Long-term consequences of perinatal high-fat feeding on dopamine function and metabolism in rats

This research project investigates the long-term consequences of perinatal exposure to high-fat (HF) on the mesocorticolimbic dopamine (DA) system. Adult offspring of mothers fed a HF diet (30% fat, compared to 5% in control mothers (C)) during the last week of gestation and throughout lactation displayed decreased locomotion in response to an acute amphetamine challenge and decreased behavioral sensitization to repeated amphetamine compared to C animals. These behavioral effects were accompanied by small increases in tyrosine hydroxylase expression in the ventral tegmental area and significant increases in DA and DOPAC content in the NAc, suggesting an elevated DA tone in this target field. In the NAc, there were no significant changes in D1, D2 receptors or DA transporter (DAT) levels between diet groups. The behavioural and biochemical data were collected in adulthood, long after the termination of the diet suggesting that a HF perinatal diet is inducing permanent changes within the DA system and might contribute to the development of metabolic disturbances.

Improvement of Release Criteria for Immediate Release Solid Oral Dosage Forms

Herewith are presented the results of an investigation the statistical power of USP compendial release tests and recommended alternatives. <br>The U.S. drug supply chain, formerly protected by a closed distribution network, is now threatened by the legal and illegal importation of drug products. Whereas quality can never be inspected into final products, compendial release standards may represent the only valid assessment that products of dubious origin would receive. Reliable tests for content uniformity and dissolution are required to protect the safety of the supply chain. A study was designed to test the hypothesis that existing compendial tests for content uniformity and dissolution would protect the supply chain against substandard and counterfeit drugs if basic field tests failed. <br>Compendial tests for content uniformity and dissolution were evaluated for statistical power using simulation studies. The results revealed that the revised content uniformity test, based on tolerance analysis, was subject to an unacceptable level of consumers' risk. The Bergum method proved to be an excellent secondary standard for product assessment and is recommended as an alternative to the USP method. Simulations with the USP dissolution test revealed significant weaknesses and inconsistencies in the test structure. Theoretical models and power assessments confirmed that the coverage specification of the dissolution test was an unacceptably high 50% coverage with 50% confidence. <br>A Bayesian D-optimal design program was used to investigate alternative methods to improve the coverage capability of the USP dissolution test. The result of this program was the identification of two alternatives to the existing USP procedure. The first alternative is based on the addition of attribute coverage tests to stages 2 and 3 of the USP test, whereas the second alternative is based on the concept of tolerance analysis. <br>Validation studies confirmed that both alternatives significantly improved the statistical power of the USP dissolution test without increasing the sample size or modifying the current three-stage procedure. <p>Infusions, decoctions, alcoholic preparations and other dosage forms of Artemisia afra are frequently used in South African traditional medicine. Generally when these preparations are made without applying good manufacturing practices they do not meet microbial quality control standards, safety and toxicity criteria and encourage poor patients compliance. To overcome the aforementioned disadvantages of traditional dosage forms a sold dosage form, i.e. a table might be recommended. The first objective of this study was to formulate and manufacture a rapid release tablet dosage of Artemisia afra that would contain an amount of plant material equivalent to that found in its traditional liquid dosage forms and that would meet conventional pharmaceutical standards. The second objective was to conduct a pilot study to obtain a preliminary profile of the bioavailability of select flavonoids presents in both the tablet and traditional liquid preparation of Artemisia afra in humans.</p>

Dosage compensation is the equalisation of gene expression from unequal doses of genes. Drosophila males up-regulate transcription from their single X chromosome to equal that from the two female X chromosomes. Five malespecific lethal (msl) genes are required in males, and encode the main agents of the up-regulation. At least these proteins, together with either or both of two noncoding RNAs, form the MSL chromatin-modifying complex. Female-specific translational repression of a key component, msl2, limits the complex to males. The MSL complex binds to the X chromosome at hundreds of distinct loci, acetylates nucleosomes, and de-condenses the chromatin. Together with possibly many co-factors, the transcriptional up-regulation caused by MSL complex appears to counteract repressive factors to achieve an average effect of transcriptional doubling. Here, I have studied the initiation of MSL regulation on the X chromosome with a variety of approaches. In order to study early events, dosage compensation was induced in females with ectopic expression of msl2 from the tetracycline system. However, low background expression without activation prohibited further studies. To identify novel factors that affect dosage compensation, a reporter gene system based on variable eye size was evaluated. The system provided a dose-dependent phenotype, but could not report additional up-regulation by the MSL complex, and was thus unsuitable for the proposed mutational screen. The quantifiable lacZ gene was measured in a strict comparison of expression from an eye-specfic (GMR) or a constitutive (armadillo) promoter. At defined locations on the X chromsome, armadillo-lacZ acquired local compensation, but GMR-lacZ did not. Further modifications upstream of GMR-lacZ increased the response, and confirmed the importance of the promoter in attraction of dosage compensation. To corroborate this with the established importance of genic sequences in MSL attraction, a combinatorial model of attraction is proposed. The relative importance of early or constitutive expression was also tested, by providing GMR-lacZ with extra expression through the tetracycline system. A burst of embryonic expression, and constitutive expression, were both insufficient to increase dosage compensation of the transgene. Finally, the compensation of GMRmediated transgenes was confounded by ‘transvection’ effects of chromosome pairing. This effect may have wider implications on the study of compensation at individual genes.
