Citologia de lavado vesical preparado por citocentrifugação : padronização do método para diagnóstico de doenças vesicais em cães /

Orientador: Marileda Boanfim Carvalho / Banca: Alessandre Hataka / Banca: Marcia Mery Kogika / Banca: Fábio Nelson Gava / Banca: Andrigo Barboza De Nardi / Resumo: O objetivo do presente estudo foi padronizar a metodologia de citologia de lavado vesical preparado pelo método de citocentrifugação, avaliar o padrão das células epiteliais de transição em cães e aplicabilidade clínica da citopatologia esfoliativa como método complementar de diagnóstico de doenças vesicais em cães. O estudo foi realizado em duas fases. A primeira incluiu 34 pacientes sem anormalidade ou doença vesical (cães adultos, machos e fêmeas), para padronização da metodologia e caracterização da celularidade normal. Na segunda fase foram avaliados 95 pacientes (cães adultos, machos e fêmeas), com ou sem sinais do trato urinário inferior. Em ambas as fases os cães foram avaliados por meio de exames de rotina, com destaque para sedimentoscopia de urina, urocultura e exame ultrassonográfico da bexiga, além da citologia de lavado vesical preparada pelo método de citocentrifugação (colorações de Papanicolaou, Giemsa, Rosenfeld modificado e Gram - somente na segunda fase). Os resultados obtidos indicaram que a técnica empregada no estudo para recuperação celular realizada com barbotagens de três ciclos utilizando 1mL/kg de solução salina, se mostrou eficaz para avaliação de células uroteliais de cães sadios. Com o segundo estudo, que incluiu pacientes com doença vesical, concluiu-se que o conjunto de avaliações para o diagnóstico de doença vesical empregado são complementares e as contribuições de cada exame variam de acordo com o tipo de enfermidade a ser diagnosticada. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The objective of this study was to standardize the methodology for cytology bladder washing prepared by cytospin method to evaluate the pattern of epithelial transition cells in dogs and the clinical applicability of exfoliative cytology as a complementary method for of bladder diseases diagnosis in dogs. The study was conducted in two phases. The first one included 34 patients without bladder abnormality or disease (adult dogs, male and female), for standardization of the methodology and characterization of the typical cellularity. In the second phase it was evaluated a group of 95 patients (adult dogs, male and female), with or without lower urinary tract signs. In both phases the dogs were evaluated by routine tests, especially sediment of urine, urine culture and ultrasound bladder examination, as well as cytology of bladder washing prepared by cytospin (stains of Papanicolaou, Giemsa, Rosenfeld modified and Gram - only in the second phase). The results indicated that the technique used in the study for cell recovery, performed with three barbotage cycles using 1 mL/kg of saline, was effective for urothelial cells evaluation from healthy dogs. In the second study, which included patients with bladder disease, it is concluded that the employed set of evaluations for bladder disease diagnosis are complementary and the contributions of each test vary with the type of disease to be diagnosed. Foram realizados exames ultrassonográficos da região pélvica, a fim de comparar as dimensões uterinas com o peso, idade (infantil, juvenil, subadulta, adulta jovem e adulta) e número de partos (nulípara, primípara e multípara) e comparar as dimensões ovarianas com o peso e a idade. O volume uterino (VU) foi diretamente proporcional ao número de partos, fator mais importante no crescimento uterino das fêmeas adultas (p < 0,05). O peso e a idade demonstraram uma correlação positiva com o VU (r = 0.5354, r = 0.6489, p < 0.01), respectivamente. O volume dos ovários cresceu proporcionalmente a idade das fêmeas (p < 0,05). We performed pelvic ultrasound exams to compare the uterine measurements with weight, age (infant, juvenile, subadult, young adults, and adults) and the number of live births (nulliparous, primiparous, and multiparous) and to compare the ovarian measurements with weight and age. Diagnosis of airway affections may be reach by using tracheobronchial (TB) wash and pulmonary biopsy as well. The aim of this study was to evaluate the pulmonary and TB microbiota in healthy horses submitted to different stabling. Eighteen healthy adult male quarterhorses were distributed in two groups: Group 1 (nine animals maintained in extensive regimen); and Group 2 (nine animals kept in stall conditions). The washes were carried out using physiologic solution in TB lumen through the intravenous catheter (14G x 30 cm). Pulmonary biopsies were collected using semiautomatic Tru-cut needles (16G x 20 cm). The material collected was submitted to microbiological culture. No trickiness was observed in the animals. At least one species of bacteria grown in wash tracheobronchial culture in the horses studied. In TB washes was observed predominance both of Gram negative and facultative aerobic microorganisms. In 38.5% out of the washes were isolated Gram-positive coccus, predominantly Staphylococcus. Escherichia coli and Enterobacter were isolated in 3% and 11%, respectively. The culture of airway in stabling animals showed a higher number of isolates in both wash and biopsy techniques comparing to the horses kept in extensive regimen.

A punção por capilaridade foi realizada em quatro áreas diferentes do tumor com agulhas de insulina. Os esfregaços citológicos foram corados com May Grünwald Giemsa. Já os fragmentos tumorais colhidos para o exame histopatológico foram fixados imediatamente em formol tamponado a 10% durante 48 horas e, após o processamento, foram corados com hematoxilina - eosina e classificados segundo Cassali et al., 2011. The classic presentation of the disease is characterized by elderly patients not spayed or later neutered with multiple nodules. Definitive diagnosis of lesions is given by histopathological examination. Nowadays, especially in veterinary oncology, the early detection of a neoplastic state can alter the course of disease. In this context, fine-needle cytology has great value because it allows differentiation between inflammatory processes, benign and malignant neoplastic formations quickly, accurately and safely. However, the application mammary tumors in veterinary medicine has several controversies, due to the diverse histology. The present study aimed to compare the cytological and histological bitches and thus evaluate the accuracy of cytology by capillarity with insulin needle in neoplastic lesions of the mammary glands. Therefore, we studied 69 breast lesions obtained from patients treated at the Service of Oncology Veterinary, Unesp Jaboticabal, and Veterinary Institute Dr. Daleck. The capillary puncture was performed in four different areas of the tumor with insulin needles. The cytology smears were stained with May Grünwald Giemsa. Already tumor fragments harvested for histologic examination were fixed immediately in 10% buffered formalin for 48 hours and, after processing, were stained with hematoxylin - eosin and classified according Cassali et al., 2011.
