Cianopeptídeos inibidores de proteases produzidos por cianobactérias brasileiras / Cyanopeptides proteases inhibitors produced by Brazilian cyanobacteria

As cianobactérias são micro-organismos reconhecidos por seu potencial em produzir cianotoxinas que afetam não só o ecossistema e a outros organismos dos ambientes aquáticos, mas também aos seres humanos, agindo em diversos órgãos e tecidos. Cerca de 600 metabólitos secundários produzidos por cianobactérias já foram descritos na literatura, sendo que muitos deles possuem potencial biológico. Os peptídeos de baixo peso molecular, produzidos por cianobactérias chamados cianopeptídeos, dos quais se podem citar as anabaenopeptinas, aeruginosinas, microviridinas, cianopeptolinas e microgininas, são compostos provindos do metabolismo secundário de cianobactérias e são descritos como inibidores de proteases e fosfatases em alguns sistemas biológicos. Sendo assim, o objetivo do estudo foi identificar a ocorrência de cianopeptídeos em cianobactérias brasileiras e testar seu efeito de inibição da atividade sobre enzimas proteases. Os objetos de estudo foram: uma linhagem da espécie Sphaerospermopsis torques-reginae, duas de Cylindrospermopsis raciborskii, duas de Microcystis sp, uma de Oscillatoria e uma de Pseudanabena sp. A partir dos resultados obtidos pode-se afirmar que do total de linhagens analisadas, ao menos 4 destas parecem produzir os cianopeptídeos de interesse, quando avaliados por cromatografia líquida de alta eficiência com detector de arranjo de diodos (HPLC-DAD). Em seguida, culturas de duas linhagens de C. raciborskii (uma produtora e outra não produtora de saxitoxina) foram amostradas a cada 3 dias, para a avaliação do crescimento celular, produção de cianopeptídeos e de saxitoxina e suas variantes. Não foi possível confirmar a produção de cianopeptídeos nas duas linhagens desta espécie. Por outro lado, foi evidenciado um aumento da produção de saxitoxinas quando cultivada num meio sem nitrogênio em comparação com a condição controle. Quando analisada a linhagem de Microcystis sp. (LTPNA 08), produtora de microcistinas, foi possível confirmar por cromatografia líquida acoplada a espectrometria de massas (LC-MS) a produção de dois cianopeptídeos, sendo estes, duas microgininas. Desta forma, desenvolveu-se um método cromatográfico para a separação desses compostos e purificação por cromatografia líquida semi-preparativa, onde foi possível a obtenção de frações enriquecidas de microgininas e microcistinas-RR e LR, com cerca de 70%, 86% e 97% de pureza. Por fim, realizaram-se ensaios avaliando a atividade da enzima conversora de angiotensina (ECA) e aminopeptidase M (AMP M) com as frações isoladas de microgininas e microcistina-LR. A atividade da ECA foi ± 50% inibida pelas frações testadas de cianopeptídeos, microcistina-LR isolada e microcistins-lR comercial. A atividade da AMP M foi 100% e 24,5% inibida quando incubada com 20 µM da fração de microgininas e microcistina-LR, respectivamente. Desta forma, as microgininas isoladas de cianobactérias brasileiras, mostraram-se como importantes inibidores da ECA e AMP M, podendo, no futuro, serem utilizadas como compostos para o tratamento de patologias cardiovasculares e renais. / Cyanobacteria are micro-organisms recognized for their potential to produce cyanotoxins that affect not only the ecosystem and other organisms of aquatic environments, but also humans, acting in various organs and tissues. Around 600 secondary metabolites produced by cyanobacteria have been described in the literature; many of them have biological potential. Low molecular weight peptides produced by cyanobacteria are called cyanopeptides, among them we can cite the anabaenopeptins, aeruginosins, microviridins, cyanopeptolins and microginins, these compounds are derived from secondary metabolisms of cyanobacteria and apparently cause inhibition of proteases and phosphatases in some biological systems. Therefore, this study targeted the identification of the occurrence of cyanopeptides in Brazilian cyanobacterias and testing its effect on the inhibition of proteases activity. The targets of study were: a strain of species Sphaerospermopsis torques-reginae, two strains of Cylindrospermopsis raciborskii, two strains of Microcystis sp. and, one strain of Pseudanabena and Oscillatoria sp. From the results obtained in this study it can be stated that at least four of the total of strains analyzed appear to produce cyanopeptides of interest, when analyzed by high-performance liquid chromatography whit phodo diodo array detector (HPLC-PDA). The cultures of two strains of C. raciborskii (a producer of saxitoxin and a non-producer) were sampled every 3 days for assessment of cell growth, production of cyanopeptides and saxitoxins. It was not possible to confirm the production of cyanopeptides in strains of this species. Nevertheless, an increase in production of saxitoxins was shown when cultivated in an environment without nitrogen, as compared to the control condition. When the strain of Microcystis sp. (LTPNA 08), a producer of microcystins, was analyzed, the production of two cyanopeptides was confirmed by using liquid chromatography-mass spectrometry (LC-MS). After confirmation, a method using HPLC-PDA was used to do the separation and purification of these compounds by semi-preparative chromatography, in which it was possible to obtain an enriched fraction of microginins, microcystin-RR and microcystin-LR with approximately 70%, 86% and 97% purity, respectively. Lastly, inhibition experiments were run with angiotensin-converting enzyme (ACE) and aminopeptidase M (AMP M) with the isolated fractions. The microginins fractions, MC-LR commercial and MC-LR isolated, showed an inhibition of ± 50% of the angiotensin-converting enzyme. The activity of AMP M was 100% and 24.5% inhibited when incubated with microginins and microcystin-LR fractions in a concentration of 20 µM, respectively. Thus, isolated microginins from Brazilian cyanobacteria have exhibited properties as potential therapeutic agents in development of inhibitors of ACE and AMP M, which can be a benefit of using these molecules in the treatment of cardiovascular and renal pathologies.

Identification of agricultural and industrial pollutants in the Kat River, Eastern Cape and their effect on agricultural products found along the river banks

There is growing concern that commonly used Pharmaceuticals and Personal Care Products (PPCPs) and pesticides are entering and contaminating drinking water supplies. The use of targeted quantitation of PPCP has been well established but there is an emerging trend to also screen for and identify unexpected environmental pollutants. Chemicals like pesticides hormones and antibiotics are especially of interest because of proven endocrine disrupting effects and a possible development of bacterial resistance. Powerful screening methods are required to detect and quantify the presence of these compounds in our environment. PPCP encompass a wide range of pollutants, including Endocrine Disrupting Compounds (EDC), pesticides, hormones, antibiotics, drugs of abuse, x-ray contrast agents and drinking water disinfection by-products to name a few. In order to properly assess the effects of these compounds on our environment, it is necessary to accurately monitor their presence. The diversity of chemical properties of these compounds makes method development challenging. LC/MS/MS is able to analyse polar, semi-volatile, and thermally labile compounds covering a wide molecular weight range. The new AB SCIEX TripleTOF™5600 LC/MS/MS was used to profile environmental samples for unexpected pollutants, to identify and characterise the chemical composition and structure of the pollutants, and to quantify (based on intensity) the concentration in collected water samples. Liquid Chromatography coupled to tandem Mass Spectrometry (LCMS/ MS) is able to analyse polar, semi-volatile, and thermally labile compounds covering a wide molecular weight range, such as pesticides, antibiotics, drugs of abuse, x-ray contrast agents, drinking water disinfection by-products etc. More recently there is a growing interest from environmental researchers to also screen for and identify non-targeted compounds in environmental samples, including metabolites and degradates, but also completely unexpected pollutants. The new AB SCIEX TripleTOF™5600 LC/MS/MS system is capable of performing highly sensitive and fast MS scanning experiments to search for unknown molecular ions while also performing selective and characteristic MS/MS scanning for further compound identification and, therefore, is the instrument of choice for this challenging task. General unknown screening workflows do not use a target analyte list and compound detection is not based on any prior knowledge, including retention times and information on possible molecular and fragment ions. Therefore, acquired chromatograms are very rich in information and can easily contain thousands of ions from both any compounds present in the sample as well as from the sample matrix itself. Thus, powerful software tools are needed to explore such data to identify the unexpected compound. Water samples were collected both upstream and downstream of two WWTPs (Seymour and Fort Beaufort) and were directly injected on the AB SCIEX TripleTOF™5600 LC/MS/MS after being filtered. 15 sample points along the Kat River, ranging from a point as close to the source as possible to a point just before it joins the Great Fish River were used. The samples collected from the source were used as the control in each of the experiments, the assumption being the closer you get to the source, the less contaminated the water would be for the analysis of pesticides. Points were selected where the Kat River crosses the R67 or on farms where the river was accessible using farm roads. Samples were collected from October 2013 to November 2014.The Peak view software and Analyst software were used in the analysis of PPCPs. The XIC Manager allows you to manage large lists of compounds and perform automatic extracted ion chromatogram (XIC) calculations and review results operations. The results were displayed in the chromatogram pane and the XIC table (see results). The results reported here in this thesis indicate that there is contamination in the Kat River water due to both pesticides and PPCPs. The results also indicate that the food products are also contaminated and hence both the Kat River agricultural produce and its water need to be closely monitored for both pesticide and PPCPs contaminants. Further studies to investigate the quantitative levels of pesticides and PPCPs in the Kat river water to determine if the concentration levels of the detected pesticides are below the reported Maximum Residues Limits will be explored in the future.

As microcistinas constituem uma família de heptapeptídeos cíclicos com cerca de 70 variantes caracterizadas. São biossintetizadas em condições específicas por cianobactérias, principalmente em corpos d\'água eutrofizados. Apresentam organotropismo ao fígado e DL50 entre 50-500 µg.kg-1 peso (rato, i.p.). A exposição aguda pode impactar a saúde humana ao desarranjar o parênquima hepatocelular por hiperfosforilação do citoesqueleto e produzir intenso sangramento, podendo levar à morte por choque hipovulêmico um homem adulto e são promotoras tumorais, quando ocorre exposição crônica, por inibição de fosfatases 1 e 2A. Os congêneres das microcistinas apresentam toxicidades distintas e é importante caracterizá-los em corpos d\'água para se estimar o risco à saúde humana. Na literatura, não há convergência quanto à forma de extração das toxinas, preparo das amostras para análise e condições cromatográficas para o isolamento e quantificação. Neste trabalho, diversas análises foram conduzidas para se determinar as melhores formas de extração das toxinas, clean up da amostra e condições cromatográficas para amostras de campo ou cultura de cianobactérias. A extração hidro-alcoólica (MeOH:H2O, 3:1, v/v) parece ser a melhor forma de extração, seguida de partição líquido-líquido para amostras de campo (florações) e extração em fase sólida (cartuchos C18) para algas cultivadas em laboratório. Por meio de isolamento por cromatografia líquida de alta eficiência (HPLC) com detectores UV-VIS e pela análise do padrão de fragmentação das moléculas por espectrometria de massas (MS) com ionização por eletrospray em Tandem (ESI-MS/MS) determinou-se a presença das variantes de microcistinas MC-RR, MC-LR, MC-YR e MC-hRhR na amostra de água do Reservatório Billings e [Asp3]MC-LR na cepa Microcystis aeruginosa BCCUSP 235. Este método parece ser adequado para a extração e caracterização de microcistinas presente em corpos d\'água e em cultura de laboratório e a variante MC-hRhR foi caracterizada pela primeira vez na literatura mundial. / It is well known that eutrophicated lakes and water reservoirs can be a serious concern to wildlife, domestic livestock and human health since cyanobacterial blooms are prone to occur. Some cyanobacteria (blue-green algae) species which form bloom may biosynthesize a family of hepatotoxic peptides named microcystins under certain circumstances, related specially to nitrogen: phosphorus ratio, temperature, pH and light intensity of the water body. There are at least 70 variants of microcystins with different toxicity, DL50 ranging from 50 to 500 µg.kg-1 (in mouse, i.p.). Acute exposure provoke hyperphosphorylation of the cytoskeleton, in liver cells, leading to cellular disruption with haemorrhage and the long term exposure can cause tumor promotion by inactivation of serine/threonine phosphatases 1 and 2A (PP1 and PP2A). Therefore, it is very important to determine and quantify microcystin variants present in water reservoirs. There are several methods available to evaluate qualitative and quantitatively microcystin and its variants in water reservoir and estimate human risks. Such methods comprise biological, biochemical and physico-chemical assays. In this work, we have isolated and analyzed hepatotoxins by High Performance Liquid Chromatography (HPLC) with photodiode array (PDA) detector and their structures were elucidated by Tandem - MS/MS with eletrospray ionization (ESI-MS/MS). MC-RR, MC-LR, MC-YR and MC-hRhR were determined in samples collected at Billings reservoir and [Asp3]MC-LR was found in the strain Microcystis aeruginosa BCCUSP 235. The procedures developed herein can be a powerful tool for isolation and determination of microcistin and its derivatives founded in water reservoirs. Also, we have described for the first time a new variant of microcistin: MC-hRhR.

Does Thermo-tolerance in Daphnia depend on the mitochondrial function?

The thermotolerance, an adaptive phenomenon that is accompanied by the phenotypic plasticity which is the adjustment of physiology, biochemistry and metabolism of every cellular function by the hidden mechanism. Mitochondrion, the powerhouse of the cell that determines the functional integrity of every cellular homeostasis and functional phycological processes should provide its association in regulating the thermotolerance as well. This study assessed the mitochondrial function in regulating and determining the limit of thermo tolerance in the Daphnia magna of different geographical regions of the world, mainly sub grouped as temperature tolerant clones (IL) and temperature sensitive clones (GB). The acclimation effects or the adjustment of the preexisting biological properties help the organism adjust its biological processes to the changing habitat to maintain the cellular functional integrity. The clonal divergence as well as the acclimation show a clear pattern in limiting the thermotolerance and the prediction is the temperature tolerant clones should show higher adjustment of the mitochondrial function than temperature sensitive ones. We hypothesize that the damage in the mitochondrial membrane integrity by different mito-toxins should decrease the heat tolerance by decreasing the membrane potential and fluidity. The integrated mitochondrial function was assessed in acclimated clones by using the molecular studies as well as observation of behavioral and phenotypic plasticity. Due to the specific effects of each mito-toxins (CCCP, NaN3 and DNP) on different complexes (I-IV and ATP synthase) in ETC, we determined the mitochondrial membrane integrity by the Rhodamine 123 alongside with the lactate assay for measuring the mitochondrial integrity. Among all these three mito-toxins, CCCP show significant effect on limiting the heat tolerance. The lower lactate accumulation was observed in the temperature-tolerant clones acclimated in cold temperatures (18°C) which indicates the higher mitochondrial adjustment than the temperature sensitive clones. The concluding remark is that thermal tolerance is determined by the adjustment of mitochondrial function which accompanied with the adjustment to the mitochondrial respiration as well as the adjustment to membrane potential and fluidity.

Analysis of organochloro-pesticides in Eritrean water and sediment samples

Thesis (MSc)--Stellenbosch University, 2004. / ENGLISH ABSTRACT: The choice of the sample preparation technique used in combination with a chromatographic technique in environmental analysis strongly influences the performance of the method as a whole. The main aim of this work was to evaluate methods like: micro liquid-liquid extraction (цLLE)and solid phase micro extraction (SPME) in combination with gas chromatography-electron capture detector (GCECD) and a recently developed technique named stir bar sorptive extraction (SBSE) combined with thermal desorption system-programmable temperature vaporizationgas chromatography-mass spectrometer (TDS-PTV-GC-MS) for the analysis of organochloro-pesticides (OCPs) in natural water and to make a comparison between them. The most suitable method was then selected for the analysis of water samples collected from Eritrea. An additional goal was to develop a sensitive method for the analysis ofOCPs in soil and sediment samples. цLLE-GC-ECD and SPME-GC-ECD methods were evaluated for the analysis of OCPs in water. The former allowed achieving a sensitivity of less than 100 ppt and the latter was able to detect down to the 1 ppt level. Both methods were found to be repeatable with %RSD of3% to 19% and 5% to 12% as well as linear with a range of 1:103 and 1:5x103 respectivey. SBSE-TDS-GC-MS was also investigated for the analysis of OCPs in water samples. With this technique a good sensitivity down to 1 ppt was also obtained. The less sensitive nature of the MS compared to ECD was hereby compensated for by the large amount of polydimethylsiloxane (PDMS) phase on the stir bar. The method was repeatable and linear with a range of 1:5000, which is similar to the one obtained for the SPME-GC-ECD method. All three methods were also tested with real contaminated water samples and a comparison of the three techniques in terms of sensitivity, linearity, repeatability, availability and cost effectiveness was done. As a method of choice in terms of the above criteria, SPMEGC- ECD was applied to the water samples collected from Eritrea. The presence of some oePs such as a-BBC (benzenehexachloride), heptachlor, heptachlor epoxide, endosulfan I, p,p'-DDE (dichlorodiphenylchloroethylene), endosulfan II, p,p'-DDD (dichlorodiphenyldichloroethane), endosu!fan sulfate and p,p'-DDT (dichlorodiphenyltrichloroethane) in some of the Eritrean water samples was demonstrated The SPME-GC-ECD technique for the analysis OCPs in soil and sediment samples was investigated. It was shown to be able to detect down to 1 pg/g (lppt) with good linearity and repeatability. The method was also evaluated for authentic soil samples. Due to a lack of time the method could not be applied for the analysis of the soil and sediment samples collected from Eritrea. / AFRIKAANSE OPSOMMING: Die keuse van 'n gepaste monster-voorbereidingsmetode wat gebruik word tesame met 'n chromatografiese skeidingtegniek in omgewings-analise het 'n beduidende effek op die algehele metode. Die doel van hierdie werk was om metodes soos mikrovloeistof- vloeistof ekstraksie (цLLE) en soliede fase mikro ekstraksie (SPME) in kombinasie met gaschromatografie met elektronvangs deteksie (GC-ECD) te ontwikkel vir die analise van organochloor-pestisiede (OCPs) in natuurlike water. Vervolgens is hierdie metodes vergelyk met die onlangs-ontwikkelde roerstaaf sorptiewe ekstraksie tegniek (SBSE) in kombinasie met termiese desorbsie-GCmassaspektrometriese deteksie (TDS-GC-MSD). Die mees geskikte metode is vervolgens gebruik vir die analise van Eritreaanse watermonsters. Daarbenewens was die doelook om 'n sensitiewe metode te ontwikkel vir die analise van OCPs in gronden sedimentmonsters. Beide цLLE en SPME-GC-ECD metodes is gekarakteriseer deur goeie herhaalbaarheid en lineariteit. Die sensitiwiteit van bogenoemde metodes was minder as 100 dele per triljoen (ppt) en 1 ppt, respektiewelik. Ook met SBSE-TDS-GC-MSD kon 'n deteksielimiet van 1 ppt bereik word. In hierdie geval is vir die verlies van sensitiwiteit van die MSD in vergelyking met die ECD, gekompenseer deur die groter hoeveelheid PDMS op die roerstaaf in vergelyking met 'n SPME apparaat. Die herhaalbaarheid en lineariteit van die metode is vergelykbaar met dié van die SPMEGC- ECD metode. Al drie metodes is vergelyk vir die analise van gekontamineerde watermonsters in terme van sensitiwiteit, liniêre bereik, herhaalbaarheid, toeganklikheid en kosteeffektiwiteit. SPME-GC-ECD is vervolgens gekies as metode van voorkeur vir die analise van Eritreaanse watermonsters. Die teenwoordigheid van sekere OCPs soos a-BHC, heptachloor, heptachloorepoksied, endosulfan I, p,p'-DDE, endosulfan II, p,p' -DDD, endosulfan sulfaat en p,p' -DDT in die monsters is gedemonstreer. Die toepasbaarheid van die SPME-GC-ECD metode vir die analise van OCPs in grond-en sedimentmonsters is ook geëvalueer, en dit was moontlik om so min as 1 pg/g waar te neem. Weens tydbeperkings was dit nie moontlik om Eritreaanse grondmonsters met die ontwikkelde metode te analiseer nie.

Ecotoxicity and environmental fate of diesel and diesel blends produced by Sasol’s Fischer-Tropsch processes using natural gas and coal as feedstock as well as biodiesel and biodiesel blends

Thesis (PhD)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: World crude oil demand and production is set to increase in the long term and is projected to increase from 82 barrels per day in 2007 to an estimated 104 million barrels per day in 2030 according to the International Energy Agency. The environmental challenges posed by the current and projected increased future fuel use, with specific reference to air, aquatic and terrestrial impact, are driving producers and legislators to change fuel specifications and consequently fuel properties to be less harmful to the environment. Traditionally transportation fuels are produced through crude oil refining but in South Africa more than one third of the liquid fuels are produced synthetically through catalytic conversion of gassified coal via the Fischer-Tropsch process by Sasol. Diesel from syncrude is referred to as synthetic diesel and the refiner must blend various hydrocarbon streams, effectively tailoring the diesel to its final composition. Biodiesel from renewable sources like vegetable oils is considered environmentally more acceptable than petrodiesel because of its high biodegradability in the environment, lower sulphur and aromatic hydrocarbon content as well as lowered particulate content in the exhaust emissions. The present research was aimed at evaluating whether the composition of diesels derived from different feed stocks, that included coal, natural gas, crude oil and soybean oil, would influence its biodegradability and ecotoxicity. Acute aquatic tests that included freshwater fish, crustaceans, algae and marine bacteria were used to determine the acute toxicity of diesels. In addition, quantitative structure activity relationship models were used to estimate the biodegradation and ecotoxicity properties of the diesels in an attempt to develop a cost effective tool to determine those properties. The results indicated that the 2-D GC technique quantitatively and qualitatively identified the hydrocarbon constituents in the diesels. The relevance of using the 2-D GC technique was in identifying and quantifying the hydrocarbon breakdown products and being used in a mass balance to confirm the potential biological breakdown processes of the materials used in the present study. The differences in theoretical oxygen demand (ThOD) of the different experimental diesel blends using various blending materials and biodiesel, emphasised and confirmed the importance of calculating the ThOD for the respective blending materials when measuring the biodegradation rates. Furthermore, the biodegradation hierarchy of Pitter and Chudoba (1990) in order of decreasing biodegradability: alkanes > branched alkanes > cyclo-alkanes > aromatic hydrocarbons, could be expanded to include FAME: FAME > alkanes > branched alkanes > cyclo-alkanes > aromatic hydrocarbons. The biochemical pathways identified for the biodegradation of all the diesels was enzyme-enhanced β-oxidation. The present research also indicated that biodiesel addition to crude-derived diesels to increase the density to within the current required specifications for diesels cannot be a reality in SA because of the underdeveloped biodiesel industry. To increase the density by using biodiesel to within the specification for GTL diesel, more than 27% biodiesel would be required, which is currently is not achievable from an economic perspective as well as governmental national strategy perspective. The addition of biodiesel as lubricity enhancer seems more plausible, because less than 5% would be required for petrodiesels. The results on the ecotoxicity of the diesels and diesel blends demonstrated a general lack of acute toxic effect, especially for the fish and crustaceans used during the present study. Although algal and bacterial tests showed an effect at most of the WAF loading rates, none were high enough to enable the calculation of a median effect loading rate (EL50). QSAR‟s, like EPI Suite, together with prediction models, like the Fisk Ecotoxicity Estimation Model, can be used to screen for ecotoxicity and biodegradability of hydrocarbons found in Petrodiesels. It was less applicable for the prediction of biodiesel constituents. The use of different cut-off values for the constituents of biodiesel could be developed in future research. The use of this combination enabled the present research into the potential toxicity of hydrocarbon mixtures to be conducted, especially since tests on individual constituents are impractical. QSAR‟s may provide a relatively cost-effective way to screen for potential environmental acceptability of such mixtures. The contributors to the toxicity of mixtures of hydrocarbons found in diesels were evaluated and it appears that paraffins contribute more to the overall toxicity than previously thought and aromatics less. By putting well-defined policies and incentives in place, a robust biodiesel industry could be created that will enable SA to contribute to the mitigation of the threat of climate change, to become less dependent on foreign oil and to develop rural agriculture. The key to energy security is not one solution to South Africa‟s energy needs, but a multifaceted approach to the complex subject of sustainable energy security. The end of the hydrocarbon era of energy is not in sight, at least for the near future, but soon even hydrocarbon energy in the form of coal and crude oil will have to be re-evaluated as SA‟s major energy resource for economic and energy security. In SA the potential of developing natural gas resources through fracking, nuclear, solar, wind, biological and even wastes to energy processes as well as better energy efficiency, in a balanced and diverse energy portfolio, could pave the way toward energy security in the long run. / AFRIKAANSE OPSOMMING: Ru-olie aanvraag en produksie wêreldwyd is besig om toe te neem en die Internasionale Energie Agentskap projekteer dat wêreld ru-olie verbruik sal toeneem van 82 vate per dag in 2007 tot „n beraamde 104 vate per dag in 2030. Die omgewings uitdagings wat huidige en toekomstige toename in brandstof verbruik, spesifiek die impak op lug gehalte, water- en grond, mag hê, is dryfvere vir produseerders en reguleerders om brandstof spesifikasies te verander om minder omgewings impak te veroorsaak. Brandstof vir vervoer doeleindes word oor die algemeen van ru-olie gemaak, maar in Suid Afrika word ongeveer „n derde van die vloeibare brandtof gemaak deur middel van gekatiliseerde omskakeling van vergasde steenkool via die Fischer-Tropsch proses by Sasol. Diesel wat uit sintetiese ru-olie gemaak is, is sinteties en die raffineerder moet verskillende koolwaterstof strome meng om „n finale produk te lewer. Biodiesel wat uit hernubare hulpbronne soos plant-olies en diervet gemaak word, kan oorweeg word vir die vervaardiging van meer omgewings aanvaarbare brandstof met laer swael en aromatiese koolwaterstof inhoud en ook minder partikel inhoud in die uitlaatgas. Die huidige navorsing het beoog om te evalueer of die samestelling van diesels wat vervaardig is uit verskillende hulpbronne, wat steenkool, aardgas, ru-olie en sojaboon olie ingesluit het, die biodegradeerbaarheid en ekotoksisiteit kan beïnvloed. Akute akwatiese toetse wat varswater vis, krustaseë, alge en marine bakterieë ingesluit het, was aangewend om die akute toksisiteit van die diesels te bepaal. Kwantitatiewe struktuur aktiwiteit verwantskaps modelle is ook gebruik om die biodegradeerbaarheid en ekotoksisiteits eienskappe van die diesels te beraam om vas te stel of 'n bekostigbare alternatief beskikbaar is om daardie eienskappe te bepaal. Die resultate het aangedui dat die 2D GC tegniek kwantitatief en kwalitatief gebruik kan word om die koolwaterstowwe in die diesels te identifiseer. Die benutting van die 2D GC tegnieke is egter om die koolwaterstof afbraak produkte te identifiseer en ook om die massa balans gedurende die biodegradering te bevestig. Die verskil in teoretiese suurstof aanvraag van die verskillende diesels het die belangrikheid daarvan blemtoon en bevestig om die teoretiese suurstof aanvraag korrek te bereken en sodoende die biodegradasie korrek te bepaal. Verder kan die biodegradasie hierargie van Pitter en Chudoba (1990) volgens afnemende biodegradasie: alkane > vertakte alkane > siklo-alkane > aromatiese koolwaterstowwe, uitgebrei word om vetsuur-metielesters in te sluit: vetsuur-metielesters > alkane > vertakte alkane > siklo-alkane > aromatiese koolwaterstowwe. Die biochemiese roetes wat geïdentifiseer is vir die biodegradasie van die diesels, was ensiem-verbeterde β-oksidasie. Die huidige navorsing het ook aangedui dat biodiesel toevoeging tot ru-olie vervaardigde diesel om die digtheid te verhoog to binne huidige spesifikasies is nog nie lewensvatbaar in Suid Afrika nie as gevolg van die onderontwikkelde biodiesel industrie. Om die digtheid te verhoog met biodiesel tot binne spesifikasie verg meer as 27% biodiesel en is huidiglik nie haalbaar vanuit 'n ekonomiese persketief en ook nie vanuit 'n regerings nasionale strategie perspektief nie. Die toevoeging van biodiesel as lubrisiteits vervetering blyk meer van toepassing te wees aangesien minder as 5% biodiesel toevoeging benodig sou wees. Die resultate van die ekotoksisiteits toetse het 'n algemene gebrek aan akute toksisiteits effek aangedui, veral vir vis en skaaldiere wat in die huidige studie gebruik is. Howel alge en bakteriële toetse daarop gedui het dat 'n toksiese effek wel aanwesig was, was dit gering en kon die median effektiewe ladings koers (EL50) nie bepaal word nie. QSARs, soos Epi Suite, tesame met voospellings modelle, soos die Fisk Ecotoxicity Estimation Model, kan gebruik word om ekotoksisiteit en biodegradeerbaarheid van koolwaterstowwe in petrodiesels te beraam, alhoewel dit minder van toepassing was op biodiesel. Die gebruik van ander afsny waardes spesifiek vir biodiesel kan oorweeg word in toekomstige navorsing. Die molecules wat bygedra het tot die toksisiteit van die koolwaterstof mengsels was geëvalueeren daar is gevind dat die paraffiniese molekules meer begedra het tot die totale toksisiteit en die aromate minder. Deur goed gedefinieerde beleid en aansporings meganismes inplek te sit, kan 'n biodiesel industrie in SA geskep word wat SA sal help om by te dra tot die bekamping van klimaats vendering en sodoende minder afhanklik te wees van buitelandse olie en ook landbou in SA te bevorder. Die sluetel tot energie sekuriteit is nie een oplossing vir SA se energie aanvraag nie, maar eerder 'n veelsydige benadering tot die komplekse onderwerp van volhoubare energie sekuriteit. Die einde van koolwaterstof energie is nog nie in sig nie, ten miste nie in die nabye toekoms nie, maar binnekort sal selfs koolwaterstof energie in die vorm van steenkool en ru-olie heroorweeg moet word as SA se hoof energie hulpbronne vir ekonomiese en energie sekuriteit. In SA moet die potensiaal van natuurlike gas ontginning deur middel van hidrauliese breking, kernkrag, wind energie, biologiese energie en selfs afval tot energie prosesse bestudeer word, so-ook beter energie doeltreffendheid om sodoende 'n gebalansweerde energie portefuelje te skep wat die weg sal baan na energie sekuriteit op die lang termyn.

Undersökning av avgasemissioner till vatten från dieselinombordsmotorer i fritidsbåtar : En jämförande studie av olika bränslen

<p>In this Masters Thesis emissions to water from diesel engines in pleasure boats when driven with alternative fuels have been investigated. Two alternative fuels, Fischer-Tropsch-diesel (FT-diesel) and a blend of FT-diesel and rapeseed methyl ester (RME) have been compared with diesel of Swedish environmental class 1 (diesel EC1). The alternative fuels have been compared using two older marine diesel engines, and water samples have been taken from the water in the exhaust system before the exhaust compounds reaches the recipient. The water samples have been analysed with gas chromatography (GC) and mass spectrometry (GC/MS), to analyse volatile and semi volatile compounds including aldehydes and ketons. To investigate the acute toxicity of the exhaust water a 24 h ecotoxicological test was preformed with the crustacean Artemia fransiscana.</p><p>The results showed considerable differences in emissions of poly aromatic hydrocarbons (PAHs) and alkylated benzenes. From diesel EC1, the amounts of PAHs and alkylated benzenes were significantly higher than from pure FT-diesel and FT-diesel containing 20 % of RME. Pure FT-diesel generated the lowest amounts of PAH and alkylated benzenes. The FT-diesel containing 20 % RME generated higher amounts of benzene than diesel EC1. FT-diesel and FT-diesel with a 20 % blend of RME generated the same amount of acetone, which in turn was higher than the concentration of acetone in the exhaust water, using diesel EC1. Based on the analysis, FT-diesel is the best alternative from an exhaust emission point-of-view, concerning both the amounts and the difference in chemical composition of the combustion products released into the water.</p><p>The ecotoxicological test showed no effect on the test organism using either of the samples.</p> / <p>Fritidsbåtsintresset är stort i Sverige och båtlivet betyder mycket för många svenskar. Tyvärr är användningen av fritidsbåtar förknippad med en rad miljöproblem. Avgasutsläpp till vatten och luft från fritidsbåtsmotorer påverkar den lokala miljön avsevärt. Gammal teknik med dålig förbränning används fortfarande i stor utsträckning. På kort sikt, innan den gamla tekniken är utbytt, är det viktigt att den äldre motorparken använder miljöanpassade bränslen för att utsläppen till sjöar och hav ska bli så skonsamma som möjligt. Det är dock viktigt att utvärdera miljöpåverkan av dessa alternativa bränslen, så att det verkligen är ett bättre alternativ ur miljösynpunkt.</p><p>I detta examensarbete har utsläpp till vatten från dieselinombordsmotorer vid drift med olika bränslen undersökts. Två miljöanpassade dieselbränslen; FT-diesel (Fischer-Tropsch) och en procentinblandning av rapsmetylester (RME) i FT-diesel, har jämförts med diesel av svensk miljöklass 1 (MK1). Bränslena har testkörts i två äldre, marina dieselmotorer och prover har tagits på det avgasblandade kylvattnet innan det når recipienten. Det avgasblandade kylvattnet har analyserats med avseende på delvis flyktiga och flyktiga föreningar, aldehyder samt ketoner. Analyserna har skett med gaskromatografi och masspektormetri (GC/MS-screening) på externt laboratorium. För att testa avgasvattnets akuta giftighet har även ett akut (24h) ekotoxikologiskt test utförts på kräftdjuret Artemia fransiscana.</p><p>Analysresultatet visade skillnad i utsläpp av polyaromatiska kolväten (PAH) och alkylbensener. Vid körning med diesel MK1 genererades betydligt högre halter av PAH:er och alkylbensener, än vid körning med FT-diesel och FT-diesel med 20 % inblandning av RME. Ren FT-diesel gav de lägsta halterna av PAH:er och alkylbensener.</p><p>Utsläppshalterna av bensen uppvisade inte lika stor skillnad mellan de olika bränslena. Blandningen FT-diesel med 20 % RME gav lite högre halter bensen än vad diesel MK1 gjorde. Ren FT-diesel uppvisade lägst halt bensen. Utsläpp av aceton visade sig vara lika för FT-diesel och FT-diesel med 20 % RME och gav högre halter än vad diesel MK1 gjorde. Baserat på analysresultaten är FT-diesel det bästa bränslet ur emissionssynpunkt med avseende på vilka ämnen som hamnar i vattenfasen.</p><p>Det ekotoxikologiska testet visade ingen skillnad mellan de olika bränslena. Avgasvatten från bränslena hade inte någon akut toxisk effekt på testdjuret.</p>

Copepods comprise an ecologically important role in freshwater and marine ecosystems, which is why they are often considered an important ecotoxicological model organism. The International Organization for Standardization’s (ISO) 14669 protocol is the only guideline for the determination of acute toxicity in three European marine copepod species: Acartia tonsa. The goal of this project was to assess the feasibility of establishing and maintaining cultures of Acartia tonsa, as well as to refine current culturing and egg separation methods. Initial culture methodology proved difficult for consistent production of eggs and collection of nauplii. The development of an airlift system for the separation of eggs from nauplii and adults, based on size, successfully increased the availability of eggs, nauplii and adults. The sensitivity and relative conditions of the copepod species was assessed by running a series of 48h acute toxicity tests with the reference toxicants 3,5-dichlorophenol, 4,4’-methylenebis(2,6-di-tert-butylphenol. The acute 48 hour median lethal dose concentration (LC50), the no observed effect concentration (NOEC), and the lowest observed effect concentration (LOEC) was analyzed for the three reference compounds for of A. tonsa.

O crescimento desordenado das cidades tem trazido à tona problemas de saneamento e degradação dos recursos naturais, entre eles a água. O despejo de efluentes domésticos e industriais têm ocasionado a eutrofização dos mananciais, culminando com a proliferação dos fitoplânctons. Esta proliferação tem chegado ao ponto, em certos momentos, de acarretar episódios de floração de algas. Entre os organismos fitoplanctônicos que se desenvolvem no ambiente, estão as cianobactérias, com vários gêneros capazes de produzir diversas cianotoxinas, tais como as microcistinas, anatoxinas, cilindrospermopsinas (CY), saxitoxinas (PSPs), entre outras. Com o aumento da freqüência dos episódios de floração de algas, a probabilidade da ocorrência destas toxinas também aumenta. Como conseqüência disto e devido às exigências legais, os corpos dágua devem ser monitorados para garantir a qualidade da água para consumo humano. Com vistas ao monitoramento da presença das cianotoxinas CY e PSPs, foram realizadas investigações em alguns corpos dágua do Estado de São Paulo. Das investigações realizadas, a neosaxitoxina foi identificada pela primeira vez no Reservatório Billings e os congêneres, saxitoxina, goniautoxina 2, goniautoxina 3, foram identificados de forma inédita em amostras de água deste reservatório. Com relação à CY, foi desenvolvido um método analítico, parcialmente validado. Entretanto, esta cianotoxina não foi localizada nas amostras de água ou cianobactérias das águas superficiais dos corpos dágua estudados. Este estudo mostra a importância do monitoramento da qualidade das águas dos mananciais quanto à presença de cianotoxinas, especialmente daqueles corpos dágua com a finalidade do consumo humano. / Cities growth usually occur in an unorganized manner. This tendence can generate a variety of sanitary problems, including the degradation of natural resources, such as water bodies. As a consequence, domestic and industrial efluents cause eutrofication of water reservoir, increasing the natural level of phytoplancton, what may form algal bloom. Among the phytoplanktonic organisms that grow in this modified environment it is found the cyanobacteria. Some of them can produce different types of cyanotoxins such as microcystin, anatoxin, cylindrospermopsin (CY) and saxitoxin (PSPs). The probability of production of these cyanotoxins increase according to frequent occurrence of algal blooms episodes. Consequently, water bodies monitoring becomes important to assure water quality. The aim of this project was to develop a specific method to identify the presence of cyanotoxins CY and to investigate PSPs in water bodies in São Paulo State. The results confirmed the presence of neosaxitoxin (NEO), a toxin of PSPs family. It was the first time that Neo was indentified in Billings Reservoir along with other PSPs types: saxitoxin, gonyautoxin 2, gonyautoxin 3. Although the study also included CY monitoring, CY was not identified in the tested samples. The present study confirmed the importance of continuous searching and monitoring of water bodies to grant quality to water used for human consumption.
