Estudo dos mecanismos envolvidos na migração celular induzida pelo hidróxido de cálcio para bolha de ar subcutânea em camundongos /

Resumo: O objetivo deste trabalho foi investigar a migração celular induzida pelo hidróxido de cálcio para bolha de ar subcutânea em camundongos. Observou-se que a migração de neutrófilos ocorreu de maneira dose-dependente, alcançando o pico de migração 96 horas após a injeção do estímulo. Esta migração foi inibida com o pré-tratamento dos animais com indometacina (5mg/Kg), meloxican (5mg/Kg) e dexametasona (1mg/Kg), diferentemente, o MK886 (1mg/Kg) foi inefetivo neste processo. Constatou-se também que os animais estimulados com hidróxido de cálcio apresentaram um aumento significativo na liberação de citocinas e quimiocinas (KC, MIP-2, IL1ß e TNFa). O pré-tratamento com meloxican ou aminoguanidina inibiu a liberação de KC, MIP-2, IL1ß, mas não de TNFa. O pré-tratamento com Tioglicolato a 3% aumentou cerca de 167% a população de macrófagos na bolha de ar subcutânea, entretanto, não alterou a migração de neutrófilos. Pôde-se concluir que a migração de neutrófilos para bolha de ar subcutânea induzida pelo hidróxido de cálcio, em camundongos, foi dependente de KC, MIP-2, IL1ß e TNFa, sendo a liberação destes mediadores independente de mastócitos e de macrófagos. / Abstract: The aim of this study was to investigate the cellular migration induced by calcium hydroxide to air-pouch in mice. It was observed that the migration developed dependently to the doses and achieved the peak 96 hours after the stimulation. Neutrophil migration was inhibited with the pre-treatment with indometacin (5mg/Kg), meloxican (5mg/Kg) and dexamethasone (1mg/Kg). Differently, MK886 (1mg/Kg) was ineffective in this process. It was seen that the animals stimulated with calcium hydroxide showed an increase of cytokines and chymiokines (KC, MIP-2, IL1ß e TNFa) release. Pre-treatment with meloxican or aminoguanidine inhibited KC, MIP-2, IL1ß release, but not TNFa. Release of TNFa was not dependent of prostaglandines and nitric oxide because it was not inhibited by aminoguanidine and meloxican. Pretreatment with Tioglocolate 3% increased 167% the macrophage population in the air-pouch. However, it did not change neutrophil migration.
