Isolamento ambiental de Cryptococcus spp em hospital de Porto Alegre-RS

Ocasionalmente ocorre a aerolização destes propágulos que podem causar infecção humana por inalação destas partículas. O objetivo deste estudo foi identificar a ocorrência de espécies do Complexo Cryptococcus em matéria orgânica e ar interno em ambiente hospitalar. Neste projeto, um total de 60 amostras de excretas de pombos na área externa e 216 amostras de ar de ambiente interno de um Hospital de Porto Alegre foi analisado. Na amostragem microbiológica de ar, a levedura não foi encontrada, entretanto 33% das amostras de excretas de pombos apresentaram positividade para Cryptococcus neoformans. Dos 2015 isolados de Cryptococcus presentes nas excretas de pombos, 238 foram selecionados para delinear o perfil molecular e bioquímico. O genótipo destes isolados foi analisado por RFLP com dupla digestão, fundamentado no gene URA5. A determinação do feromônio Mat α e Mat a foi amplificado por PCR direto, e para análise de variedade das espécies foi utilizado o método PCR-Multiplex. A presença da cápsula e melanização também foi observada. Todos os isolados foram sorotipo A (Var. grubii), genótipo VNI e Mat α. / Yeasts from the Cryptococcus genus are present in multiple locales in nature associated to vegetal and organic remains. Occasionally these airborne particles are inhaled by humans, which could lead to infection. In this research, a total of 60 samples from pigeon droppings in the outside area, and 216 samples of indoor air were analyzed. This study aims indentifying the occurrence of the Cryptococcus complex species in organic matter and indoor air in a hospital in the city of Porto Alegre, Rio Grande do Sul state, Brazil. In the microbiological air sampling, the yeast was not recovered; however, 33% of the excreta samples were positive for Cryptococcus. From the 2.015 isolates of Cryptococcus present in pigeon droppings, 238 were selected to outline molecular and biochemical profiles. The genotype of these isolated samples was analyzed through RFLP with double digestion, based on the URA5 gene. The determination of Mating type α e Mating type a pheromones was amplified through direct PCR, while for the species variety analysis the PCR-Multiplex method was used. The capsule and melanization presence was also noted. All isolates were Serotype A (Var. grubii ), VNI genotype and Mating type α.

Studies have shown that proton pump inhibitors (PPI´s) have an inhibitory effect on melanin production, in melanocytes,produced by a phenoloxidase, called tyrosinase. This effect on inhibition of melanogenesis was associated with inhibition of tyrosinase.Since melanocytes and Cryptococcus spp. produce the melanin with a similar oxidation action of phenolic compounds, the objective of this work was to test the effect of PPI´s on the inhibition of melanin production in Cryptococcusneoformans/gattii lato sensu. For this, a new methodology was initially developed for the quantification of melanin, through the gray scale. In this experiment, 12 strains of Cryptococcus spp., 1 strain of Candidaparapsilosis ATCC 22019 (negative control) and 1 strain of Hortaeawerneckii (positive control) were analyzed, resulting in an adequate production and quantification of melanin in testedstrains. Next, PPIs were tested (omeprazole, rabeprazole, lansoprazole, esomeprazole and pantoprazole), classical antifungal agents (fluconazole and amphotericin B) and glyphosate (inhibitor of melanogenesis in Cryptococcus spp.) on the susceptibility of 20 strains of Cryptococcus spp.and 1 strain of C. parapsilosis ATCC 22019, based on the protocol M27-A3 of the Clinical Laboratory Standart Institute (CLSI).The following MIC50 values were observed: for omeprazole 166.7 ± 91.29 μg/ml, esomeprazole 256 ± 115.1 μg/ml, lansoprazole 226 ± 200.1 μg/ml, rabeprazole 142.6 ± 74.6 μg/ml, pantoprazole 428.6 ± 297.3 μg/ml, glyphosate 952.4 ± 150.4 μg/ml, fluconazole 4,757 ± 1,6 μg/ml and amphotericin B 1,464 ± 2.6 μg/ml. Finally, subinhibitory concentrations of PPIs were tested on melanin production in Cryptococcus spp., resulting in an inhibition of melanin for protons pump inhibitors, the best effect being obtained with omeprazole, lansoprazole and pantoprazole. Therefore, PPI´s has an important effect on the inhibition of melanin in Cryptococcus spp., and further studies are needed to understand the mechanisms of inhibition and its potential in reducing virulence, dissemination and infection of the CNS. / Cryptococcus neoformans lato sensu e Cryptococcus gattii lato sensu são leveduras hialinas capazes de produzir melanina, estando associados auma doença chamada criptococose. Para sua instalação e disseminação, vários fatores de virulência têm sido associados, como a produção de melanina, que é produzida por uma fenoloxidase chamada lacase que oxida os compostos fenólicos. Estudos demostraram que os inibidores de bomba de prótons (IBP´s) têm efeitos inibitórios na produção de melanina, em melanócitos, produzidos por uma fenoloxidase chamada tirosinase. Este efeito inibitório da melanogênese foi associado ao bloqueio da tirosinase. Uma vez que os melanócitos e Cryptococcus spp. produzem melanina por uma via semelhante à oxidação de compostos fenólicos, o objetivo deste trabalho foi testar o efeito de IBP´s na inibição de produção de melanina em C.neoformans/C. gattiilatosensu. Para tanto, inicialmente foi desenvolvida uma nova metodologia para quantificação de melanina por meio da escala de cinza. Neste sentido, foram analisadas 12 cepas de Cryptococcus spp., uma cepa de Candidaparapsilosis ATCC 22019 (controle negativo) e uma cepa de Hortaeawerneckii (controle positivo), resultando em uma adequada produção e quantificação de melanina nas cepas testadas. A seguir, foram testados os IBP´s (omeprazol, rabeprazol, lansoprazol, esomeprazol e pantoprazol), os antifúngicos clássicos (fluconazol e anfotericina B) e glifosato (inibidor da melanogênese em Cryptococcus spp.) sobre a sensibilidade de 20 cepas de Cryptococcus spp. e uma cepa de C. parapsilosis ATCC 22019, baseado no documento M27-A3 do Clinical Laboratory Standart Institute (CLSI). Foram observados os seguintes valores de MIC50: para omeprazol 166,7 ± 91,29 µg/mL; esomeprazol 256 ± 115,1 µg/mL; lansoprazol 226 ± 200,1 µg/mL; rabeprazoL 142,6 ± 74,6 µg/mL; pantoprazol 428,6 ± 297,3 µg/mL; glifosato 952,4 ± 150,4 µg/mL; fluconazol 4,757 ± 1,6 µg/mLe; anfotericina B 1,464 ± 2,6 µg/mL. Finalmente, foram testadas concentrações subinibitórias dos IBP´s sobre a produção de melanina em Cryptococcus spp., obtendo como resultado uma inibição da melanina para os inibidores de bomba de prótons, sendo o melhor efeito obtido com omeprazol, lansoprazol e pantoprazol. Portanto, IBP´s têm um efeito importante na inibição de melanina em Cryptococcus spp., sendo necessário estudos mais aprofundados para compreender os mecanismos de inibição e seu possível potencial na diminuição de virulência, disseminação e infecção do SNC.

Identification and characterisation of a Cryptococcus laurentii Abo 510 Phytase

Thesis (MSc)--University of Stellenbosch, 2004. / ENGLISH ABSTRACT: Phosphorus is vital for growth of all life forms and is a fundamental component of nucleic acids, ATP and several other biological compounds. Oilseeds and cereal grains, two major constituents of the diet of animals, contain phytic acid, which is the main storage form of phosphorus in plant cells. Monogastric animals, such as poultry and pigs, are not capable of utilising the bound phosphorus in phytic acid since they do not produce phytase, the essential hydrolysing enzyme. Microbial phytase is therefore added to the animal feed to enhance the availability of phosphorus and thus minimise phosphorus pollution and phosphorus supplementation in diets. For a phytase to be effective in the poultry and swine industry, it needs to be able to release phytic acid phosphorus in the digestive tract, it must be thermostable to resist feed processing and must be inexpensive to produce. One approach for developing an efficient phytase for the animal feed industry is by identifying new phytases from microorganisms, plants and animals. In this study, 11 strains of the genus Cryptococcus were screened for 'phytase activity. Initially, a differential agar plate screening method was employed to determine if any Cryptococcus species were able to express phytase, after which production was confirmed in different liquid media. Cryptococcus laurentii Abo 510 was identified as a strain with significant phytase activity. The C. laurentii Abo 510 strain showed clear zones on the differentialmedia agar plates and the production of phytase at high levels was observed when using wet cells grown in liquid media. The C. laurentii Abo 510 strain produced maximal phytase activity at a relatively high temperature (62°C) and in an acidic pH range (pH 5.0). This phytase also showed a broad substrate specificity that may assist in the release of other phosphate compounds captured in feedstuff. Although the phytase did not require any metal ions for its activity, several metal ions caused inhibition of the phytase activity. The enzyme was stable when exposed to 70°C for up to 180 minutes with only 40% loss in activity. Phosphorus addition to the culture media and enzyme assay solution at concentrations exceeding 500 f.!Minhibited the phytase activity completely. Different carbon sources in the culture media also influenced the phytase activity. The enzyme was determined to be a cell wall-associated phytase with little intracellularactivity. / AFRIKAANSE OPSOMMING: Lewende organismes benodig fosfaat vir groei en oorlewing en fosfaat vorm 'n fundamentele komponent van nukleïensure, ATP en verskeie ander biologiese verbindings. Veevoer bestaan meestal uit twee groot bestanddele, naamlik oliesade en graansoorte wat fitiensuur bevat. Fitiensuur is die vernaamste vorm waarin fosfaat in veevoer gestoor word. Enkelmaagdiere soos pluimvee en varke is nie in staat om die fosfaat van die fitiensuur te benut nie, aangesien hierdie diere nie die geskikte hidrolitiese ensiem, fitase, vir die vrystelling van fosfaat besit nie. 'n Mikrobiese fitase-ensiem word derhalwe by veevoer gevoeg om die fosfaatbeskikbaarheid te verhoog. Sodoende word fosfaatbesoedeling en fosfaataanvullings tot die dieet van diere ook verminder. Vir 'n fitase om effektief in die pluimvee en vark-industrie te wees, moet dit fosfaat vanaf fitiensuur in die spysverteringskanaal vrystel, dit moet behandeling by hoë temperature tydens die veevoervervaardiging oorleef en die ensiem moet goedkoop geproduseer kan word. Een van die benaderings om 'n effektiewe fitase vir die dierevoer-industrie te ontwikkel, is om nuwe fitases in mikroërganismes, plante of diere te identifiseer. Die fitase het ook 'n wye substraatspesifisiteit wat tot die vrystelling van fosfaat vanaf ander komponente in die veevoer mag bydra. Die fitase het geen metaalione vir sy aktiwiteit benodig nie, maar sekere metaalione het die fitase-aktiwiteit onderdruk. Die ensiem was redelik stabiel by 70°C en het na 180 minute blootstelling slegs 'n 40% verlies in aktiwiteit getoon. Die byvoeging van fosfaat in die kultuurmedium en in die ensiem reaksiemengsel teen konsentrasies bo 500 f.lM, het die fitase aktiwiteit heeltemalonderdruk. Verskeie koolstofbronne het ook 'n effek op die optimale fitase-aktiwiteit getoon. Die fitase ensiem is met die selwand geassosieer en het baie min intrasellulêre aktiwiteit getoon.

These species are capable of expressing multiple virulence factors that contribute to the development of resistance to antifungal therapeutic use. Research seeking new effective therapy measures are necessary. Heat shock proteins (Hsp) have been shown to be a promising molecular target for medical treatment of various diseases such as various forms of cancer and atherosclerosis. Studies have demonstrated that the expression of these chaperonins is also required for the maintenance of cellular homeostasis in different fungal pathogens. From this perspective, this study aimed to investigate the effect of blocking the protein Hsp90 front of sensitivity to antifungal agents in vitro planktonic cells and sessile, production of virulence factors in vitro and in vivo virulence in Cryptococcus neoformans species Complex. Therefore, the action of yeast Hsp90 protein was blocked with radicicol drug - an antibiotic capable of inhibiting the ATPase activity of Hsp90 - and then the cultures were evaluated for in vitro susceptibility to antifungal agents for therapeutic use in planktonic cells and associated in biofilms. We also assessed the effect of inactivation of Hsp90 on the production of virulence factors capsule, melanin and proteases. In addition, the in vivo virulence was investigated in a model of experimental infection using the nematode Caenorhabditis elegans. The results revealed that radicicol inhibited the growth of planktonic cells Cryptococcus spp. (n=12) with values of minimum inhibitory concentration (MIC) ranging from 0.5 to 2 μg/mL. We observed synergy between pharmacological and radicicol azoles. Furthermore, blockade of Hsp90 potentiated the effect of in vitro antifungal agents, particularly for the fluconazole and itraconazole, which MIC values were reduced by up to 64 times the face of isolated cells (n=12). Although the blockade of Hsp90 by isolated radicicol was not able to inhibit biofilm formation and biofilm formed of Cryptococcus spp. (n=12) (p>0.05), the inactivation of Hsp90 potentiated the effect of amphotericin B, azoles, especially fluconazole, when tested in combination with radicicol (p<0.05). Moreover, blockade of Hsp90 significantly reduced the volume of capsular Cryptococcus spp. (n=4) (p<0.05). However, no difference was observed in production of melanin by radicicol treated cells (n=4) (p>0.05) as well as the production of protease by strains of Cryptococcus spp. (n=4) (p>0.05). By contrast, inactivation of Hsp90 decreased virulence Cryptococcus spp. in vitro and in vivo in C. elegans (p<0.0001) as well as maximize the effect of fluconazole in vitro and in vivo. Therefore, Hsp90 is an important target for the development of antifungal strategies. However, further studies are needed to better understand the role of Hsp90 in fungal pathogens of the genus Cryptococcus spp. / O Complexo Cryptococcus neoformans/C. gattii compreende os principais agentes da criptococose, uma importante micose de natureza sistêmica e caráter oportunista. Essas espécies são capazes de expressar múltiplos fatores de virulência que contribuem para o surgimento de resistência aos antifúngicos de uso terapêutico. Pesquisas que buscam novas medidas eficazes de terapia são necessárias. Proteínas do choque térmico (Hsp) têm se demonstrado um alvo molecular promissor em área médica para o tratamento de diversas doenças, tais como diferentes tipos de câncer e aterosclerose. Estudos demonstram que a expressão dessas chaperoninas também é necessária para a manutenção da homeostase celular em diferentes patógenos fúngicos. Nessa perspectiva, o presente estudo teve como objetivo investigar o efeito do bloqueio das proteínas Hsp90 frente à sensibilidade a antifúngicos in vitro em células planctônicas e sésseis, produção de fatores de virulência in vitro e virulência in vivo no Complexo Cryptococcus neoformans/C. gattii. Para tanto, a ação das proteínas Hsp90 fúngicas foi bloqueada com o fármaco radicicol – um antibiótico capaz de inibir a atividade ATPásica das Hsp90 – e, em seguida, as culturas foram avaliadas quanto à sensibilidade in vitro a antifúngicos de uso terapêutico em células planctônicas e associadas em biofilmes. Também foi avaliado o efeito da inativação das Hsp90 sobre a produção dos fatores de virulência cápsula, melanina e proteases. Além disso, a virulência in vivo foi investigada em modelo de infecção experimental utilizando o nematódeo Caenorhabditis elegans. Os resultados revelaram que o radicicol inibiu o crescimento das células planctônicas de Cryptococcus spp. (n=12), com valores de concentração inibitória mínima (CIM) que variaram de 0,5 – 2 μg/mL. Observou-se sinergismo farmacológico entre o radicicol e os azólicos. Ademais, o bloqueio das Hsp90 potencializou o efeito dos antifúngicos in vitro, sobretudo para o fluconazol e o itraconazol, os quais tiveram os valores de CIM reduzidos em até 64 vezes frente às células isoladas (n=12). Embora o bloqueio das Hsp90 pelo radicicol isolado não tenha sido capaz de inibir a formação de biofilme e biofilme formado de Cryptococcus spp. (n=12) (p>0,05), a inativação das Hsp90 potencializou o efeito da anfotericina B e derivados azólicos, sobretudo o fluconazol, quando testados em combinação com o radicicol (p<0,05). Além disso, o bloqueio das Hsp90 reduziu significativamente o volume capsular de Cryptococcus spp. (n=4) (p<0,05). Porém, não observou-se diferença na produção de melanina por células tratadas com o radicicol (n=4) (p>0,05) assim como na produção de proteases pelas cepas de Cryptococcus spp. (n=4) (p>0,05). Em contrapartida, a inativação das Hsp90 diminuiu a virulência de Cryptococcus spp. in vitro e in vivo em C. elegans (p<0,0001) além de potencializar o efeito do fluconazol in vitro e in vivo. Portanto, as Hsp90 são um importante alvo para o desenvolvimento de estratégias antifúngicas. Todavia, estudos mais aprofundados são necessários para melhor compreender o papel das Hsp90 nos patógenos fúngicos do gênero Cryptococcus spp.

Caracterização ambiental do complexo de espécies Cryptococcus neoformans e C. gattii no interior do estado de São Paulo, Brasil /

Orientador: Márcia Marinho / Resumo: A criptococose é uma doença sistêmica causada pela inalação de basidiosporos desidratados de espécies patogênicas do gênero Cryptococcus presentes no ambiente. Os complexos espécies mais associados à doença são os complexos de espécies Cryptococcus neoformans e C. gattii. O complexo de espécies C. neoformans está associado a casos de criptococose em pacientes imunossuprimidos, sendo comumente isolados do solo e de fezes de aves. O complexo de espécies de C. gattii, por sua vez, afeta principalmente indivíduos imunocompetentes e geralmente é isolado em espécies de árvores nativas ou exóticas em estado de biodegradação. Este estudo teve como objetivo realizar o mapeamento ambiental de espécies patogênicas do gênero Cryptococcus em amostras ambientais arbóreas em locais públicos da cidade de Birigui, São Paulo. Para a coleta das amostras, foram selecionadas regiões com maior trânsito neste município. Após processamento, foram realizados testes morfo-fisiológicos, bioquímicos, teste de assimilação de carbono e nitrogênio e PCR URA5 e RFLP com dupla digestão enzimática para caracterização do complexo de espécies/espécie de Cryptococcus. Entre fevereiro de 2018 e março de 2019, foram coletadas 100 amostras de diferentes árvores em locais arborizados públicos. A porcentagem de colonização das árvores por C. gattii VGII foi de 5%, por C. neoformans VNIV de 1% e foram encontrados 6% de C. laurentii e 3% de C. albidus. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Cryptococcosis is a systemic disease caused by the inhalation of dehydrated basidiospores of pathogenic species of Cryptococcus genus present in the environment. The two species complex associated with the disease are the Cryptococcus neoformans and C. gattii species complex. C. neoformans species complex are associated with cases of cryptococcosis in immunosuppressed patients, being commonly isolated from the soil and birds feces. C. gattii species complex affects mainly immunocompetent individuals and are often isolated in native or exotic tree species in some state of biodegradation. This study aimed to perform the environmental mapping of pathogenic species of the genus Cryptococcus in tree samples of public places of the city of Birigui, São Paulo. For collection, the samples were collected from regions with greater traffic in this city. After processing, morphophysiological, biochemical, carbon and nitrogen assimilation and URA5 PCR and RFLP with double enzymatic digestion tests were performed to characterize the Cryptococcus species/species complex. Between february 2018 and march 2019 was collected a total of 100 samples from different trees in public arborized locations. The percentage of tree colonization by the C. gattii VGII was 5%, for C. neoformans VNIV was 1% and was found 6% of C. laurentii and 3% of C. albidus species. Genes diferencialmente expressos por Cryptococcus neoformans e Cryptococcus gattii durante a infecção de macrófagos

Cryptococcus neoformans e Cryptococcus gattii são leveduras encapsuladas e agentes etiológicos da criptococose. Estes microrganismos são patogénos intracelulares facultativos os quais interagem com macrófagos do hospedeiro durante o processo de infecção. C. neoformans é capaz de se replicar no interior de células fagocíticas utilizando uma estratégia única que envolve permeabilização do fagolisossoma e produção de polissacarídeos. No presente trabalho, a metodolologia da Análise da Diferença Representacional (RDA) foi aplicada para identificar genes diferencialmente expressos por C. neoformans e C. gattii durante o desenvolvimento intracelular em macrófagos peritoneais de ratos. C. neoformans internalizados por macrófagos expressaram genes relacionados ao tráfego vesicular, transporte de membrana, atividade de chaperona, regulação da meiose, metabolismo de monossacarídeo e resposta ao estresse. Em C. gattii, foram induzidos transcritos relacionados à respiração aeróbica, endocitose, tráfego vesicular, metabolismo de monossacarídeo e nitrogênio, sinalização celular e resposta ao estresse. Nossos resultados revelaram novos genes envolvidos no parasitismo intracelular de C. neoformans e C. gattii.

Proteínas bioluminescentes : biomarcadores para o monitoramento in vivo da infecção por Cryptococcus neoformans e Cryptococcus gattii

A criptococose é uma doença fúngica invasiva causada majoritariamente pelas espécies patogênicas Cryptococcus neoformans e Cryptococcus gattii. Estudos epidemiológicos recentes sugerem que a infecção causada por C. gattii desenvolve mais frequentemente criptococomas pulmonares, enquanto que C. neoformans estabelece principalmente quadros de meningoencefalite. Embora ocorra essa associação, o mecanismo de disseminação dessas espécies não está completamente elucidado, instigando a utilização de diferentes abordagens na caracterização da infecção. O desenvolvimento de microrganismos bioluminescentes tem permitido o monitoramento em tempo real da infecção em modelos animais. No presente trabalho, diferentes estratégias foram utilizadas para a construção de linhagens de C. neoformans e de C. gattii expressando o gene repórter luciferase Renilla, sem sucesso. Paralelamente, em uma segunda abordagem, bactérias bioluminescentes (BL) foram isoladas de amostras de água marinha coletadas nas adjacências da zona estuarina do rio Tramandaí, com o objetivo de selecionar o cassete lux (CDABE) para a utilização como gene repórter. Todos os isolados apresentaram luminescência a 28°C, mas, quando incubados a 37°C, somente o isolado BL6 permaneceu com uma atividade luminescente consideravelmente reduzida. Portanto as bactérias bioluminescentes pertencentes aos gêneros Vibrio, Photobacterium e Enterovibrio isoladas do rio Tramandaí não apresentaram níveis de emissão de luminescência adequados à temperatura fisiológica humana para a utilização do cassete lux como gene repórter para monitoramento de infecções in vivo. / Cryptococcosis is an invasive fungal disease caused mainly by pathogenic species Cryptococcus neoformans and Cryptococcus gattii. Recent epidemiological studies have suggested that infection with C. gattii develops more frequently pulmonary cryptococcomas, while C. neoformans establishes mainly neurological damages. Although exists this association, the mechanism of dissemination of these species is not completely understood, instigating the use of different approaches for the characterization of the infections. Alternatively, the development of bioluminescent organisms has allowed the real-time monitoring of infection in animal models. In this study, different strategies were used to construct strains of C. neoformans and C. gattii expressing Renilla luciferase reporter gene without success. In a second approach, bioluminescent bacteria (BL) were isolated from seawater samples collected in the vicinity of the river Tramandaí estuarine zone, in order to select the lux cassette (CDABE) for use as a reporter gene. All isolates showed luminescence at 28°C, but when incubated at 37°C, only the isolate BL6 remained with a considerably reduced luminescent activity. Thus, bioluminescent bacteria belonging to the genera Vibrio, Photobacterium and Enterovibrio isolated from Tramandaí river estuary zone presented unsuitable luminescence emission levels at human physiological temperature, preventing the use of lux cassette as a reporter gene in experiments to monitoring cryptococcosis infection.
