No. of bitstreams: 0 / Os compostos semicondutores com alto número atômico e energia de banda proibida larga vêm sendo pesquisados como detectores de radiação X e gama, com alta resolução energética, operando à temperatura ambiente. O denominador comum dos materiais semicondutores, que operam à temperatura ambiente, é a dificuldade em crescer cristais com pureza química elevada e boa estequiometria. O desenvolvimento deste tipo de detectores semicondutores de radiação é ainda um desafio tecnológico e tem deparado com muitos fatores limitantes, tais como: material de partida com qualidade compatível para o uso no crescimento de cristal, baixa estabilidade do detector ao longo do tempo, oxidação superficial e outras dificuldades relatadas na literatura, que limitam o seu uso. Neste trabalho, estabeleceu-se a metodologia de transporte físico de vapor (PVT) para a purificação e crescimento do cristal semicondutor de Iodeto de Mercúrio (HgI2). Cristais de HgI2 com orientação cristalina, estequiometria e morfologia da superfície adequadas foram obtidos por essa técnica. Uma redução nítida de impurezas após a purificação pode ser observada e o nível de impureza presente nos cristais não interferiu nas suas estruturas cristalinas. Uma boa morfologia com uniformidade nas camadas da superfície foi encontrada nos cristais, indicando uma boa orientação na estrutura cristalina. Um estudo inédito foi realizado no Laboratório da University of Freiburg, sob a coordenação do Prof. Michael Fiederle, com o intuito de aumentar a estabilidade do detector de HgI2 ao longo do tempo. A aplicação de diferentes tipos de resina polimérica para encapsulamento dos detectores HgI2 foi realizada e estudada, no intuito de proteger o cristal de HgI2 das reações com os gases atmosféricos e isolar eletricamente a superfície dos cristais. Quatro resinas poliméricas foram analisadas, cujas composições são: Resina n 1: 50% - 100% de heptano, 10% - 25% metilcicloexano, <1% de ciclo-hexano; Resina n2: 25% - 50% de etanol, 25% - 50% de acetona, <2,5% de acetato de etilo; Resina n3: 50% - 100% de acetato de metilo, 5% - 10% de n-butilo e Resina 4: 50% - 100% de etil-2- cianoacrilato. A influência dos tipos de resina polimérica utilizada na espectroscopia de desempenho do detector semicondutor HgI2 é, claramente, demonstrada. O melhor resultado foi encontrado para o detector encapsulado com resina n3. The role of pharmaceutical excipients in the solid-state degradation of Gabapentin

Drug instability in solid dosage forms includes chemical or physical processes involving covalent or polymorphic transformations wherein different polymorphs possess crystal structure differences. Gabapentin chemically degrades by intramolecular cyclization to gabapentin-lactam (lactam) in the solid-state. Additionally, gabapentin undergoes polymorphic solid-state transformations. A kinetic model was developed to describe the environmental and excipient effects on chemical and physical instability associated with milling induced stress and subsequent storage under controlled temperature and humidity conditions. Reaction mixtures were generated by co-milling gabapentin Form II with various excipients. The effects of environmental conditions were studied by storing reaction mixtures at 40-60 ºC and 5-50 %RH. The chemical and polymorphic compositions of the reaction mixtures were measured as a function of time using a combination of chromatographic method, 13C ssNMR and XRPD. Degradation models that describe the relationship between polymorphs and degradation product in a series of sequential or parallel steps were devised based on analysis of the resultant concentration time profiles. Model parameters were estimated using non-linear regression and Bayesian methods and evaluated in terms of their quantitative relationship to compositional and conditional variations. In reaction mixtures composed of co-milled gabapentin and excipients, gabapentin was found to exist in three forms: anhydrous polymorph II and III and gabapentin-lactam. A fourth form (II*) was observed based on initial degradation kinetics and was hypothesized to be a crystal-disordered form generated by mechanical stress. The effect of environment moisture was to decrease the net rate of lactam formation by facilitating polymorphic transformation kinetics and crystal annealing. However, excipient blocked the catalytic moisture effect on polymorphic transformations. The key features of our model are first-order physical state transitions of II* and III to II, first-order degradation of II* to lactam and autocatalytic lactamization of II and III. For chemical transitions, no humidity effect was present but the catalytic effects of excipients on the conversion of II and III → lactam were observed. For physical transitions, excipient primarily influenced the physical state transitions of II*and III → II through its ability to interact with humidity and the degree of contact between excipient and substrate.

Local Structural and Optical Characterization of Photonic Crystals by Back Focal Plane Imaging and Spectroscopy

This thesis establishes methods to locally and effciently detect the fluorescence from photonic crystals (PCs) in dependence on wavelength and direction. These are applied to three dimensional (3D) PCs grown by vertical deposition of polystyrene beads. The experiments allow conclusions about the local 3D structure of a sample, about defects in its volume and about spatial structural variations. They thus provide more information than typical spectroscopy measurements that average over large areas and methods that only image the surface structure like scanning electron microscopy. A focused laser is used to excite emitters in the sample only locally. The fluorescence is then collected by a microscope objective. Every point in this objective’s back focal plane (BFP) corresponds to a certain direction. This property is utilized in two ways. When observing a small spectral range of the emission in the BFP, stop bands appear as intensity minima since they hinder the emission into the corresponding directions. Thus, back focal plane imaging (BFPI) allows to visualize stop bands of many directions at the same time. The detected patterns permit to find the in-plane and out-of-plane orientation of the PC lattice and to conclude on the presence of stacking faults. Spatial variations of the structure are observed on a length scale of a few micrometers. The depth of the stop band is reduced at sample positions, where structural changes occur. In back focal plane spectroscopy (BFPS), a slit selects light from certain points in the BFP, which is spectrally dispersed subsequently. This allows to record spectra from many directions simultaneously. From them, a lattice compression along the sample normal of about 4% is found. Small deformations are also observed for other directions. Scattering at defects redistributes the emission. This increases the detected intensity compared to homogeneous media at some stop band edges in a broad spectral range for samples thicker than the scattering mean free path. Thinner samples show a narrow enhancement due to an increase in the fractional density of optical states and thus in emission. BFPI and BFPS are also used to observe the growth of PCs from drying droplets. The experiments show that the beads initially form a non-close packed lattice. This causes stress as the lattice constant decreases, which is released by cracking of the PCs.
