Such studies have not been carried out on canine visceral leishmaniasis. This study investigated the seric levei of IgG against Leíshmanía total antigens, the levels of IL-10 and IFN-y in the supernatant of spleen and liver extracts, the regulatory cytokine TGF-131 in the supernatant of spleen and liver extracts, and its natural production in the supernatant of spleen cell culture, in male and female asymptomatic dogs naturally infected by Leíshmanía (L.) chagasi. The levels of anti-L. (L.) chagasí IgG antibodies in male and female asymptomatic dogs did not differ significantly between the sexes. IL-10 levei was high in the supernatant of the liver extract of male and female asymptomatic infected dogs and predominated over the other cytokines. TGF-131 levei was increased in the spleen extract. The levei of IFN-y in the spleen of the group evaluated was quantitatively lower than the IL-10 and TGF-131 cytokine levels, different from what was observed in the liver, where a low productíon of TGF-131 was observed when compared to IFN-y. In the spleen, IFN-y presented a significant difference in the asymptomatic females when compared to the asymptomatic males. The observation of a predominance of TGF-131 cytokines in the spleen and of IL-10 in the spleen and liver of an ex vívo model suggests a polarization of the immune response towards the Th2 pattern in dogs naturally infected by Leíshmanía (L.) chagasí.

Aspectos clínicos, epidemiológicos e laboratoriais de cães sororreagentes para leishmaniose visceral, em foco de transmissão no Distrito Federal-DF, Brasil

No Brasil, é causada pela Leishmania (Leishmania) chagasi, que tem flebotomíneos da espécie Lutzomyia longipalpis como vetores e grande variedade de mamíferos, inclusive pessoas, como hospedeiros. As infecções mais comuns ocorrem em mamíferos silvestres; em áreas urbanas e periurbanas, o cão é o hospedeiro de maior importância epidemiológica. Assim, o objetivo do estudo foi verificar aspectos clínicos e epidemiológicos, e avaliar a bioquímica sérica, inclusive proteínas de fase aguda, em proteinograma em gel de poliacrilamida contendo dodecil sulfato de sódio (SDS-PAGE), de 120 cães de região periurbana com foco de transmissão no Distrito Federal. Sessenta cães eram sororreagentes e sessenta animais não eram reagentes à reação de imunofluorescência indireta (RIFI) para LVC. A soroprevalência observada foi de 24,5%, sendo animais assintomáticos a maioria entre os cães sororreagentes ou não-reagentes à RIFI para LVC. Nos animais sororreagentes oligossintomáticos, os sinais clínicos mais evidentes foram lesões cutâneas (primárias e secundárias), inclusive alopecia de nariz e/ou orelhas. As médias das concentrações séricas de uréia, creatinina, colesterol, cálcio, fósforo, magnésio e das atividades de AST, ALT, ALP e GGT situaram-se na faixa de normalidade para a espécie canina. Os teores séricos de colesterol e de HDL foram inferiores e as concentrações séricas de proteínas totais e imunoglobulinas foram mais elevadas em cães sororreagentes à RIFI. Notou-se uma proteína de peso molecular 33 kDa em cães sororreagentes à RIFI, não constatada em cães soronegativos. Ainda, em animais sororreagentes, evidenciaram-se teores séricos superiores embora não significativos, mas com provável importância biológica das proteínas de fase aguda ceruloplasmina, transferrina, haptoglobina e glicoproteína ácida. / Canine visceral leishmaniasis (CVL) is an anthropozoonotic infection of cosmopolitan distribution. In Brazil, it is caused by Leishmania (Leishmania) chagasi and transmitted by vector Lutzomyia longipalpis (sandfly), having a great variety of hosts (including humans). The most common infections take place in wild mammals; in urban and periurban areas, dogs are the most important epidemiological host. The aim of this study was to verify clinical and epidemiological aspects of 120 dogs from the periurban area of transmission at Distrito Federal. Serum biochemical evaluation (including acute phase proteins) was performed using SDS-PAGE method. Sixty dogs were seroreagents and sixty were not reactive to the indirect immunofluorescence assay (IFA) for CLV. Seroprevalence rate was of 24.5%; asymptomatic dogs were the majority of seroreagent or non-reagent to IFA for CLV. In the seroreagent oligosymptomatic animals the most evident clinical signs were skin lesions (primary and secondary) including alopecy of nose and/or ears. Mean concentration of urea, creatinine, cholesterol, calcium, phosphorus, magnesium and the activities of AST, ALT, ALP and GGT were within normal limits for canine specie. Serum cholesterol and HDL were inferior and total serum protein concentration and immunoglobulins were more elevated in dogs seroreagent to IFA. In the dogs seroreagent to IFA, it was noticed a protein with molecular weight of 33 kDa, not noticed in seronegative animals. In the seroreagent animals, it was noticed increased serum levels of acute phase proteins (ceruloplasmine, transferrin, haptoglobin and acid glycoprotein), although not significative but with a probable biological importance.
