Partindo das compreensÃes de Bourdieu, as aÃÃes dos agentes no seu espaÃo social, indicados pelo habitus, criam uma luta simbÃlica por posiÃÃes de prestÃgio atravÃs do acÃmulo de capital simbÃlico dentro da cena. AtravÃs do resgate histÃrico de suas carreiras, à possÃvel identificar as possÃveis instÃncias de consagraÃÃo e legitimaÃÃo dos agentes. Propus-me a pensar os sujeitos e suas atribuiÃÃes de sentido que estabelecem a partir das vivÃncias, das ideias e do conhecimento necessÃrio para se tornar consagrado, reconhecido, lembrado pelos demais DJs da cidade. Os dados utilizando sÃo entrevistas com trÃs agentes (Fil, Fran Viana e Rodrigo LobbÃo), associadas Ãs notÃcias e entrevistas veiculadas em jornais, sites, revistas e blogs especializados na disseminaÃÃo das atividades da cena local. Como resultado, percebe-se uma lÃgica particular, no qual hà relaÃÃo direta entre o acÃmulo de conhecimento, proporcionado pelas experiÃncias de sua carreira, e as posiÃÃes de deferÃncia aos demais.

The Mechanisms of Protective Function of DJ-1 in Parkinson’s Models of Neuronal Loss: VHL and PON2

Parkinson's disease (PD) is the most common neurodegenerative motor disorder, whose clinical features are rest tremor, bradykinesia, muscular rigidity and postural instability. Although most reported cases are sporadic, a handful of familial cases and their causative genes have been identified. Loss-of-function mutations in DJ-1, one of these genes, are responsible for 1% of familial PD cases. Our laboratory has previously reported that DJ-1- lacking neurons are sensitive to oxidative stress, induced by hydrogen peroxide or the neurotoxin MPTP. To investigate the possible mechanisms through which DJ-1 protects against oxidative stress, we performed a proteomic screen and identified Von Hippel Lindau (VHL) and Paraoxonase2 (PON2) as potential DJ-1 interacting partners. VHL is an E3 ubiquitin ligase which, in normal conditions, poly-ubiquitinates HIF-1 , a subunit of a master hypoxic/oxidative stress transcription factor, whose function is protective in oxidative and hypoxic stresses. In the present study, we provided further evidence of interaction of DJ-1 with VHL. We also demonstrated that HIF-1 protein level, as an indicator of VHL activity, is lower in cells lacking DJ-1, suggesting the inhibitory role of DJ-1 on VHL. Our in vitro studies also showed that DJ-1 inhibits ubiquitin ligase activity of VHL on HIF-1 by reducing the VHL-HIF-1 interaction. Importantly, accumulation of HIF-1 protects embryonic cortical neurons against MPP+ induced neuronal death. Finally, we confirmed the impairment of HIF-1 response to oxidative stress in human lymphoblastoids of DJ-1-linked PD cases. In the second part of this study, we demonstrated the interaction of DJ-1 and PON2. Interestingly, PON2 lactonase activity is reduced in DJ-1 deficient cells which could be rescued by re-introduction of DJ-1, suggesting a modulating role of DJ-1 on PON2 activity. In addition, PON2 deficiency, like DJ-1 deficiency, hypersensitizes neurons to MPP+, which could be rescued by over-expression of PON2 in both cases. Taken together, our data provide evidence that DJ-1 exerts its protective role by inhibiting VHL activity, enhancing HIF-1 stability, and increasing PON2 pro-survival function in PD models.

Análisis de fenómenos de transporte turbulento en la interacción de dos jets calentados a diferentes temperaturas

Ingeniera Civil Mecánica / El presente trabajo de título corresponde al análisis de la influencia de los términos de transporte de calor y momentum turbulentos, de las ecuaciones de Navier Stokes y del calor, en el confinamiento de una fuente de calor, que pudieran originar fugas de calor y gases en el uso de cortinas de aire con configuración Doble Jet Doble Flujo (DJ DF) como método de confinamiento al interior de túneles viales. Se estudiaron dos casos, considerando una cortina de aire DJ DF cuyo jet caliente está siempre en contacto con la zona confinada. Uno de ellos consiste en el uso del jet caliente con la mayor velocidad inicial de la configuración, y otro en que el jet frío posee la máxima velocidad inicial. Se realizó un estudio del campo, perfil y evolución en las zonas de capas de mezcla y línea central de la cortina, de los siguientes términos: esfuerzo de Reynolds, transportes axial y transversal de calor turbulentos, y sus derivadas parciales, las cuales evidencian su contribución en el incremento de flujo de momentum medio y de calor del flujo medio debido a fluctuaciones turbulentas. Dichos términos turbulentos fueron calculados con el método de diferencias finitas de orden O(h^4), utilizando datos numéricos extraídos de una simulación previa, que utilizó el modelo LES en ANSYS Fluent. Los esquemas forward , backward y central fueron aplicados en puntos extremos y central del dominio, respectivamente. Los resultados evidenciaron la existencia de un traspaso transversal de momentum y calor en niveles de la cortina de aire ubicados entre x/H=0.4 y 0.7, para ambos casos estudiados, siendo mayor en el del jet caliente más veloz. Esto, debido a peaks negativos en gráficos de evolución del transporte transversal de calor en capas de mezcla, que justificarían el intercambio de momentum y calor debido a fluctuaciones de las partículas fluidas. El rango de niveles mencionado coincide con la zona en donde ambos jets de la cortina se unifican para formar uno nuevo, con distintas características a los dos iniciales. La interacción entre ambos jets con distintas características termodinámicas disminuye la capacidad de confinamiento de la configuración propuesta. El análisis de transporte axial de calor muestra una inversión en el flujo de calor axial a partir de x/H=0.6 para el caso 1. Se concluye que la configuración que utiliza el jet frío de mayor velocidad inicial funciona un 60% mejor como barrera confinatoria respecto al caso del jet caliente más veloz. Se sugiere estudiar la influencia de los términos turbulentos en cortinas de aire inclinadas y analizar el paso de las partículas fluidas de manera de concluir sobre la naturaleza de los procesos de transporte de momentum responsables del esfuerzo de Reynolds.

Nothobranchius Fish: An Untapped Resource for Studying Aging-Related Neurodegeneration

New models in which aging-related neurodegeneration more closely resembling the combination of pathologies that develop in aging humans, are needed. The fish Nothobranchius, which naturally develops such pathologies over the course of its short lifespan, is one such model. This review compares the lifespans and pathologies of different Nothobranchius strains to those of current vertebrate models of aging. Furthermore, existing data pertaining to neurodegeneration in these fish is discussed in the context of their reported neuropathologies, along with open questions related to mammalian chronopathologies. Specifically, the evidence for a Parkinson’s disease-like pathology is discussed. Neurogenesis and age-related changes therein are discussed in the context of siRNA and neurodegeneration. We also discuss changes in the expression of neuropeptide Y in relation to the brain-gut axis and how these change with age. Age-related behavioral changes are discussed, along with the assays used in their evaluation. Genetic discoveries are outlined and discussed with a view on DJ-1/NRF2 signaling in N. furzeri, and insights gained from comparative genomics and siRNA studies. Finally, research focus areas are highlighted, and a case is made for the utility of these fish in the study of aging-related neurodegeneration, and to screen for environmental risk factors of aging-related neuropathology.

Man brukar säga att en bild säger mer än tusen ord. Att marknadsföra ett företag genom att använda bilder och video är en strategi som används i allt större utsträckning då det ses som ett starkt marknadsföringsverktyg. I moderna samhällen kan företag inte bara nöja sig med att erbjuda sina tjänster, de måste också sälja dem genom att skapa ett tydligt varumärke. Ett varumärke skapas av en image och denna image ska som mål kunna representera varumärket alldeles självt. Syftet med detta examensarbete är att få en bredare förståelse av marknadsföring via sociala och digitala medier. Examensarbetet har utförts på uppdrag av DJkollektivet “Håll Käften och dansa” där målet har varit att skapa en internationell webbsida för att marknadsföra dem. Syftet med webbsidan är att besökaren ska få en känsla av hur det kan vara att besöka en av deras spelningar. Den primära datainsamlingen har bestått av en kartläggning över vilken sorts information som visas på befintliga DJ's webbsidor där fokus har legat på menyer, sociala nätverk och vilken information de har valt att ha med på webbsidan. Detta för att skapa en förståelse för hur marknadsföring via internet ser ut idag. Vi har även utfört ett användartest på webbsidan för att analysera användarens reaktion på hur de upplever webbsidan och dess innehåll. Rapporten beskriver produktionens arbetsprocess och avslutas med att koppla samman den teoretiska delen till det slutgiltiga utformandet av webbsidan. Resultatet av detta arbete består av ett grafiskt kit som levereras i form utav; en webbsida, en egenproducerad video, en marknadsföringsguide samt våra egna fotograferade bilder till webbsidan. / It is often said that a picture is worth a thousand words. Marketing a company by using images and video is a strategy that is increasing in use as it is seen as a strong marketing tool. In modern societies, companies cannot limit themselves to merely offering their services. They must also sell by creating a clear brand. A brand is created by an image and this image should aim to represent the brand itself. The purpose of this thesis is to gain a broader understanding of marketing through social and digital media. This project was assigned by the DJ collective "Shut up and dance" where the goal has been to create an international website to market them. The purpose of the website is for the visitor to get a feeling of how it can be to visit one of their gigs. The primary data collection consisted of a survey orientation on the information that displayed on existing DJ websites. The focus has been on the menus, social networks and the information that they have chosen to include on the website. This is to create an understanding of how marketing on the Internet is today. We also performed a usability test on the website to analyze user's reaction to how they perceive the website and its content. The report describes the work process and concludes by linking the theoretical part of the final design to the website. The result of this work has a graphic kit supplied with the collection of; a website, a showreel, marketing guide and photos we have taken to the website.

Att stärka innovation i mjukvaruföretag

<p>Detta examensarbete har två huvudsakliga mål. Det ena är att utöka funktionaliteten i mjukvaran C3Loops för att underlätta för musiker som producerar elektronisk musik som vill använda mjukvaran i liveframträdanden. Det andra är att titta närmre på hur processmodeller för mjukvaruutveckling stödjer innovation. Denna rapport ger läsaren ett tvärvetenskapligt perspektiv. Tre för ämnet relevanta teoretiska områden, innovation, software-engineering och design tas upp. C3Loops projektet används som ett exempel på ett innovativt mjukvaruprojekt och det är arbetet inom detta projekt som står som modell och bakgrund till resultatet och slutsatsen.</p><p>Idén till mjukvaran C3Loops är resultatet av tekn.Dr Rikard Lindellʼs avhandling ”jag älskar att allt ligger överst”. Arbetet resulterade i en interaktiv prototyp som Lindell beslöt sig för att ta vidare och kommersialisera. Syftet med prototypen var att utforska bruksvärdet av ytinteraktion med pekskärmar. Mjukvaran är ett preformance verktyg för kollaborativa multimedia-framträdanden. Den skall skapa nya möjligheter för liveframträdanden och samarbeten mellan elektroniska musiker, Dj och Vj.</p><p>C3Loops projektet är fortfarande pågående. Plattformen som projektet produceras för är iPad plattformen. Release datum för projektet är tänkt att överenstämma med iPads releasedatum i Sverige. Rapporten leder till ett förslag på hur organisationer som vill förbättra sin innovationsförmåga kan gå tillväga. Detta i form av en projektmetod tänkt att stödja processen för att ta fram och pröva idéer till V1.0 mjukvara. Det återstår att testa och utvärdera metoden i skarpa projekt.</p> / <p>This thesis work has two main goals. The first is to extend the functionality of the C3Loops software in such a way as to make it usfull for a wider variety of electronic musicians The second is to examine how well software engineering process models support innovation. The C3Loops project is used as a case wich is studied in relation to this inquiry.</p><p>The idea for C3Loops is the result of the doctoral thesis ”i love the fact that everything is on top” by tekn.Dr.Rikard Lindell. The work on this thesis resulted in an interactive prototype, which Lindell decided to try and commercialise as a product. The original purpose of this prototype was to explore the value of surface interaction using touch screens in a collaborative multimedia preformance context. It creates new posibilities for live preformance and collaboration between electronic artists, Djs and Vjs.</p><p>The C3Loops project is still ongoing. The software is being produced for the iPad platform and its release is planned to coincide with the iPad release in Sweden. The result of this report is a suggestion on how organizations that develop software can improve their ability to innovate. This suggestion takes the form of a process model specifically designed to help support the creation and evaluation of idéas for V1.0 software. Future work includes testing this new method on real projects.</p>

Signalling pathways in renal cell carcinoma with a focus on telomerase regulation

Telomerase is a ribonucleoprotein complex that catalyses telomeric repeat addition at the ends of chromosomes. The catalytic subunit, hTERT, acts as a key determinant for telomerase activity control; the induction of hTERT expression is required for telomerase activity. hTERT participates in cellular immortalization and is elevated in certain malignant tissues. Several tumours exhibit telomerase activity, which contributes to the infinite proliferation capacity that promotes tumour progression. Renal cell carcinoma (RCC) represents 2% of all adult malignancies and has a high mortality rate. The WHO classifies RCC into several sub-types based on cytogenetic aberrations and morphological features; the most prevalent sub-types are clear cell (ccRCC), papillary (pRCC), and chromophobe RCC (chRCC). The aims of this thesis were to study the expression patterns of various signalling molecules, to elucidate the functional links among them, and to define the roles of these signalling molecules in the regulation of hTERT gene expression and telomerase activity in RCC. The first paper included in this thesis revealed mRNA overexpression of DJ-1 (a PTEN inhibitor), cMyc, and hTERT in clinical ccRCC samples compared to tumour-free kidney cortex tissues. Significant, positive correlations were detected for DJ-1, cMyc, and hTERT mRNA levels in ccRCC, but not in pRCC. In vitro knockdown of DJ-1 by siRNA in ccRCC cells induced downregulation of p-Akt, cMyc, hTERT, and telomerase activity. Forced overexpression of DJ-1 in an ovarian carcinoma cell line was followed by increased hTERT promoter activity, which appeared to be dependent on cMYC binding to the promoter. Collectively, the in vitro studies verified a functional link among DJ-1, cMyc, and hTERT as implied in the clinical ccRCC samples. The second paper included in this thesis demonstrated overexpression of NBS1 mRNA levels in ccRCC compared to the kidney cortex. NBS1 mRNA levels exhibited significant, positive correlations with DJ-1, cMyc, and S phase, but not with hTERT. In vitro experiments suggested that DJ-1 could regulate NBS1 gene expression. The role of the hTERT transcriptional repressor WT1 in RCC was evaluated in the third paper included in this thesis. ccRCC samples displayed low WT1 mRNA levels compared to kidney cortex samples. Interestingly, WT1 expression was negatively associated with hTERT and cMyc both of which were elevated in ccRCC. Forced overexpression of WT1 isoforms in a ccRCC cell line increased the expression of several negative transcriptional regulators of hTERT and diminished the expression of hTERT positive regulators. In consequence, hTERT mRNA levels and telomerase activity were reduced. Chromatin immunoprecipitation verified direct binding of WT1 to the cMyc, Smad3, and hTERT promoters. Taken together, these data suggested that in ccRCC, WT1 affects hTERT at the transcriptional level via a combined effect on both positive and negative regulators. In conclusion, DJ-1 can regulate hTERT and telomerase activity through the PI3K pathway encompassing PTEN, NBS1, p-Akt, and cMyc in ccRCC, but not in pRCC. WT1 negatively regulates hTERT and telomerase activity directly and indirectly through multiple pathways in ccRCC.
