E atualmente CVV ? um problema significativo na sa?de da mulher. As ? glucanas s?o polissacar?deos estruturais da parede celular da levedura Saccharomyces cerevisiae e existem v?rios relatos que demonstram o efeito imunomodulador desta estrutura em infec??es bacterianas, virais, f?ngicas e parasit?rias. Sabendo disso, o presente estudo avaliou se a ?-glucana tem atividade imunomoduladora em camundongos com candid?ase vulvovaginal sob a influ?ncia do estr?geno. Os animais foram inoculados com C. albicans, por via intravaginal, e tratados com glucana, vaginal e intraperitonealmente, e os animais do grupo de controle receberam por via intraperitoneal, solu??o salina. Os animais tratados com glucana intraperitoneal e vaginal mostraram menor n?mero de UFC, no fluido vaginal, em compara??o com animais controles. No entanto, os resultados mais marcantes foram do intraperitoneal, confirmando com o histopatol?gico, por?m sem diferen?a estatisticamente significante. Em rela??o ao influxo dos neutr?filos polimorfonucleares (PMNs), os grupos tratados com glucana mostraram maior infiltra??o desses. Al?m disso, observou-se que os animais tratados com glucana apresentaram uma maior quantidade de IFN-?, no lavado vaginal, em compara??o com o controle. Os dados sugerem que glucana pode ter uma atividade importante na prote??o contra C. albicans. / Vulvovaginal candidiasis (VVC) is an inflammatory disease, on vaginal tissue, caused mainly by pathogenic yeasts of Candida albicans. And actually, VVC is currently a significant problem in women's health. The ?-glucans are structural polysaccharides of the cell wall of the fungi Saccharomyces cerevisiae and there are several reports that demonstrate the immunomodulatory effect this structure in infections of bacterial, viral, fungal and parasitic. Knowing this, the present study evaluated whether ?- glucan has immunomodulatory activity in mice with vulvovaginal candidiasis under the influence of estrogen. The animals were inoculated with C. albicans, intravaginally, and treated with glucan, vaginally and intraperitoneally, and the control group animals received saline, intraperitoneally. The animals treated with intraperitoneal and vaginal glucan showed smaller number of the CFU, in the vaginal fluid, compared with control animals. However the results more marked was of the intraperitonel, confirmed with histopathological, but in neither case the results were statistically significant. Relative to influx of the polymorphonuclear neutrophilis (PMNs), the groups treated with glucan showed greater infiltration. Moreover, it was observed that animals treated with glucan showed a higher quantity of IFN-?, in vaginal washed, compared with the control. The data suggest that glucan may have an important activity in protection against C. albicans.

Candid?ase vulvovaginal: sintomatologia, fatores de risco e coloniza??o anal concomitante

No. of bitstreams: 1 AntonioARH.pdf: 1355873 bytes, checksum: c7ebee3518ab1fe810e379bbdf3b4446 (MD5) Previous issue date: 2007-08-31 / Post-menarche patients with clinical signs of vulvovaginitis were analyzed in this study, whose aims were the following: identify the frequency of C. albicans and non C. albicans species and negative results, correlate the vaginal culture for yeast with risk factors and symptomatology; compare positive and negative results for yeast in the vaginal and anal cultures; compare the positive results for C. albicans with other results found in the vaginal and anal cultures; and compare concomitant positivity for C. albicans and non C. albicans in the vaginal and anal cultures. Sample selection occurred between May, 2003 and May, 2005, and included 99 patients from Natal, Brazil. The laboratory methods used consisted of CHROMagar Candida culture medium, thermotolerance test at 42-45?C and hypertonic NaCL, in addition to the classic methods of carbohydrate assimilation and fermentation. We used absolute numbers, percentages, means of central tendency, chi-squared test (χ2) with Yates correction, Fisher s exact test and odds ratio for statistical analysis. The most frequent species was C. albicans in 69% of the cases. The positivity for Candida spp showed an association with the use of tight-fitting intimate clothing and/or synthetics, allergic diseases and the occurrence of itching, leukorrhea and erythema. Anal colonization increased the likelihood of vaginal contamination by 2.8 and 4.9 times, respectively, for Candida spp and C. albicans. When compared to the other species, C. albicans-positive anal colonization increased by 3.7 times the likelihood of vaginal positivity. These data suggest likely vaginal contamination originating in the anus / Pacientes ap?s a menarca, com quadro cl?nico compat?vel com vulvovaginite foram analisadas no presente estudo, cujos objetivos foram: identificar a frequ?ncia de esp?cies de C. albicans, n?o C. albicans e resultados negativos; correlacionar a cultura vaginal para leveduras com fatores de risco e sintomatologia; comparar resultados positivos com os negativos para leveduras nas culturas vaginais e anais; comparar os resultados positivos para C. albicans com outros resultados encontrados nas culturas vaginais e anais; comparar a positividade concomitante para C. albicans e n?o C. albicans nas culturas vaginais e anais. A sele??o da casu?stica ocorreu entre maio de 2003 e maio de 2005, sendo inclu?das 99 pacientes, procedentes de Natal, RN. Os m?todos laboratoriais utilizados consistiram em cultivo em CHROmagar Candida, provas de termotoler?ncia a 42-45oC e NaCl hipert?nico, al?m dos m?todos cl?ssicos de assimila??o e fermenta??o de carboidratos. Para a an?lise estat?stica foram utilizados n?meros absolutos, percentuais, medidas de tend?ncia central, Teste do qui-quadrado (χ2) com corre??o de Yates e Teste exato de Fisher, al?m de Odds ratio. A esp?cie mais freq?ente foi C. albicans em 69% dos casos.
