The interaction of deprivation and delay of reinforcement under a fixed ratio schedule of respondingHilgert, Larry D. January 1975 (has links)
Motivation is an important factor in psychology, and much attention, therefore, has been devoted to the study of drives. Interaction of deprivation (hunger drive) and delay of reinforcement was studied in an operant framework in this experiment.Four male Sprague Dawley rats at approximately 250 days of age served as subjects in this experiment. The rats were exposed to a series of conditions within a counter-balanced design. The four conditions were: (a) 85 percent adjusted body weight with no delay; (b) 85 percent adjusted body weight with a 10 second delay; (a) 75 percent adjusted body weight with no delay; and (d) 75 percent adjusted body weight paired with a 10 second delay of reinforcement.Two major conclusions were made from this study. The independent variables may have individually influenced performance, and interaction between deprivation and a signaled delay also occurred when the two were placed in the same environment. Experimental evidence also suggested that discrimination of the delay was not as accurate at higher levels of deprivation.I would like to thank Gary F. This thesis analyzed data collected during a previous field fatigue study conducted at the Naval Officer Indoctrination School (OIS) in Newport, Rhode Island. An analysis of the effectiveness of a new watchstanding schedule for U.S. submarinersOsborn, Christopher M. 09 1900 (has links)
Approved for public release, distribution unlimited. / This study compares an experimental watchstanding schedule derived at Naval Submarine Medical Research Laboratory (NSMRL) with the schedule currently used onboard the submarine USS HENRY M. JACKSON (SSBN 730 GOLD). It analyzes subjective and objective data to determine if the new schedule is compatible in an operational submarine environment. This study reviews sleep and fatigue literature to emphasize important concepts needed to make schedule comparisons. Results from this study indicate a need exists among the U.S. submarine force to employ an operational schedule which provides more sleep and which is in better alignment with human circadian rhythms, thus improving cognitive effectiveness. One of the experimental schedules tested in this study yielded results similar to those of the existing submarine watchstanding schedule. This experimental schedule employed a validated model of human performance and fatigue to assess individual cognitive effectiveness. However, the results also indicate that the existing schedule is better suited in its accommodation of operational scheduling constraints which, in turn, allow watchstanders to receive more sleep. Recommendations address the need for the U.S. submarine force to continue to pursue a watchstanding schedule that provides better sleep while still accommodating operational constraints. Recommendations also address improvements in experiment implementation which can be integrated into future studies. / Lieutenant, United States Navy

Characterizing the Impact of Glucose Deprivation on the Lysine Acetyltransferase Complex NuA4Czosniak, David January 2017 (has links)
Upon the loss of glucose as the main carbon source cells have developed different mechanisms in order to adapt to this stress and promote survival. In Saccharomyces cerevisiae one such mechanism is acetylation, a post-translational modification performed by a lysine acetyltransferase (KAT) complex, such as NuA4, which has been previously shown to regulate different glucose metabolic pathways. Despite its known role upon glucose starvation, it is not currently understood how NuA4 itself is regulated in response to acute glucose deprivation (GD). I determine here that NuA4 complex protein levels (including catalytic protein Esa1), structure, activity, and localization are not impacted by acute GD. Despite GD showing no impact to NuA4 itself, it does result in the remodelling of both the interactome and acetylome of the complex where 160 proteins were identified to change interaction with Esa1-TAP and 93 acetylation sites were identified. As well GD results in a shift in localization of interacting proteins from nuclear upon standard growth to cytoplasmic. As well the changing interactome shows enrichment for proteins related to regulation of transcription and translation, metabolic pathways like glycolysis and gluconeogenesis, and others related to the cellular stress response. From the interactome three sets of proteins, Pab1, Eaf5/7/3, and Fas1 and Fas2, were studied further to greater characterize their interaction with NuA4 as they change interaction upon GD. Eaf7 protein levels were shown to decrease upon GD and both Fas1 and Fas2 levels were shown to increase in response to NuA4 deletion mutants. Together this work provides a greater understanding of the cellular response to acute GD stress, and how NuA4 plays a role in response to that stress in order to promote cell survival.
