Ochratoxin A: endocrine disruption potential,transplacental kinetics and maternal exposure assessment

Mycotoxin contamination in food commodities is an age-old problem. Due to the detrimental impact of mycotoxins on human health, exposure to mycotoxins and their health implications have been increasingly recognized. Ochratoxin A (OTA), one of the mycotoxins, has been found to cause diverse toxicities in animals, with potential impact on human health. OTA has been reported to be teratogenic and interfere with steroidogenesis in vivo. Chronic exposure of pregnant women to OTA may be hazardous for the human foetus, especially when endocrine and developmental toxicities are taken into consideration. Accordingly, in the first part of this project, I hypothesized that OTA may interfere with enzymes involved in human placental steroidogenesis. By evaluation of human placental 3β–hydroxysteroid dehydrogenase/isomerase (3β-HSD) at both mRNA and protein (hormonal) levels, my results showed that OTA could up-regulate 3β-HSD1 expression in human placental cells with concentration relevant to human exposure. This study is the first to report the endocrine disruption potential of OTA in human placental cells. As several mycotoxins have been demonstrated previously to cross human placental barrier and OTA has been associated with developmental toxicity in vivo, I further hypothesized that OTA may be transferred through human placenta and accumulate in foetal compartment. In the second part of this project, human perfused placenta was used to investigate the placental toxicokinetics of OTA using concentrations found in serum of pregnant women. Findings from this study clearly showed that the transfer of OTA through term human placenta was minimal, contradicting the existing epidemiological studies reporting higher foetal OTA levels than maternal. This is the first study where transplacental kinetics of OTA has been studied in human perfused placenta. To assess the relevance of the study findings, it is very important to provide information on maternal OTA exposure during pregnancy. Currently there is limited information regarding OTA exposure of pregnant women. The third part of this project aimed at evaluating the frequency and level of exposure to OTA in pregnant women from Egypt, where exposure to dietary mycotoxins is common due to the environmental conditions. Biomonitoring of both serum and urinary OTA levels showed that more than 70% of pregnant women were exposed to OTA with a geometric mean of 0.27 ng/ml in serum and 37.21 pg/mg creatinine in urine indicating frequent exposure of this subpopulation. As an ultimate aim, maternal-foetal risk assessment served as a conclusive part of this project to predict and evaluate both maternal and foetal risk of exposure to OTA during pregnancy. Data from the exposure of pregnant women in Egypt to OTA were further ultilized to conduct maternal-foetal risk assessment in relation to OTA exposure. Based on the refined Klaassen equation for exposure estimation during pregnancy and the benchmark dose approach for risk assessment, this subpopulation of pregnant women generally was not exposed to OTA in a high-risk manner. However, considering the suspected chronic exposure beginning from early pregnancy with high foetal susceptibility and diverse toxic effects, and in particular the potential endocrine disruption of OTA, keeping OTA exposure to a minimum is recommended. / published_or_final_version / Biological Sciences / Doctoral / Doctor of Philosophy

This thesis reports the effects of an altered intrauterine environment on the offspring???s renal, cardiovascular and renin angiotensin systems. After a midgestational asphyxial episode in fetal sheep (30 min total umbilical cord occlusion at 90 days; term 150 days) the hydrops that resulted had not completely resolved by 130 days. While the heart and kidneys were apparently unaffected, the brain and lung weights were 37% and 50% lower than sham values, respectively and there were joint contractures. The effects of maternal renal disease on the offspring were investigated. Although in utero fetuses of subtotally nephrectomised ewes (STNx) had altered urine flow rates, sodium excretion, haematocrits, plasma chloride and plasma renin levels, by 1-2 weeks after birth these values in the lambs (STNxL) were similar to controls (ConL) under baseline conditions. Body weight and the weights of most organs were similar, including the kidney, in which glomerular number was normal. In the neonatal period, the lambs were subjected to four challenges: furosemide (2 mg/kg intravenous bolus), infusion of angiotensin II and phenylephrine, intravenous infusion of 0.15M saline (50 ml/kg over 30 min) and haemorrhage (20% estimated blood volume over 10 min). These challenges revealed evidence of programming of several aspects of the renal, cardiovascular and renin angiotensin systems in the STNx offspring. As young adults at 6 months of age, male and female offspring of STNx ewes were normotensive and had normal renal function. On a high salt diet (HSD, 0.17M NaCl in 8L water for 5-7days), female offspring of both groups did not become hypertensive. However, the STNx offspring must have retained salt and water as plasma sodium was increased and haematocrit was decreased. In the STNx offspring only, there was a relationship between glomerular filtration rate (GFR) and mean arterial pressure, indicating an inability to maintain a constant GFR in response to changes in arterial pressure.

A leucina é usada experimentalmente para minimizar as alterações do metabolismo protéico durante o crescimento tumoral. Este trabalho investigou os efeitos do crescimento tumoral sobre o metabolismo protéico e estresse oxidativo nos fetos provenientes de ratas alimentadas com dieta suplementada com leucina. Os grupos de fêmeas prenhes foram divididos da seguinte forma: controle (C), controle com dieta suplementada com leucina (L), tumor (W), tumor com dieta suplementada com leucina (WL), dieta pareada á ingestão diária das ratas do grupo W (Cp), dieta suplementada com leucina e pareada á ingestão diária das ratas do grupo WL (Lp). Após 20 dias de prenhes os músculos fetais foram analisados. Os resultados mostraram que a síntese protéica fetal foi reduzida no grupo W quando comparados aos outros grupos. Houve significativa recuperação da síntese protéica no músculo dos fetos WL sugerindo efeito positivo do uso de aminoácido de cadeia ramificada nessa situação. Ocorreu aumento significativo da degradação protéica nos fetos W. Essa perda protéica foi amenizada pelo uso da dieta suplementada com leucina indicando efeito benéfico sobre a degradação protéica no grupo WL que obteve valores similares ao observado no grupo C. A atividade da enzima glutationa-S-transferase ficou elevada no músculo de fetos WL em comparação com o grupo W. Também houve aumento na atividade da enzima fosfatase alcalina no grupo WL indicando maior atividade celular nesse grupo quando comparado ao grupo W. A presença de MDA (Malondialdeído) no músculo fetal também ficou reduzida nos fetos WL. Com a análise desses parâmetros conclui-se que a dieta suplementada com leucina pode alterar o metabolismo muscular fetal aumentando a síntese protéica e reduzindo o efeito danoso do estresse oxidativo. / Abstract: Cancer occurs in approximately 1 per 3.000 pregnancies and accounts for one-third of maternal deaths. Leucine has been used experimentally to minimize the protein metabolism changes during tumor growth. The present work investigated the effects of tumor growth on muscle protein metabolism and oxidative stress in fetus pregnant rats fed a leucine-rich diet. Fetal groups from pregnant Wistar rats were: control (C), control fed leucine-rich diet (L), tumor-bearing (W), tumor-bearing fed leucine-rich diet (WL), Pair-fed control (P) and Pair-fed Leucine (PL). After 20 days the fetal muscles were analyzed. The results showed that fetal protein synthesis was decreased in W group, when compared to the other groups. The significantly recover on muscle protein synthesis in fetus of WL group suggested a positive effect of this branched-chain amino acid. There was a significant increase on protein degradation in W fetus and a protective effect of the leucine-rich diet on muscle protein waste in WL group, since it was similar to C group. The muscle enzyme glutathione-S-transferase activity was increased in WL fetus in comparison to W group. The muscle alkaline phosphatase activity increased in WL group. The malondialdehyde content (MDA) in fetal muscle was decreased in WL fetus.
