Studium antimikrobiálního efektu hroznových semen

This paper is dedicated to gathering information about utilization of Vitis vinifera L. extracts, mainly grape seed extract (GSE), production of GSE in two variants (aqueous or methanol GSE) and description of its characteristics. Extract was rich in polyphenols (13462 mg/l GAE), measured by Folin-Ciocaltel method. Also 3615 mg/l GAE of flavonols was present in GSE. Antioxidant activity measured by DPPH method was high, 7978 mg/l GAE. Disc diffusion method was used for antimicrobial activity analysis. GSE's effect was tested on five common microorganisms. Results of the experiment show that GSE strongly inhibits growth of Bacillus cereus, Candida tropicalis and Enterococcus faecalis. However, it was not effective against Escherichia coli and Pseudomonas aeruginosa.

The aim of this thesis is to summarize the options of hop products leaded by hop extracts. The literature research focuses on chemical composition of hop, key transformation of hop bitter acids used in the brewing industry and also describes the technology of production of individual hop preparations and possibility of their application. The next part summarizes the beer production process including associated mikrobiology, which is an importand part of all stages of beer production. The experimental part describes production of six variations of beer chosen from different hop products, their chemical analysis and sensory evaluation. The results of the tests are evaluated statistically and are compared with each other at the end of the thesis.

Produkce alelopatických látek u vybraných bylin a jejich biologická účinnost

The thesis is focused on the study of allelopathy and allelopathic effects of Ambrosia artemisiifolia and Festuca rubra on selected species of herbs. The first theoretical part describes plants relationships and available information about allelopathy. The second part deals with the influence of Ambrosia artemisiifolia on the growth of Plantago lanceolata, Lotus corniculatus, Arrhenatherum elatius, Festuca rubra, Phacelia tanacetifolia and Pisum sativum in outdoor conditions in different cultivation substrates. The effect of extracts from roots of Festuca rubra and synthetic m-tyrosine on the germination of seeds Ambrosia artemisiifolia was evaluated. The obtained results confirmed the effect of Ambrosia artemisiifolia on the growth of the studied plants and the effect of the tested solutions on germination seeds of Ambrosia.

Vliv hroznových semínek na krevní parametry laboratorních potkanů

The aim of this master thesis was to experimentally determine the effect of grape seeds (their metabolically active compounds) on blood parameters of laboratory rats. The experiment was conducted in a laboratory of The Mendel University, Brno in late 2015. Four groups of laboratory rats were supplied with; 1) water, 2) water in combination with grape seed extract, 3) water with grape seed extract and cadmium, 4) water in combination with cadmium. Half of the laboratory animals were killed after 14 days and second half were killed after 28 days. Blood samples were analyzed in the veterinary laboratory (VLAB). The results were evaluated and compared. From our results, we can assume that feeding grape seed extract for its protective effect against damage of organism caused by cadmium, cannot be recommended because this effect was not proved.

Primarily its influence on microcrystalline cellulose compressibility was studied. Its effects were compared to sodium stearylfumarate and colloidal silicon dioxide. The used lubricants were wixed with the filler in the concentrations ranging from 0.1 to 1%. The mass flow rate was evaluated in the filler itself and its mixtures. Subsequently, the tablets were made using the compression forces ranging from 2.5 to 7.5kN. The compaction process was evaluated using the force- displacement method. Furthermore, the tablet hardness, friability and disintegration time was tested. The results showed that the rice extract can be used as lubricant for the used model filler. Unlike other evaluated glidants, it must be used at concentrations up to 0,25%. The use of higher concentrations does not improve the flow properties of tableting mixtures. It affects the properties of intermediate and final products. In this thesis the properties of tablets made of two lactose types, which were used as model fillers, were studied. These were Tablettose® 80 and Lactopress® Anhydrous. In both lactose types, five sieved size fractions of the particle size ranging from 9 to 346µm and their original unsieved mixture were used. The compaction forces ranging from 2 to 10kN were used for tablet preparation. Magnesium stearate was used for external lubrication. The evaluation of the prepared tablets was done mainly by using the pharmacopoeial methods. The evaluated parameters were friability, disintegration time and hardness. Furthermore, the elasticity of the tablets and the evaluation using the force-displacement method parameters were assessed. Einfluss von Mistelextrakt auf die Migration von kaninen Mammatumorzellen und die Genexpression und das Wachstum von humanen B-NHL-Zelllinien in Bezug auf den Einsatz der Misteltherapie bei Mensch und Tier

Für diese Arbeit wurden in-vitro-Studien an kaninen Mammatumorzellen und humanen follikulären B-NHL-Zellen durchgeführt. Es wurden erstmals die migratorischen Eigenschaften einer kaninen Mammatumorzelllinie (CMT-U-27, infiltrierendes duktuläres Karzinom mit Metastasen) und deren Beeinflussbarkeit untersucht. Die Experimente zeigten einen leichten promigratorischen Einfluss von Noradrenalin und EGF; mit dem Tumorpromotor PMA wurde eine sehr deutliche Stimulation der Migration erreicht. Es konnte gezeigt werden, dass Iscador® M in der Lage ist, die PMA-stimulierte Migration der CMT-U-27-Zellen zu hemmen, wobei die spontane, Matrix-induzierte Zellmigration unbeeinflusst blieb. Zwischen der migratorischen Aktivität von Tumorzellen und ihrer Fähigkeit Metastasen zu bilden, besteht ein Zusammenhang. Durch einen inhibierenden Effekt auf die Migration von Tumorzellen könnte eine Progression der Erkrankung durch Metastasenbildung verhindert oder zumindest verzögert werden. IL-6 dient als prognostischer Faktor bei lymphoproliferativen Erkrankungen der B-Zellen und kann sich negativ auf den Krankheitsverlauf bei B-Lymphompatienten auswirken. Unterstützend zu den guten in-vivo Erfahrungen in der Lukasklinik in Arlesheim, Schweiz, sollte in dieser Arbeit untersucht werden, ob Iscador® P bei follikulären B-NHL-Zelllinien (DoHH2, WSU-NHL, Sc-1) einen autokrinen IL-6-Loop auslösen kann. Dafür wurden die Zellen über definierte Zeiträume mit verschiedenen Iscador® P-Konzentrationen inkubiert. Es wurden Proben entnommen und durchflusszytometrische Messungen auf membrangebundenen IL-6R und gp130, Messungen der mRNA-Expression von IL-6, IL-6R und gp130 mittels Real-Time-RT-PCR und ELISA-Messungen zur Bestimmung von IL-6, löslichem IL-6R (sIL-6R) und löslichem gp130 (sgp130) durchgeführt. Es konnte kein Einfluss von Iscador® P auf die Oberflächen- oder Genexpression von IL-6 und den dazugehörigen Rezeptorkomponenten gp130 und IL-6R nachgewiesen werden. Zudem wird kein IL-6, sgp130 oder sIL-6R von den Zellen freigesetzt. Damit kann ein autokriner IL-6-Loop und ein durch Freisetzen von sIL-6R ermöglichtes Transsignalling bei den in dieser Arbeit untersuchten follikulären B-NHL-Zelllinien ausgeschlossen werden. In Wachstumskurven konnte gezeigt werden, dass Iscador® P den proliferativen Einfluss von IL-6, bei gleichzeitiger Inkubation, aufhebt. Genexpressionsmessungen der apoptoserelevanten Gene bcl-2, bax, bcl-xl und bad mittels Real-Time-RT-PCR zeigten bei den WSU-NHL-Zellen, dass die Apoptose über ein Verschieben des bcl-2/bax-Quotienten ausgelöst wird, welches mittels durchflusszytometrischer Analyse bestätigt werden konnte. Im Gegensatz dazu konnte bei Sc-1 Zellen keine Verschiebung des bcl﷓2/bax-Quotienten in Richtung Induktion der Apoptose beobachtet werden, sodass die bei dieser Zelllinie beobachtete Hemmung der IL-6 mediierten Proliferation durch Iscador® P, durch einen anderen, bislang noch nicht geklärten, Mechanismus ablaufen muss.
