Modified chitosan nano-substrates for mycobacterial capture

Thesis (MSc)--Stellenbosch University, 2016. / ENGLISH ABSTRACT: Tuberculosis (TB) is one of the world’s deadliest diseases, with one third of the population being infected by it. The diagnosis of active tuberculosis entails finding and identifying Mycobacterium tuberculosis (Mtb), the causative pathogen in a specimen of bodily fluid from the patient. Multiple samples will improve the diagnostic yield and specimen volumes should therefore be as large as possible, which is often challenging for patients and especially younger children. Alternatively, a smaller volume could be required if there was a manner in which to concentrate the bacteria within a specimen, through use of a substrate which has an affinity for the pathogenic species. Polymers having intrinsic cellular activity are of interest as such substrates, one such being the natural polysaccharide, chitosan. In this thesis, a variety of modified chitosan derivatives were prepared as potential Mtb-capturing substrates. This was achieved by modifying chitosan with a variety of moieties, selected based on possible interactions with the Mtb cell wall, to render various quaternary ammonium salts of the polymer chitosan. The quaternized chitosan derivatives were then used to synthesize nano-substrates having an affinity for Mtb. Polymer coated superparamagnetic magnetite nanoparticles (SPMNs) were synthesized via an in situ co-precipitation technique, in which modified chitosan is able to chelate with the metal core. Polymer nanofibers were also electrospun via the electrospinning technique. The prepared derivative, N-trimethylammonium chitosan chloride (TMC), was electrospun into nanofibers by blending with suitable non-ionogenic polymers, namely polyvinyl alcohol (PVA), polyethylene oxide (PEO), polyvinyl pyrrolidone (PVP) and polyacrylamide (PAM), required to facilitate nanofiber formation. Affinity studies were conducted between the modified chitosan nano-substrates and the bacillus Calmette-Guérin (BCG) strain of Mycobacterium bovis, the attenuated Mtb-mimic bacteria, for evaluation as mycobacterium capturing substrates. The successful capture of BCG onto the surfaces of the various modified chitosan nanofibers and modified chitosan coated superparamagnetic nanoparticles was confirmed by fluorescence microscopy (FM), light microscopy (LM), transmission electron microscopy (TEM) and field emission scanning electron microscopy (FE-SEM). Analysis of the FM, TEM and FE-SEM images indicated that the chitosan coated nanoparticles functionalized with a C12 aliphatic quaternary ammonium moiety (CS-qC12), captured the most BCG through a combination of ionic and hydrophobic interaction. TMC blended with PVA, to produce nanofibers crosslinked with genipin, were found to have the strongest interaction with BCG of the nanofibrous mats tested. These findings were corroborated by water contact angle measurements, which established that PVA was the least hydrophilic of the non-ionogenic polymers and had hydrogen bond donating groups only, factors influencing the cellular adhesive properties of affinity substrates. / AFRIKAANSE OPSOMMING: Tuberkulose (TB) is een van die wêreld se mees dodelikste siektes, met ‘n derde van die bevolking wat geïnfekteer is daarmee. Ten einde aktiewe TB te diagnoseer moet Mycobacterium tuberculosis (Mtb), die voorsakende patogeen in ʼn monster van die pasiënt se liggaamlike vloeistof, gevind en ïdentifiseer word. Veelvuldige monsters sal die diagnotiese opbrengs verhoog en monster volumes moet dus so groot as moontlik wees wat dikwels ʼn uitdaging vir pasiënte en veral jonger kinders kan bied. Alternatiewelik kan ʼn kleiner monster van die pasiënt vereis word indien daar ʼn manier was om die bakterieë in ʼn monster te konsentreer deur die gebruik van ʼn substraat wat ʼn affiniteit toon vir die patogeniese spesie. Polimere met ʼn intrinsieke sellulêre aktiwiteit, wek belangstelling as sodanige substraat, een synde die natuurlike polisakkaried, chitosan. In hierdie tesis is ʼn verskeidenheid gemodifiseerde chitosan afgeleides voorberei as potensiële Mtb-vaslegging substrate. Dit is gedoen deur chitosan te modifiseer met ʼn verskeidenheid funksionele groepe, gekies op grond van moontlike interaksies met die Mtb selwand, ten einde ʼn verskeidenheid kwaternêre ammonium soute van die chitosan polimeer te bekom. Die kwaternêre chitosan afgeleides is gevolglik gebruik om nano-substrate te sintetiseer wat ʼn affiniteit toon vir Mtb. Polimeer bedekte superparamagnetiese magnetiet nanopartikels (SPMNs) is gesintetiseer via ʼn in situ mede-neerslag metode, waarvolgens die gemodifiseerde chitosan polimere in staat is om met die metaal kern te chelaat. Polimeer nanovesels is ook geëlektrospin deur die elektrospin tegniek te gebruik. Die voorbereide afgeleide N-trimetielammonium chitosan chloried (TMC) is tot nanovesels geëlektrospin deur vermenging met geskikte nie-ionogeniese polimere, naamlik poliviniel-alkohol (PVA), polietilene-oksied (PEO), poliviniel-pirrolidoon (PVP) en poliakrielamied (PAM), wat vereis word ten einde nanovesels te produseer. Affiniteit studies is uitgevoer tussen die gemodifiseerde chitosan nano-substrate en die bacillus Calmette-Guérin (BCG) stam van Mycobacterium bovis, die verswakte Mtb-mimiek bakterieë vir evaluering as mycobakterium-vaslegging substrate. Die suksesvolle vasvang van BCG op die oppervlaktes van die verskillende gemodifiseerde chitosan nanovesels en gemodifiseerde chitosan bedekte SPMNs is bevestig deur fluoressensie mikroskopie (FM), lig mikroskopie (LM), transmissie elektron mikroskopie (TEM) en veld-emissie-skandering elektron mikroskopie (FE-SEM). Analise van die FM, TEM en FE-SEM beelde het getoon dat die chitosan bedekte nanopartikels met byvoeging van ʼn C12 alifatiese kwaternêre ammonium groep, die meeste BCG vasgevang het deur ʼn kombinasie van ioniese en hidrofobiese interaksie. TMC vermeng met PVA om nanovesels te vorm, gekruisbind met genipin, is gevind om die sterkste interaksie met BCG te toon. Hierdie bevindings is bevestig deur water-kontak-hoek-metings, wat getoon het dat PVA die minste hidrofilies van die nie-ionogeniese polimere was en slegs waterstof-binding skenkings groepe het, alles faktore wat die sellulêre bindingskwaliteite van affiniteit-substrate sal beïnvloed.

Potencialidades da succinil quitosana como emulsificante e substituto de fração lipídica em massa de bolo / Potentialities of succinyl chitosan as emulsifier and lipid fraction substitute on cake batter

A necessidade da criação de produtos inovadores com características funcionais está relacionada com a demanda de consumidores que priorizam o bem-estar vinculado a uma alimentação saudável e equilibrado. Nesse sentido, o objetivo deste estudo foi avaliar os efeitos da adição de succinil quitosana (SQ) como um substituto da fração lipídica e agente emulsificante sobre a qualidade de bolos. Para tanto, no capítulo 1 foi realizado um levantamento conciso da literatura para a compreensão dos temas abordados. No capítulo 2, o estudo da SQ se baseou na sua obtenção e caracterização para aplicaçãoem bolos. A análise de infravermelho e difração de raio X validaram as reações de desacetilação e succinilação, sendo que os estudos térmicos demonstraram estabilidade térmica (acima de 300 °C) viabilizando a utilização da SQ em produtos de panificação. A aplicação da suspensão de SQ (2,0 g/100 g) em massas de bolos com níveis reduzidos de gordura (0 % (controle), 25 %, 50 %, 75 % e 100 % (ausência total de gordura)) com ajustes na quantidade de água, para adaptação das consistências das massas, resultou em massas de bolos com parâmetros de viscosidade semelhantes. A redução de gordura até um nível de 50 % e presença de SQ contribuiu para o aumento da área 2D (de 13, 83 para 19,05 mm2) e razão largura/altura, além da umidade e atividade de água. A SQ influenciou significativamente (p < 0,05) na vida de prateleira, visto que a taxa de endurecimento foi de 27,8 (N/dia) para os bolos controle, e em bolos com nível de 25 % apresentou o valor de 26,1 (N/dia) e 49,2 (N/dia) para àqueles com presença total de SQ. Os teores de lipídeos diminuíram significativamente (p < 0,05), embora o conteúdo total de minerais e proteínas tenham apresentado ligeiras variações. Em relação à análise sensorial, foram atribuídas aos bolos com redução de até 50 % de gordura notas entre 6 e 7, e àqueles com 75 % de redução, nota inferior para os atributos de textura, sabor e aspecto geral. Dos provadores, 35 % \"provavelmente comprariam\" os bolos com 50 % de gordura. Nesse sentido, a SQ pode ser considerada um potencial substituto de gordura em bolos, podendo-se reduzir à metade a quantidade deste ingrediente em sua formulação original, agregando qualidades tecnológicas, sensoriais e nutricionais a esses produtos. / The need to create innovative products with functional characteristics is related to the demand of consumers which prioritize the well-being combined to a healthy and balanced diet. In this sense, the objective of this study was to evaluate the effects of the addition of succinyl chitosan (SQ) as an emulsifying agent and lipid fraction substitute on the qualities of cakes. Therefore, in chapter 1 a concise survey of the literature was carried out to understand the themes covered. In chapter 2, the SQ study was based on its obtaining and characterization for application in cakes. Infrared analysis and X-ray diffraction validated the deacetylation and succinylation reactions, and the thermal studies demonstrated thermal stability (above 300 °C), allowing the use of SQ in bakery products. The application of SQ suspension (2.0 g / 100 g) with reduced levels of fat (0 % (control), 25 %, 50 %, 75 % and 100 %) adjusting the amount of water, to adapt the batter\'s consistencies, resulted in cakes with similar viscosity parameters. The reduction of fat up to 50 % and presence of SQ contributed to the increase of the 2D area (from 13,83 to 19,05 mm2) and width / height ratio, as well as moisture and water activity. SQ significantly influenced (p <0.05) shelf life, since the hardening rate was 27,8 (N / day) for control cakes, and in cakes with a 25% level the value was 26,1 (N / day) and 49.2 (N / day) for those with total presence of SQ. The lipid contents decreased significantly (p <0.05), although the total content of minerals and proteins showed slight variations. Regarding sensory analysis, scores between 6 and 7 were attributed to cakes with a fat reduction of up to 50%, and those with a 75% reduction were attributed a lower score, according to the attributes of texture, taste and overall appearance. From consumers, 35% \"I would probably purchase\" the cakes with 50% fat. O presente trabalho estudou duas vias reacionais para a preparação de derivados cationizados de quitosana (QCat), sendo que a N-metilação extensiva via reação de quitosana com iodometano (CH3I) resultou em N,N,N-trimetilquitosana (TMQ) enquanto que a reação de quitosana com cloreto de glicidiltrimetilamônio (CGTMA) produziu N-(2-hidroxi)-propil-3-trimetilamônio quitosana (HPTAQ). Planejamento fatorial completo 23 foi aplicado para analisar os efeitos das condições reacionais sobre o rendimento das reações (R) e as características de TMQ e HPTAQ. As características estruturais da quitosana de partida e seus derivados foram analisadas por espectroscopias na região de infravermelho (FTIR) e de ressonância magnética nuclear de hidrogênio (RMN-1H), sendo que esta última também foi utilizada para determinar os graus médios de acetilação (GA), de quaternização (GQ) e de substituição (GS) de quitosana, TMQ e HPTAQ, respectivamente. Viscosimetria capilar foi utilizada para determinar a viscosidade intrínseca ([&eta;]) de quitosana, TMQ e HPTAQ, permitindo estimar a ocorrência de despolimerização e sua relação com as condições reacionais. Todas as amostras de TMQ foram solúveis em água e o valor de GQ variou no intervalo 21,0 - 67,0%; a [&eta;] variou entre 13,7 mL/g - 213,0 mL/g e o rendimento da reação atingiu até 82,0%, sendo que entre as variáveis analisadas a concentração de hidroxido de sódio e excesso de CH3I são as mais relevantes. Quanto às amostras de HPTAQ, apenas aquelas com GS &gt; 12,0% foram solúveis em água,GS ficou no intervalo 1,0 % - 45,5%; [&eta;] variou entre 283,0 mL/g - 446,0 mL/g e o rendimento não foi superior a 33,0%, e entre as variáveis analisadas, temperatura e excesso de CGTMA foram as mais importantes na produção de HPTAQ. As amostras TMQ2 GQ = 46,0%; [&eta;] = 290,0 mL/g) e HPTAQ6 GS = 33,0%; [&eta;] = 293,0 mL/g) foram empregadas em estudo visando a formação de complexos surfactante/polieletrólito (CSP) com decanossulfonato de sódio (DS). As propriedades condutimétricas e tensiométicas dos TMQ2, HPTAQ6 e CSP\'s foram estudadas e mostraram que todos apresentam atividade interfacial (&gamma; <= 52,0 mN.m-1), além de elevado valor de módulo de elasticidade (E >= 31,0 mN.m-1), indicando que as espécies analisadas geram filmes interfaciais com boas propriedade mecânicas e por isso podem ser aplicados como agentes estabilizantes de emulsões. As emulsões do tipo óleo/água preparadas pela incorporação de QCat (TMQ ou HPTAQ) ou de CSP (TMQ/DS ou HPTAQ/DS) foram estáveis por mais de 4 meses, conforme avaliado por medidas de fluorescência e de turbidez. / In this work were studied two different methods for preparing cationized derivatives of chitosan (ChCat), a consisted extensive N-methylation by chitosan reaction with iodomethane (CH3I) resulted in N,N,N-trimethylchitosan (TMCh), and other a reaction of chitosan with chloride glycidyltrimethylammonium (GTMAC) resulted in N-(2-hydroxy)-propyl-3-trimethylammonium chitosan (HPTACh). A complete factorial design (23) was used to analyze the influence of reaction conditions on the yield of the reaction (R) as well as the characteristics of TMCh and HPTACh produced. The structure of chitosan pattern and its derivatives were characterized by infrared spectroscopy (IR) and nuclear magnetic resonance (1H-NMR), considering that the latter was also used to determine the average degree of acetylation (DA), quaternization (DQ) and substitution (DS) of chitosan, TMCh and HPTACh, respectively. Capillary viscometry was used to determine the intrinsic viscosity ([&eta;]) of chitosan, TMCh and HPTACh, allowing to estimate the occurrence of depolymerization and its relationship with the reaction conditions. All samples of TMCh were soluble in water and it was found that DA ranged as 21.0% - 67.0%; the [&eta;] ranged as 13.7 mL/g - 213.0 mL/g and the yield reaction was up to 82.0%, among the variables studied, the concentration of sodium hydroxide and excess CH3I were the most relevant. The HPTACh samples were soluble in water with DS &gt; 12.0%, DS ranged as 1.0% - 45.5%; the [&eta;] ranged as 283.0 mL/g - 446.0 mL/g and the yield was lower than 33.0%, among the variables studied temperature and excess CGTMA were the most important in the production of HPTACh. Among the reaction conditions studied for the synthesis of TMCh the best condition was 2, while for HPTACh the best condition was 6. The samples TMCh2 DQ = 46.0%, [&eta;] = 290.0 ml/g) and HPTACh6 DS = 33.0%, [&eta;] = 293.0 mL/g) were used in a study aiming at the formation of surfactant/polyelectrolyte complex (SPEC) with sodium 1-decanessulfonate (SD). The conductivity and surface tension properties of TMCh2, HPTACh6 and SPEC\'s have been studied and show that all have interfacial activity (&gamma; <= 52.0 mN.m-1), and high value of elasticity modulus (E >= 31.0 mN.m-1), indicating that all species generate interfacial film with good mechanical property and thus can be applied as a stabilizing agent in emulsions emulsions oil/water that were prepared by incorporating ChCat synthesized and SPEC\'s in their formulations, they were stable for more than 4 months, as evaluated by fluorescence and turbidity measurements.

As emulsões do tipo óleo/água preparadas pela incorporação de QCat (TMQ ou HPTAQ) ou de CSP (TMQ/DS ou HPTAQ/DS) foram estáveis por mais de 4 meses, conforme avaliado por medidas de fluorescência e de turbidez. / In this work were studied two different methods for preparing cationized derivatives of chitosan (ChCat), a consisted extensive N-methylation by chitosan reaction with iodomethane (CH3I) resulted in N,N,N-trimethylchitosan (TMCh), and other a reaction of chitosan with chloride glycidyltrimethylammonium (GTMAC) resulted in N-(2-hydroxy)-propyl-3-trimethylammonium chitosan (HPTACh). A complete factorial design (23) was used to analyze the influence of reaction conditions on the yield of the reaction (R) as well as the characteristics of TMCh and HPTACh produced. The structure of chitosan pattern and its derivatives were characterized by infrared spectroscopy (IR) and nuclear magnetic resonance (1H-NMR), considering that the latter was also used to determine the average degree of acetylation (DA), quaternization (DQ) and substitution (DS) of chitosan, TMCh and HPTACh, respectively. Capillary viscometry was used to determine the intrinsic viscosity ([&eta;]) of chitosan, TMCh and HPTACh, allowing to estimate the occurrence of depolymerization and its relationship with the reaction conditions. All samples of TMCh were soluble in water and it was found that DA ranged as 21.0% - 67.0%; the [&eta;] ranged as 13.7 mL/g - 213.0 mL/g and the yield reaction was up to 82.0%, among the variables studied, the concentration of sodium hydroxide and excess CH3I were the most relevant. The HPTACh samples were soluble in water with DS &gt; 12.0%, DS ranged as 1.0% - 45.5%; the [&eta;] ranged as 283.0 mL/g - 446.0 mL/g and the yield was lower than 33.0%, among the variables studied temperature and excess CGTMA were the most important in the production of HPTACh. Among the reaction conditions studied for the synthesis of TMCh the best condition was 2, while for HPTACh the best condition was 6. The samples TMCh2 DQ = 46.0%, [&eta;] = 290.0 ml/g) and HPTACh6 DS = 33.0%, [&eta;] = 293.0 mL/g) were used in a study aiming at the formation of surfactant/polyelectrolyte complex (SPEC) with sodium 1-decanessulfonate (SD). The conductivity and surface tension properties of TMCh2, HPTACh6 and SPEC\'s have been studied and show that all have interfacial activity (&gamma; <= 52.0 mN.m-1), and high value of elasticity modulus (E >= 31.0 mN.m-1), indicating that all species generate interfacial film with good mechanical property and thus can be applied as a stabilizing agent in emulsions emulsions oil/water that were prepared by incorporating ChCat synthesized and SPEC\'s in their formulations, they were stable for more than 4 months, as evaluated by fluorescence and turbidity measurements.

O presente trabalho apresenta a síntese e a caracterização de derivados de quitosana, bem como o preparo e caracterização de filmes de nanocompósitos à base de quitosana comercial (ou seus derivados), argila (MMT) e nanopartículas de prata (NPs-Ag), obtidas via Fotoquímica. Para tanto, foram preparados, a partir da quitosana comercial (QC), os derivados: quitosana desacetilada (Q30des), quitosana purificada (QP), quitosanas parcialmente despolimerizas (QD30, QD21 e QD5), quitosanas hidrofílicas (QD21-40DEAE e QD5-49DEAE) e quitosanas anfifílicas (QD21-40DEAE-6DD, QD21-40DEAE-18DD, QD5-49DEAE-6DD e QD5-49DEAE-17DD). O grau médio de desacetilação das QC, QP e Q30des e de substituição por grupos DEAE e dodecila foram determinados por Espectroscopia de Ressonância Magnética Nuclear de Hidrogênio (RMN de 1H). Ademais, os biopolímeros foram caracterizados por Espectroscopia no Infravermelho (FTIR-ATR), Viscosimetria, Análise Termogravimétrica e Microscopia Eletrônica de Varredura (MEV). Em seguida, foi estudada a síntese de nanopartículas de prata, sob radiação UV, em filmes de nanocompósitos de quitosana comercial ou seus derivados e argila. Em um primeiro momento, estudou-se a formação das NPs-Ag em filmes de QC com diferentes formulações e posteriormente em filmes de derivados de quitosana contendo 10% de MMT (m/m). A técnica de Difração de Raios-X (DRX) foi utilizada para a determinação do espaçamento interlamelar da argila montmorilonita pura e nos compósitos preparados. A síntese das NPs-Ag foi acompanhada por Espectrofotometria de Absorção Molecular no UV-vis, e monitorada após um ano de sua formação, sendo suas características morfológicas, bem como a dispersão da argila nos nanocompósitos examinados por Microscopia Eletrônica de Transmissão (MET). At first, the Ag-NPs formation was studied on QC films with different formulations and secondarily, on films of chitosan derivatives containing 10 wt % of MMT. The X-Ray Diffraction (XRD) was used to determine the interlamellar spacing of pure montmorillonite clay and in the nanocomposites prepared. The synthesis of the NP-Ag was accompanied by UV-vis Spectroscopy. Its morphological characteristics, as well as the clay dispersion in the nanocomposites were examined by Electron Transmission Electron Microscopy (TEM). Finally, the antimicrobial activities of materials were investigated by the disk diffusion method against the bacteria Escherichia coli e Bacillus subtilis.
