Développement de sondes chimiluminescentes pour la détection d'activités enzymatiques / Development of chemiluminescent probes for the detection of enzymatic actvitySolmont, Kathleen 16 March 2018 (has links)
Since several years, it is easier to exploit biological phenomena in cellulo through technologies dealing with bioimaging. This method gathers fluorescence, bioluminescence and chemiluminescence. Even though fluorescence is the most employed technic, bioluminescence and chemiluminescence, being the consequence of (bio)chemical reaction, have been widely studied for decades. In fact, they can avoid the main problems encountered in fluorophore use: exciting light. Chemiluminescence is thus the appropriate approach to avoid biological autofluorescence. 1,2-dioxetane is one of the moieties that, upon decomposition, can generate an excited state on a connected fluorophore, giving rise to its intrinsic luminescence. The aim of our project was to develop and study new 1,2-dioxetan chemiluminescent probes based on phenol or naphthol moieties, for targeted enzymatic complexes detection with in cellulo and in vivo bioimaging. The strategy relied on the synthesis of chemiluminescent scaffolds comprising thermally stable 1,2-dioxetan, on which the trigger and the connected NIR fluorophore can be easily diversified. Two methods have been attempted: 1) coupling to a water-soluble version of a known fluorophore (i.e. Nile red) allowing an energy transfer through bonds (i.e. TBET) 2) connection with a lanthanide complex giving rise to an energy transfer through space (i.e. CRET).

Creating Virginia’s War Memorial Carillon in the Shadow of the Civil WarLambertz, Witek Jessica 17 April 2014 (has links)
This thesis explores the creation of the Virginia’s War Memorial Carillon in relation to the persistent significance of the Civil War in Virginia. By focusing on the debates about Virginia’s War Memorial Library, Tomb of the Unknown Soldier, and Carillon, this thesis reconstructs the sentiments and historical legacy that shaped the multitude of decisions leading to the creation of Virginia’s World War memorial. Virginia seriously considered two other proposals for war memorials—the War Memorial Library and the Virginia Tomb of the Unknown Soldier—before ultimately constructing the War Memorial Carillon. Concerns for appropriateness and residual attachments to the Lost Cause of the Civil War inform the debates about the best memorial to represent Virginia and honor the soldiers, sailors, marines, and women from Virginia who fought in the World War.

Previous issue date: 2000-08-31 / Petr?leo Brasileiro SA - PETROBRAS / Through an integrated approach, using litho, chrono and biostratigraphic data, the relative importance of climate variations and tectonics were recognized in rift sediments of the onshore Potiguar Basin, Northeast Brazil. Concepts of sequence stratigraphy were applied as a template to integrate sedimentological and geochemical data (oxygen isotopes), as well as quantitative palynologic methods to address and recognize the main depositional patterns produced in a rift basin. The main objective was to address the relative importance of climate changes and tectonics to the resultant stratigraphic architecture. The results of computer simulations of sedimentary basin fills of rift basins were quite useful to test working hypothesis and mimic the process of filling a half graben during a rift event. The studied section includes a neovalanginian-eobarremian (Lower Cretaceous) rift interval from the Pend?ncia Formation, located in the southwestern portion of Umbuzeiro Graben, in the offshore Potiguar Basin. The depositional setting is interpreted as progradational deltaic system entering a lake from its flexural margin. Sismoestratigraphyc and well logs analyses allowed to interpret two regressive intervals (Green and Yellow Sequences), separated by a broad transgressive interval (Orange Sequence), known as the Livramento Shale. The depositional history encompass
three stages: two tectonically active phases, during the deposition of the Green and Yellow Sequences, and a tectonically quiescent phase, during the deposition of the Orange Sequence. Paleoclimatic interpretation, based on quantitative palynology and geochemical data (18O), suggests a tendency to arid conditions during the tectonically active phases and wet conditions during the tectonically quiescent phase.

Previous issue date: 2012-08-06 / Conselho Nacional de Desenvolvimento Cient?fico e Tecnol?gico / The Northeast relief was described by the Pediplanation Model. This action discards the
theoretical basis of post-Cretaceous tectonic evolution of the landscape. Through this
model the Massif Pereiro - MP, Borborema Province, was established as part of the
Tablelands Area Residual Sertanejos. The present work aims to establish the post-
Cretaceous morphotectonic evolution of the MP by geomorphological and geological
mapping using Geographic Information System, Remote Sensing and dating of
sediments by Single Aliquot Regenerative-dose (SAR). The MP is contained in the core
semi-arid, annual precipitation of 600-800 mm / year. The MP is NE-SW, is limited by
Shear Zone Jaguaribe (ZCJ) and Portalegre Shear Zone (ZCPa), the same attitude, and
crossed by several other shear zones. These shear zones show evidence of brittle
Cenozoic reactivation, mostly as normal faults and shallow crustal level. The
Quaternary sedimentation around the MP focuses on fault escarpments in a general
pattern cascade, where ages decrease from the summits of the steep foothills. The ages
of 51 sediment samples indicate a correlation with global climate following pulses: Last
Interestadial-UI, the Last Glacial Maximum - LGM and the transition Pleistocene /
Holocene, while the latter focus on 18 of 51 samples dated. This study also finds
evidence of a new quaternary basin, here called Merejo Basin. Through these results it
is concluded that no evidence of post-Cretaceous tectonic evolution of morphological
MP, as their retreat along the fault scarps, invariably following the trend of the shear
zones. The erosion of cliffs in large time scale is controlled by weakness zones
generated by faults on the other hand the erosion of cliffs in short time, with the
formation of deposits and colluvial horizons pedogenizados, has climate control. It was
also found that in the study area there is a preponderance of past and current tectonic
erosion processes on the morphological evolution / O relevo do Nordeste foi descrito por meio do modelo de pediplana??o.
dep?sitos coluvionares e horizontes pedogenizados, possui controle clim?tico. Concluise
