Analise bioquimica e histopatologica do figado e pancreas em modelos experimentais de camundongos BALB/c machos e femeas infectados pela cepa RAL e RC de Trypanosoma cruzi / Biochemical and histopathological analysis from liver and pancreas in guinea pigs (male and female BALB/c mice) infected by RAL and RC strains from Trypanosoma cruzi

A infecção pode estar relacionadas com a cepa, morfologia e inóculo do agente infectante ou mesmo fatores inerentes ao hospedeiro vertebrado, como características imunológicas, genéticas, nutricionais e ainda hormonais. Este trabalho teve como objetivo analisar duas cepas RAL e RC de T. cruzi verificando a patologia com a mesma linhagem genética o Linhagem 1. Os animais experimentais foram os camundongos Balb/c; infectados com 2 x 104 formas tripomastigotas das duas cepas. A cepa RAL foi originalmente isolada de triatomíneo Triatoma infestans, proveniente de Santa Rosa do Viterbo ¿ SP e a cepa RC isolada a partir da infecção natural do cachorro-do-mato Cerdocyon thous azarae, capturado na cidade de Cássia dos Coqueiros, no Estado de São Paulo. Para a avaliação bioquímica e histopatológica foram inoculados o protozoário T. cruzi em camundongos fêmeas e machos coletando-se o sangue e retirando-se o pâncreas e fígado nos respectivos dias 1.º (grupo controle), 6.º, 9.º, 12.º e 14.º. Após a inoculação foram analisados: peso, parasitemia, glicose, fosfatase alcalina, proteínas totais, uréia, creatinina, aspartato-transaminase (AST ou TGO), colesterol e triglicerídeos. Houve diminuição do peso, variação da parasitemia de acordo com o dia, reduzida taxa de glicose sangüínea bem como de todos os outros parâmetros bioquímicos já mencionados, exceto uréia. Verificou-se que a cepa RAL foi mais virulenta que a cepa RC, machos mais susceptíveis à infecção, pois houve maior parasitemia nesse grupo de camundongo. Os dois grupos apresentaram acentuadas alterações bioquímicas e histológicas. Na análise histológica, os tecido hepático e pancreático estavam acentuadamente desorganizados, com aparecimento granulomatoso e infiltrados celulares e presença de formas amastigotas ou ninhos de amastigotas. Os resultados encontrados nos camundongos inoculados pelas cepas RAL e RC de T.cruzi mostraram alguns parâmetros similares, outros alterados mesmo sendo de semelhante linhagem genética / Abstract: The physiologic evaluations of infected hosts by Trypanosoma cruzi undergo a homeostatic imbalance, which may be related to the parasite strain factors as well as to the morphology and inoculation of the T. cruzi infective agent or also to the inherent factors of a vertebrate host such as immunologic, genetic, nutritional and hormonal characteristics. This work aimed to analyze two strains from T. cruzi - RAL and RC - checking this protozoan pathology with the same phylogenetic line (Zidodema 1). The guinea pigs were Balb/c mice infected with 2 x 104 forms from tripoamastigotes of the two strains. Primarily, RAL strain was isolated from Trachoma infestans triatomineum, came from Santa Rosa do Viterbo /SP and RC strain was isolated from a bushdog natural infection -Cerdocyon thous azarae- caught in Cássia dos Coqueiros /SP. For a biochemical and histopathological evaluation T. cruzi was inoculated in female and male mice, collecting blood and removing their pancreas and livers at the 1st ?control group? 6th, 9th, 12th and 14th days respectively. After the inoculation, weight, parasitic index, glucose, alkaline phosphatise, total proteins, urea, creatinine, aspartate-transaminase (AST or TGO), cholesterol and triglycerides were analyzed. There were weight reduction, parasitic index variation according to the specific day, blood glucose rate diminished as well as all the biochemical parameters already mentioned, except urea. It was verified that RAL strain was more virulent than RC one because the males were more susceptible to the infection, since there was a great parasitic index in this group, in both strains. Biochemical and histological changes were more emphatic in both groups. In histological evaluation, the hepatic and pancreatic tissues were stressed disorganized, appearing granulomatous formations, infiltrated cellular and the presence of amastigotes. The finding results in mice inoculated by RAL and RC strains from T. cruzi presented some parameters so similar and others were changed although the genetic line was the same / Doutorado / Doutor em Parasitologia
