Field survey and culture studies of Caulerpa in Taiwan

¡§ Sea grape ¡¨ named by Japanese is Caulerpa lentillifera having erect branches with spherical ramelli. The length observed in field is 3 cm but may elongate over 10 cm after cultivation. While it is generally recognized by Taiwanese for all edible Caulerpa with spherical ramelli. Caulerpa microphysa collected from Taiwan northeast coast having 3 cm erect branch, but was recorded as 1 cm. To find the Caulerpa with longer erect branch field survey was conducted monthly in Taiwan south coast¡]Hsiao Liu Chiu and Kenting¡^ for one year from October 2006. Six species including C. racemosa var. macrophysa, C. microphysa, C. peltata, C. serrulata, C. sertularioides and C. racemosa var. turbinata were collected. The other two species C. lentillifera and C. racemosa var. occidentalis are acquired from the aquaculture ponds. In total 8 Caulerpa species are cultivated in aquariums with steady flow and nutrients. After cultivation, the morphology of C. racemosa var.macrophysa and C. peltata changed largely. All species with spherical ramelli only had 3-4 cm erect branches, but the C. microphysa strain from the northeast coast could elongat to 12 cm, having the same growth character as the Japanese sea grape. The morphology of the erect branches of C. microphysa changed with the temperature and the illuminance. Caulerpa microphysa could grow in 25-40 psu salinity, at 22-31.5 ¢J and 100-400 £gE/m2/s, the best growth rate¡]26.4 ¡Ó 0.6 ¢H/day¡^found in 35 psu salinity, at 31.5 ¢J and 100 £gE/m2/s. The growth of C. microphysa in deep seawater was better than in the surface seawater, that indicated there are some special nutrients in the deep seawater for the Caulerpa and need to study in the future.

Studies with seaweeds present in the coast of Ceará, such as Caulerpa cupressoides and Pterocladiella capillacea has aroused interest in many biological effects. The Caulerpa cupressoides is a green algae belonging to the family Caulerpaceae and Pterocladiella capillacea is a red seaweed of the family Pterocladiacea. Given the potential of these species and their components such as lectins, as well as a few reports of kidneys toxic effects aimed to study effects of lectins cited algae on renal perfusion model, cytotoxic effects on MDCK cells, effects on Blood Pressure (BP) and Heart Rate(HR). Male Wistar rats weighing between 250 and 300g were used in the experiments. Lectins was utilized in perfusion of isolated kidneys at concentration of 10μg/mL. The effects of lectins were evaluated in Pressure Perfusion (PP), Renal Vascular Resistance (RVR), Urinary Flow (FU), Glomerular Filtration Rate (GFR), Percentage of Tubular Transport of Sodium (% TNa+), Potassium (% TK +) and Chloride (%TCl-). The mesenteric artery assay was realized with the lectin Caulerpa cupressoide- LCc at concentrations of 0.1, 0.3, 1, 3, 10, 30; 60, 100, 200, 300 and 400μg/mL., and the tissue was pre-contracted with phenylephrine (PHE) (0.3μmol and 1μmol). The BP and HR were obtained by cannulation of the carotid artery and registers performed by a data acquisition software. The LCc increased RVR and PP at 90 and 120 min, reduced GFR at 60 minutes and reduced the % TNa+, % TK+ and %TCl- at 60, 90 and 120 minutes. The lectin Pterocladiella capillaceae -LPc reduced the FU at 120 minutes and reduction % TNa+, at 90 and 120 minutes. In the study of cytotoxicity, LCc and LPc did not affect viability of MDCK cells. LCc promoted relaxation in mesenteric artery in concentrations of 100, 200, 300 and 400 µg/mL in tissue pre-contracted with PHE to 1.0 micromol, but did not it in tissue pre-contracted with PHE a 0.3 micromol, in concentrations below to 200 µg/mL. LCc promoted deposition of protein material in the tubule and expansion interstice. The LPc caused deposition protein material in the tubule, without changes at the level of interstice. LCc caused slight reduction in blood pressure at concentrations of 100 and 200μg/mL and did not change the Cardiac Frequency (CF) and LPc caused no changes in CF nor the Heart Rate (HR). It was noted that the LCc promoted renal hemodynamic effects, raising PP and RVR, which may be due to α-adrenoceptor activation. LCc reduced the BP from the concentration of 100 μgmL, suggesting possible release of agents vessel dilators, which also would cause fluid accumulation in the interstice as shown kidney histology. LPc did not change the PP and RVR. The HR and BP were not changed with the LPc, suggesting that it does not promote the release of substances that act directly as a vasoconstrictor. / As macroalgas são encontradas em todo o mundo, desempenhando um importante papel na manutenção do equilibrio marinho e na preservação da biodiversidade marinha. Estudos com espécies de macroalgas presentes no litoral do Ceará, como a Caulerpa cupressoide e Pterocladiella capillacea tem despertado interesse por diversos efeitos biológicos. A Caulerpa cupressoides é uma alga verde da família Caulerpaceae e a Pterocladiella capillacea trata-se de uma alga vermelha da familia Pterocladiacea. Diante do potencial destas espécies e de seus componentes como as lectinas, bem como de poucos relatos sobre efeitos tóxicos renais objetivou-se estudar os efeitos das lectinas das algas citadas em modelo de perfusão renal, efeitos citotóxicos em células MDCK e efeitos na Pressão Arterial (PA) e Frequencia Cardiaca (FC). Utilizou-se ratos wistar machos, pesando entre 250 e 300g. Na perfusão de rins isolados utilizou-se as lectinas na concentração de 10µg/mL. Os efeitos das lectinas foram avaliados na Pressão de Perfusão(PP), Resistência Vascular Renal(RVR), Fluxo Urinário(FU), Ritmo de Filtração Glomerular (RFG), Percentual do transporte tubular de sódio (%TNa+), de potássio (%TK+) e de cloreto (%TCl-). O ensaio com artéria mesentérica foi realizado com a lectina da Caulerpa cupressoides (Lcc) nas concentrações de 0,1; 0,3; 1; 3; 10; 30; 60; 100; 200; 300 e 400 µg/mL e o tecido foi pre-contraído com fenilefrina(PHE) (0,3µmol e 1µmol). A PA e FC foram obtidas por canulação da carótida e os registros realizados através de um software de aquisição de dados. A LCc aumentou a PP e RVR aos 90 e 120 minutos, reduziu o RFG aos 60 minutos e reduziu o %TNa+, %TK+ e %TCl- aos 60, 90 e 120 minutos. A lectina da Pterocladiela capillacea (LPc) reduziu o FU aos 120 minutos e reduziu o %TNa+, aos 90 e 120 minutos. No estudo de citotoxidade, a LCc e LPc não alteraram a viabilidade de células MDCK. A LCc promoveu relaxamento em artéria mesentérica nas concentrações de 100, 200, 300 e 400 µg/mL em tecido pré-contraido com PHE 1 µmol, mas não o fez em tecido pré-contraido com PHE 0,3 µmol abaixo de 200 µg/mL. A LCc promoveu deposição de material protéico nos espaços tubular e urinário e expansão do intersticio. A LPc causou minima desposição de material protéico, sem alterações a nivel de intersticio. A LCc causou leve redução na PA nas concentrações de 100 e 200µg/mL e não alterou a FC e a LPc não causou alterações da PA e FC. Observou-se que a LCc promoveu efeitos hemodinâmicos renais, aumentando PP e RVR o que pode ser em decorrência de ativação de α-adrenoreceptores. A LCc reduziu a PA a partir de100 µg/mL, sugerindo possível liberação de agentes vasodiltadores, o que ocasionaria também acúmulo de líquido no intersticio (edema) como mostrou a histologia dos rins. A LPc não promoveu alterações na PP e RVR. A FC e PA não foram alteradas com a LPc sugerindo que a mesma não promove liberação de substâncias que agem diretamente como vasoconstritores.

A descoberta de metabólitos secundários no ambiente marinho vem consolidando a importância deste ambiente nos últimos anos. A presença de uma grande diversidade de espécies vegetais e animais, pode fornecer uma enorme variedade de substâncias que podem ser usadas na indústria farmacêutica. Dentre todos os organismos marinhos, as algas representam o primeiro elo da cadeia alimentar e possuem um papel muito importante na manutenção da vida aquática, além de serem igualmente relevantes tanto do ponto de vista econômico quanto social. O presente estudo visa realizar prospecção química e microbiológica a partir de extratos de algas marinhas do gênero Caulerpa Lamouroux (Bryopsidades, Chlorophyta). A coleta do material foi realizada na praia de Maracaípe (município de Ipojuca), litoral de Pernambuco, limita-se ao norte pela praia de Porto de Galinhas e ao sul pela praia de Enseadinha. O objetivo principal deste estudo foi realizar uma prospecção química e a avaliação microbiológica dos metabólitos secundários de Caulerpa racemosa (Forsskal) J. Agardh nas condições bentônicas e arribadas, frente aos micro-organismos com maiores índices de infecção hospitalar. O extrato bruto foi caracterizado quanto a sua ação antioxidante (DPPH), além disso, foi realizada uma análise ficoquímica para detecção de saponinas, alcaloides, fenóis, taninos e antocianinas. Foi realizado o isolamento e purificação da fração mais ativa da alga bentônica, onde foram purificadas 4 substâncias, que foram submetidas à elucidação através de técnicas espectroscópicas convencionais (IV, UV e RMN 1H). Os testes antimicrobianos utilizados seguiram o método de difusão de disco e foi constatado que as frações e os extratos brutos das algas bentônicas apresentaram uma marcante atividade frente ao Enterococcus faecalis e cepas resistentes de Enterococcus. Além disso, foi observado que as frações apresentaram uma ação inibitória mais discreta frente a outros representantes de bactérias Gram-positivas. A triagem fitoquímica mostrou a presença de saponinas e alcaloides nesses dois grupos de algas. Os testes antioxidantes revelaram que as algas bentônicas possuem uma ação antioxidante superior as arribadas. A avaliação espectroscópica também indicou a presença de pigmentos clorofilados, esteróides e foi confirmada a presença da caulerpicina.

O presente estudo avaliou as interferências dos extratos hidroalcóolico (EHA - 50 μg/mL) e metanólico (EM - 50 μg/mL) de Caulerpa taxifolia no metabolismo energético e na Angiogênese de embriões de Gallus gallus domésticos L. Ovos vermelhos da raça Rhode Island Red de quatro lotes, foram incubados a 37º C, com viragem automática, para o estudo do metabolismo energético e da angiogênese. Os grupos foram distribuídose divididos em grupos controle, tratados EHA (50 μg/mL) e ME (50 μg/mL), e controle enriquecidos com ω-3, com e sem tumor. No estágio de 288 horas, o desenvolvimento foi interrompido. Os vasos foram quantificados e caracterizados morfologicamente e os embriões fotografados, fixados e processados. Os resultados do metabolismo energético indicaram que não houve diferença significativa entre grupos controle e tratado normal, contudo os animais com tumores induzidos com EHA e ME e os enriquecidos com ω-3 apresentaram resposta significativa quando comparados ao controle (p≤ 0,05). Foi observado que as propriedades físicas e a integridade estrutural da casca dos animais controle tumorado apresentaram anomalias de formação: casca rugosa, áspera e mole. Os parâmetros morfométricos não apresentaram significância entre os grupos (p≤ 0,05). Em relação a vasculogênese e angiogênese houve uma redução significativa entre grupos normais e tumorados. Os vasos apresentaram uma discreta redução do calibre quando comparado ao grupo controle (p≤ 0,05). O aspecto microscópico da membrana amniótica dos organismos tratados com o EHA e ME, dos grupos normal e tumorado mantiveram a morfologia preservada ao longo do tratamento. O tecido cardiovascular de embriões controle com tumor apresentou pontos hemorrágicos e uma congestão no lúmen do vaso, talvez em função da presença da malignidade das células. A microscopia da medula encefálica se manteve preservada. Nos embriões controle tumorados ela se apresentou pouco conservada, com vasos congestos na região do manto, possivelmente por interferência da ação do tumor no tecido analisado. Estes biocompostos apresentaram ainda efeito antivasculogênico e antiangiogênico, sem comprometer a anatomia macroscópica e microscópica dos organismos tratados durante o desenvolvimento embrionário. Diante do presente exposto, conclui-se que os compostos presentes nos extratos de Caulerpa taxifolia pode ser considerada um potencial agente antitumoral de origem marinha, que talvez atue nos mecanismos intrínsecos relacionados com a matriz extracelular, estabilizando e remodelando a estrutura vascular e a capacidade de indução da neovasculogênese. / The neovascularization is an important mechanism in tumor development, is responsible for nutritional support to cancer cells proliferating and establishing favorable conditions for metastatic spread. Among many mechanisms, stimulation of angiogenesis leads to increased secretion of vaso-inducing factors and decreased inhibitors. This study evaluated the interference of hydroalcoholic extracts (EHA - 50 mg / mL) and methanol (MS - 50 ug / ml) of Caulerpa taxifolia in energy metabolism and angiogenesis of Gallus gallus domesticus embryos L. Red breed eggs Rhode Island Red four batches were incubated at 37 C with automatic turning, for study of energy metabolism and angiogenesis. The groups were divided in control groups treated EHA (50 g / mL) and ME (50 ug / ml) and control enriched with ω-3, with and without tumor. In stage age of 288 hours, the development was stopped. The vessels were quantified and characterized morphologically and photographed embryos, fixed and processed. The results of energy metabolism showed no significant difference between control group and the normal treaty, yet tumorados animals treated with EHA and ME and enriched with ω-3 showed significant response compared to control (p ≤ 0.05). It was observed that the physical properties and structural integrity of the shell tumorado control animals showed formation of defects: rough skin, rough and soft. The morphometric parameters showed no significant difference between the groups (p ≤ 0.05). For vasculogenesis and angiogenesis was significantly reduced between normal groups and tumorados. The vessel had a slight reduction in size compared with the control group (p ≤ 0.05). The microscopic aspect of amniotic membrane bodies treated with EHA and ME, the tumorado groups remained normal and preserved morphology throughout the treatment. The cardiovascular tissue with tumor control embryos showed bleeding points and congestion in the vessel lumen, perhaps due to the presence of malignant cells. The brain marrow microscopy remained preserved. However, in tumorados control embryos she had just saved with congested vessels in the mantle region, possibly by tumor action of interference in the analyzed tissue. These biocompounds still had antivasculogênico and antiangiogenic effect without compromising the macroscopic and microscopic anatomy of organisms treated during embryonic development. Given the above this, it is concluded that the compounds present solution in taxifolia Caulerpa extracts can be considered as a potential antitumor agent of marine origin, which may act on the intrinsic mechanisms related to the extracellular matrix, stabilizing and reshaping the vascular structure and inducibility of neovasculogenesis.

Predicting Risks Of Invasion Of Caulerpa Species In Florida

Invasions of exotic species are one of the primary causes of biodiversity loss on our planet (National Research Council 1995). In the marine environment, all habitat types including estuaries, coral reefs, mud flats, and rocky intertidal shorelines have been impacted (e.g. Bertness et al. 2001). Recently, the topic of invasive species has caught the public's attention. In particular, there is worldwide concern about the aquarium strain of the green alga Caulerpa taxifolia (Vahl) C. Agardh that was introduced to the Mediterranean Sea in 1984 from the Monaco Oceanographic Museum. Since that time, it has flourished in thousands of hectares of near-shore waters. More recently, C. taxifolia has invaded southern Californian and Australian waters. Since the waters of Florida are similar to the waters of the Mediterranean Sea and other invasive sites my study will focus on determining potential invasion locations in Florida. I will look at the present distribution of C. taxifolia - native strain in Florida as well as the distribution of the whole genus around the state. During this study, I address three questions: 1) What is the current distribution of Caulerpa spp. in Florida? 2) Can I predict the location of potential Caulerpa spp. invasions using a set of environmental parameters and correlate them to the occurrence of the algae with the support of Geographic Information System (GIS) maps? 3) Using the results of part two, is there an ecological preferred environment for one or all Caulerpa spp. in Florida? To answer these questions, I surveyed 24 areas in each of 6 zones chosen in a stratified manner along the Floridian coastline to evaluate the association of potential indicators Caulerpa. Latitude, presence or absence of seagrass beds, human population density, and proximity to marinas were chosen as the 4 parameters expected to correlate to Caulerpa occurrences. A logistic regression model assessing the association of Caulerpa occurrence with measured variables has been developed to predict current and future probabilities of Caulerpa spp. presence throughout the state. Fourteen different species of Caulerpa spp. were found in 26 of the 132 sites visited. There was a positive correlation between Caulerpa spp. and seagrass beds presence and proximity to marinas. There was a negative correlation with latitude and human population density. C. taxifolia – aquarium strain wasn't found. Percent correct for our model was of 61.5% for presence and 98.1% for absence. This prediction model will allow us to focus on particular areas for future surveys.
