Aspectos ecotoxicológicos dos inseticidas diflubenzuron e teflubenzuron para o pacú (Piaractus mesopotamicus) /

Na aqüicultura, o tratamento com inseticidas pode ser realizado com a aplicação direta na água, ou administração na ração dos animais. Apesar de muito utilizados, poucos são os trabalhos sobre ecotoxicologia, comportamento e bioacumulação do diflubenzuron (DFB) e teflubenzuron (TFB) na água e seus efeitos em organismos aquáticos de clima tropical. Os objetivos deste trabalho foram: estimar a toxicidade (CL(50-96h)) do DFB e TFB para o pacu (Piaractus mesopotamicus), calcular o risco alimentar para o consumo humano, determinar resíduos de DFB e TFB na água, sedimento e músculo de pacus, após a exposição aos inseticidas na água ou na ração dos peixes e analisar a atividade hepática das enzimas glutationa-S-transefrase (GST) e catalase (CAT) em pacus alimentados com ração acrescida de DFB ou TFB. A CL50;96h do DFB para o pacu foi superior a 1200 mg.L-1 e a do TFB, superior a 1000 mg.L-1. Os resíduos de DFB foram detectáveis até 7 dias após a aplicação (com 18,3% de resíduo) e para o TFB, foram detectáveis ate o terceiro dia (8% de resíduo). No sedimento, as maiores concentrações de DFB e TFB foram observadas nas primeiras 24 h e em todos os períodos amostrados foram detectados resíduos dos inseticidas. Nos filés dos pacus foram detectados resíduos dos inseticidas após a alimentação com a ração tratada e após aplicação de DFB e TFB na água. A ingestão diária estimada (IDE) de DFB e TFB encontra-se acima dos valores de ingestão diária aceitável (IDA), mesmo para as amostras com menores valores de resíduos detectados nos filés do pacu. In aquaculture these pesticides can be injected directly in the water or in the fish food. Besides their large utilization, there are few works about the effects of these insecticides on the fish metabolism and biomagfication, especially in tropical aquatic organisms. This study aims to: evaluate the acute toxicities (LC(50-96h)) in pacu (Piaractus mesopotamicus), measure the risk food for human consumption, determinate residues of DFB and TFB in water, sediment and muscle of pacus after the management of the insect-powders on water or in the fish food and to analyze the hepatical activity of the enzymes catalase (CAT) and glutathione-S-transferase (GST) in fishes fed with ration plus DFB or TFB. The LC((50- 96h)) of DFB and TFB for the pacu was higher than 1.200 mg.L-1 and 1.000 mg.L-1, respectively. The residues TFB were detected until the thirth day (8% of residue) and DFB until seventh day after application in the water (18,3% of residue). In the sediment, the highest concentrations of DFB and TFB had been observed in the first 24 h and in all the samplings there were . In muscle of pacus, residues of the insecticides had been detected after the feeding with the ration treated and after application to DFB and TFB in the water. The daily ingestion estimate (DIE) of DFB and TFB meets above of the values of acceptable daily ingestion (DAI), exactly for the samples with lesser values of residues detected in filés of pacu. For the enzymes the CAT activity was not affected by both pesticides, but the GST activity was reduced five days after the feeding. Ecotoxicidade e eficácia da oxitetraciclina e do florfenicol contra infecção experimental por Aeromonas hydrophila e aspectos histopatológicos em pacu (Piaractus mesopotamicus) /

Resumo: Um dos problemas da aqüicultura é a ocorrência freqüente de infecções por parasitos e bactérias, principalmente em sistemas de criação intensiva. Entre os antibióticos, a oxitetraciclina (OTC) e o florfenicol (FFC) são os mais usados para tratar infecções causadas por bactérias em peixes. Os objetivos foram estimar a concentração letal 50% (CL50;48h) da OTC (Terramicina®) e do FFC (Aquaflor®) para o pacu (Piaractus mesopotamicus), classificar a OTC e o FFC pelo risco de intoxicação ambiental; determinar a dose eficaz de OTC e de FFC administrada na ração para o controle da infecção por Aeromonas hydrophila em pacu; avaliar o efeito da diluição da OTC e do FFC nas variáveis de qualidade de água durante os testes de toxicidade e durante o tratamento e avaliar as alterações histopatológicas após o tratamento com OTC e FFC. Para os testes de toxicidade aguda os animais (peso entre 0,5 e 1,0 g) foram expostos a 7,0; 7,5; 8,0 e 8,5 mgL-1 de OTC ou a 600,0; 700,0; 800,0; 900,0 e 1000,0 mgL-1 de FFC e um tratamento controle. A OTC e o FFC foram classificados pela toxicidade aguda e pelo risco ambiental de acordo com as classes de valores de CL50;48h. Para o tratamento de infecção induzida por A. hydrophila (3.0 x108 bactéria.mL-1), foram realizados dois testes, um com OTC (110,0; 140,0 e 170,0 mg.Kg-1) e outro com FFC (5,0; 10,0 e 15,0 mg.Kg-1). Os tratamentos foram três concentrações de cada antibiótico, um controle sem infecção e um com infecção. Para análise histopatológica foram coletados fragmentos de brânquia, fígado, rim e pele dos peixes após o tratamento com os antibióticos. Among the antibiotics, the oxytetracycline (OTC) and the florfenicol (FFC) are the most commonly used to treat infections caused by bacteria in fishes. The objective was to estimate the 50% lethal concentration (LC50, 48h) of OTC (Terramycin®) and of the FFC (AQUAFLOR®) for the pacu (P. mesopotamicus), classify the OTC and FFC of according the assessment risk and to determine the effective concentration of OTC and of FFC administered in feed for control of Aeromona hydrophila in pacu; to evaluate the effect of dilution of OTC and of the FFC in variables in water quality during toxicity tests and for treatment and to evaluate the histopathological changes after treatment with OTC and FFC. For acute toxicity tests the animals (weighing between 0.5 and 1.0 g) were exposed to 7.0, 7.5, 8.0 and 8.5 mg.L-1 OTC or 600.0, 700 , 0, 800.0, 900.0 and 1000.0 mg.L-1 of FFC and a control treatment. The OTC and the FFC were classified for acute toxicity and for environmental risk of according with the ranking of value of LC 50;48h. For the treatment of A. hydrophila induced by experimental infection (3.0 x108 bactéria.mL-1), were carried out two tests, one with OTC (110.0, 140.0 and 170.0 mg.Kg-1) and another with FFC (5.0; 10.0 and 15.0 mg.Kg-1). A fim de analisar o impacto da contaminação química e orgânica na espécie de estudo, foram realizadas análises ecotoxicológicas visando avaliar a ação de alguns dos principais componentes envolvidos com a proteção ao estresse oxidativo e metabolismo de detoxificação de fase I e II, e capacidade antioxidante. Os resultados indicam a influência de efluentes domésticos e industriais no metabolismo de detoxificação e estresse oxidativo. No entanto, apesar do aumento da atividade EROD e o efeito da atividade da GST, os valores médios de GST na área urbana estão dentro daqueles esperados em condição de hipóxia descritos na literatura. Esta observação sugere que o aumento da GST em resposta à produção de ERO pela presença de poluentes, aumenta a rede de defesa antioxidante, controlando os danos oxidativos causados pela hipóxia e pela reperfusão. Com o intuito de estimar a condição, que reflete a aptidão (fitness) individual do animal, foi avaliada a relação matemática entre peso e comprimento, que indicam alterações na forma do corpo e no incremento em peso, que permitem inferências sobre a saúde e o bem-estar animal. Os dados gerados informam diferenças em condição que estão associadas à área de avaliação, mas também ao sexo e período reprodutivo, e gradiente de contaminação indicando forte influência de estressores ambientais na fisiologia dos espécimes analisados. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The objective of this study was to evaluate the role of human activity influence, such as environmental contamination and habitat changes as drivers for changing physiological, biochemical, and genetic diversity among Geoffroy's side-necked turtle populations in one of the most impacted watersheds in southeastern Brazil. In order to analyze the impact of chemical and organic contamination, ecotoxicological analyses were performed to assess the action of some of the major components involved in oxidative stress protection and phase I and II of metabolism detoxification, and antioxidant capacity. The results indicate the influence of domestic and industrial effluents in detoxification metabolism and oxidative stress. However, in spite of increased activity and effect of EROD activity of GST, GST-average values in the urban area conform with those expected in hypoxia condition in the literature. This observation suggests that increased GST in response to the ROS production by the presence of pollutants increases the antioxidant defense network, controlling the oxidative damage caused by hypoxia and reperfusion. In order to estimate the condition, which reflects the individual ability (fitness), we evaluated the mathematical relationship between weight and length, indicating that changes in body shape and weight increase, allowing inferences about the health and well animal welfare. The data generated indicate differences in conditions that are associated with the area, but also with sex and reproductive period, and contamination gradient indicating strong influence of environmental stressors on the physiology of the specimens. Alterações hematológicas e bioquímicas em uma população de Phrynops geoffroanus (Schweigger, 1812) em reposta a estressores ambientes /

A espécie Phrynops geoffroanus, popularmente conhecida como "Cágado-de-Barbelas", tem se proliferado em ambiente poluído, sujeita a condições adversas que podem influenciar nos seus hábitos de vida e condições fisiológicas. Estudos que monitorem os efeitos da contaminação ambiental são fundamentais para o conhecimento da biologia da espécie e delineamento de estratégias de conservação efetivas. Deste modo, as análises dos parâmetros hematológicos e bioquímicos mostram-se importantes na avaliação da saúde de animais silvestres e riscos para o animal e ecossistema. O objetivo do trabalho foi verificar a influência do ambiente na fisiologia de uma população de Phrynops geoffroanus por meio da avaliação da capacidade antioxidante e respostas aos estressores ambientais, em comparação com espécimes de local sob condições controladas. Foram avaliadas, por meio do hemograma completo, dosagem de espécies reativas ao ácido tiobarbitúrico (TBARS), ensaio da capacidade antioxidante em equivalência ao Trolox (TEAC) e atividades das enzimas antioxidantes catalase e glicose-6-fosfato desidrogenase (G6PDH), amostras sanguíneas de sessenta espécimes, sendo trinta provenientes do córrego Felicidade, ambiente poluído, no perímetro urbano de São José do Rio Preto-SP; e trinta do criatório "Reginaldo Uvo Leone", Tabapuã-SP, local sob condições controladas, cujas amostras constituem o grupo controle dos experimentos. Houve grande variação nos parâmetros hematológicos de Phrynops geoffroanus de ambiente urbano. The species Phrynops geoffroanus, popularly known as "Geoffroy's side-necked turtle", has proliferated in polluted environments, subject to adverse conditions that could influence their living habits and physiological conditions. Studies that monitor the effects of environmental pollution are the key to understand the species biology and design effective conservation strategies. Thus, the analysis of hematological and biochemical parameters showed to be important in assessing the health of wild animals and for the animal and ecosystem risks. This work aimed to assess the environmental influence on the physiology of a Phrynops geoffroanus population through the evaluation of antioxidant status and responses to environmental stressors, compared to specimens from a place under controlled conditions. They were evaluated by hemogram, measuring thiobarbituric acid reactive species assay (TBARS), Trolox-equivalent antioxidant capacity evaluation (TEAC) and the activities of antioxidant enzymes, catalase and glucose-6-phosphate dehydrogenase (G6PDH), blood samples of sixty specimens, thirty from the Felicidade stream, polluted environment, within the São José do Rio Preto city, and thirty from the "Reginaldo Uvo Leone" breeding farm, Tabapuã-SP, place under controlled conditions, whose samples constitute the control group of the experiments. There was a wide variation in hematological parameters of Phrynops geoffroanus from the urban environment.
