Desenvolvimento e AplicaÃÃo de Teste de Toxicidade Aguda Utilizando como Organismo-Teste Daphnia Magna

Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico / No estado do CearÃ, nÃo se tem conhecimento da aplicaÃÃo de testes de toxicidade, quer com esgoto sanitÃrio, quer com esgoto industrial. Assim, o presente trabalho objetivou implantar o cultivo e o teste de toxicidade aguda com o organismo-teste Daphnia magna no LaboratÃrio de Saneamento (LABOSAN) da Universidade Federal do Cearà (UFC), conforme Norma Brasileira 12713/2004. Com base nas recomendaÃÃes desta norma, conseguiuse cultivar e realizar ensaios de sensibilidade e testes de toxicidade com D. magna em efluentes domÃsticos e industriais. Devido à quantidade insuficiente de neonatos gerados no inÃcio do cultivo, elaborou-se curva de crescimento das algas que foram utilizadas como alimento: Pseudokirchneriella subcaptata (ex- Selenastrum capricornutum), Scenedesmus subspicatus e Spirulina platensis, fundamentais para o estudo sobre a reproduÃÃo de D. magna, quando submetida a trÃs diferentes dietas. Neste estudo foi observado que a quantidade de alimento influenciou diretamente na reproduÃÃo de D. magna. Tanto a P. subcaptata, quanto a S. subspicatus (algas indicadas pela NBR 12713/2004) se mostraram adequadas na alimentaÃÃo de D. magna na concentraÃÃo de 6,5E+06 cÃls./mL.Daphnia . TambÃm comprovou-se que a S. platensis nÃo à tÃo adequada para a alimentaÃÃo de D. magna quanto as indicadas pela Norma Brasileira. A carta controle foi elaborada e o valor de CE 5024h mÃdio obtido foi comparado com valores reportados na literatura, comprovando que a carta poderia ser utilizada e que os organismos do cultivo estiveram dentro do intervalo de seguranÃa no perÃodo dos testes. Os resultados dos testes de toxicidade com o efluente real contendo corantes revelaram que o sistema anaerÃbio, alÃm de reduzir cor e DQO, contribuiu para a detoxificaÃÃo do efluente e que o pÃs-tratamento aerÃbio aumentou a toxicidade. Por fim, os testes de toxicidade com esgoto tÃxtil mostraram-se ser uma ferramenta imprescindÃvel na avaliaÃÃo de subprodutos gerados em condiÃÃes anaerÃbias e aerÃbias, melhorando o entendimento dos processos de tratamento. / In the Cearà state, as far as we know, toxicity tests were never conducted before, either with sewage or industrial wastewater. Thus the present work aimed to implement the cultivation and application of acute toxicity tests with Daphnia magna as test-organism in the Laboratory of Sanitation (LABOSAN) of the Federal University of Cearà (UFC), according to the Brazilian guideline NBR 12713/2004. Based on the recommendations of this guideline, it was possible to cultivate and conduct sensibility experiments and toxicity tests with D. magna with both sewage and industrial wastewaters. Because the amount of neonates in the beginning of cultivation was not enough, a growth curve was developed for each of the algae used to feed D. magna: Pseudokirchneriella subcaptata (ex- Selenastrum capricornutum), Scenedesmus subspicatus and Spirulina platensis, They were tested with three different diets. In this study the amount of food influenced directly D. magna reproduction. Both P. subcaptata and S. subspicatus (algae suggested in the NBR 12713/2004) showed to be a good food source to D. magna when the concentration was 6,5E+06 cels./mL.Daphnia. It was also verified that S. platensis was not good to feed D. magna compared to the other algae tested and reported in the guideline. The sensibility letter was created and the EC 5024h average achieved was comparable to the values reported in literature, which confirm that the letter could be used and the cultivation organisms were in the security interval of the test period. The toxicity test results using real dye-containing wastewater showed that the anaerobic system decrease color and COD and helped to detoxify the effluent. The aerobic post-treatment increased the toxicity. Finally, the ecotoxicity tests with textile wastewater showed to be an important tool to assess sub-products generated on anaerobic and aerobic conditions, therefore improving the process treatment understanding.
