A histology-based fish health assessment to determine the health status and edibility of two indicator fish species from the Roodeplaat Dam22 June 2011 (has links)
M.Sc. / A comprehensive histology-based fish health assessment was implemented at the Roodeplaat Dam (RD), Pretoria, South Africa, to determine the health status of two fresh water fish species, Clarias gariepinus (n = 20) and Oreochromis mossambicus (n = 18), after exposure to pollutants. RD is known to be polluted, being impacted from two sewage treatment plants, industry, housing and agriculture all upstream of the reserve. Excessive nutrient loads, such as orthophosphate, resulted in the dam becoming eutrophic. The quality of water in the RD was a cause for concern, as the fish from the dam are being used as a source of food by the local people. The histology-based fish health assessment included fish necropsy, calculation of gross body indices and histological assessment. Water quality (chemical and physical) analysis was also carried. Furthermore, target chemical analysis of fish tissue (skeletal muscle) was performed to determine the levels of contaminants in the consumable parts of the fish. These results were compared with existing international guidelines to determine whether the harvested fish were safe for human consumption. All the information obtained was integrated in a qualitative and quantitative scenario-based health risk assessment.

M.Sc. / The tigerfish, Hydrocynus vittatus member of the Alestidae, are ferocious and fierce predators capable of consuming prey up to 40% of their size. In South Africa H. vittatus can be found in the major east-flowing rivers from the Limpopo River to the Phongolo River flowing into the Pongolapoort Dam making up the southernmost population. The Pongolapoort Dam also known as Lake Jozini is located in the high-rainfall subtropical region of northern KwaZulu-Natal. The Pongolapoort Dam wall was completed in 1973 to make it the fifth largest dam in South Africa with a total surface area of 2 445.9 x 106 m2. It was primarily built for the irrigation of sugar-cane in the surrounding regions. The Fish Health Assessment Index is used as a quantitative index that identifies morphological alterations and assigns them a score for statistical comparison. The main purpose of the macroscopic Fish Health Assessment Index is to detect gross changes in the health of fish populations through necropsy early enough for remedial actions to be put into place. Histopathology is used as an important diagnostic tool for detecting infectious and non-infectious diseases and can therefore be used to assess the health of fish populations by analysing selected target organs. Catch and release (C&R) angling has become a very widely promoted conservation tool as sport angling continues to increase. Anglers release their fish under the assumption that the fish will survive; however, the end results have been shown to vary greatly, from death of the fish to sub-lethal end points such as exhaustive exercise, injury and/or air exposure which will affect the fitness of the fish. The first aim of the study was to determine the health status of H. vittatus and C. gariepinus from the Pongolapoort Dam by means of a histology-based fish health assessment protocol applied to a total of 45 H. vittatus specimens collected in February 2009 (n = 30) and July 2009 (n = 15) and a total of 19 C. gariepinus specimens collected in October 2009 (n = 10) and April 2010 (n = 9). In the H. vittatus specimens DDT was found in the axial muscle; the concentrations of these in February 2009 (5 403.9 ng∙g-1 lipid) and July 2009 (5 537.4 ng∙g-1 lipid). The Mean Index values showed that the Kidney Index (IK), Gill Index (IG) and Fish Index (IFISH) were higher in fish from the February survey while the Liver Index (IL) was higher in those collected during July. Liver alterations identified included intercellular oedema, granular degeneration, vacuolation, nuclear pleomorphism and lymphocyte infiltration. Kidney alterations included dilation of the glomerulus capillaries, vacuolation and hyaline droplet degeneration. Gill alterations identified included telangiectasia and hyperplasia of the secondary lamella, congestion, and rupture of pillar cells.

Fish histopathology as a tool to assess the health status of freshwater fish species in the Albasini Dam, Limpopo Province, South AfricaNibamureke, Marie Clémentine Uwineza 01 July 2015 (has links)
MSc. (Zoology) / The Albasini Dam was used as a reference site outside the DDT (1, 1, 1-trichloro-2, 2-bis (p-chlorophenyl) ethane) - sprayed area in a previous survey conducted from 2006 to 2008. DDT, endocrine disrupting chemicals and inorganic chemicals were detected in the dam. A histological analysis of fish from the dam showed histological alterations in heart, liver, gills, and gonads. Therefore, it was necessary to follow up the health status of the dam by monitoring the levels of organic and inorganic chemicals and their effects on fish. The aim of the present study was to determine the health status of three freshwater fish species, Clarias gariepinus, Oreochromis mossambicus, and Coptodon (Tilapia) rendalli from the Albasini Dam, using fish histology as a biomonitoring tool. In total, 18 fish were sampled using gill nets; these included Clarias gariepinus (n=5); Coptodon (Tilapia) rendalli (n=4) and Oreochromis mossambicus (n=9). The histology-based fish health assessment included a standard fish necropsy; a calculation of blood parameters (haematocrit, leukocrit and total plasma protein); somatic indices and the condition factor and a qualitative and semi-quantitative histological assessment of five target organs: liver, heart, gills, kidney and gonads. Water, sediment and fish muscles samples were collected and analysed for inorganic chemicals and organic chemicals. Inorganic chemicals were analysed by means of Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) and Inductively Coupled Plasma-Optic Emission Spectrometry (ICP-OES). An assessment of the health status and edibility of fish from three impoundments in the North West Province, South AfricaMooney, Amanda 01 May 2013 (has links)
M.Sc. (Aquatic Health) / The North West Province is mineral rich and known for its extensive mining and agricultural activities. These activities drain organic and inorganic pollutants in our waterways, possibly resulting in reduced water quality in dams. Organic and inorganic pollutants such as nutrients, organochlorine pesticides and metals may be present in unacceptable levels possibly affecting fish as well as human health. The North West dams are known for their high nutrient loads and are mostly classified as hypertrophic. In order to better understand the possible effects of the combined pollutant loads on fish health and edibility of fish, (1) a fish health assessment and (2) a human health risk assessment should be conducted. The fish health assessment and the human health risk assessment form an important role in the establishment of water quality standards or/and guidelines for acceptable levels of safe consumption of fish respectively. The aim of this study was to determine (1) if Oreochromis mossambicus from the Klipvoor -, Roodekopjes - and Vaalkop Dams show adverse effects and (2) if consumed, pose a human health risk. Establishing biological and environmental drivers that influence the health assessment index as a biomonitoring toolPhala, Balance Matseilane January 2019 (has links)

Zoology)) -- University of Limpopo, 2019 / In South Africa, a high anthropogenic demand of water for domestic, recreational, agricultural, urbanisation and industrial purposes has placed enormous pressure on freshwater resources and has led to a decline in water quality. In addition to measuring water quality variables, the health assessment index (HAI) advocated by AvenantOldewage and Swanepoel (1993), and modified by Crafford and Avenant-Oldewage (2009) by incorporating the inverted parasite index (IPI), and adapted further by Madanire-Moyo et al. (2012) who excluded the white blood cells counts, has been used as a rapid and inexpensive means of assessing and monitoring fish health and, in turn, the state of aquatic ecosystems. However, few studies have evaluated the latter approach in conjunction with other biomonitoring indices to assess the water quality of impoundments. Thus, the aim of this study was twofold. The first was to evaluate the HAI adapted by Madanire-Moyo et al. (2012) as a means to establish the health status of Oreochromis mossambicus (Peters, 1852) sampled from various impoundments based on a once-off survey. The second was to establish if the adapted HAI, in conjunction with selected biomonitoring indices and environmental variables, could describe monthly and seasonal fluctuations of O. mossambicus health.
Once-off surveys were conducted between April and May 2016 at five impoundments. The Luphephe-Nwanedi Dam located in the upper catchment of the Limpopo River System and Rhenosterkop Dam situated in the Elands River, a tributary of the Olifants River System, served as control sites due to little or no anthropogenic activities occurring in their catchment areas. In addition, surveys were conducted at Loskop and Flag Boshielo dams and Phalaborwa Barrage situated in the main stem of the Olifants River for comparison purposes. The latter three impoundments were selected because they vary in water quality and pollution levels. Monthly surveys were carried out at Flag Boshielo Dam from February 2016 to February 2017 to establish if the biomonitoring tools and the quantification of water and sediment quality variables, could describe and predict seasonal fluctuations in the health of O. mossambicus.
To this end, water quality variables were measured during each survey. Water and sediment samples were collected for analysis of nutrients and metals. Oreochromis mossambicus were collected using gill nets. Fish health was assessed using the adapted HAI that was based on observing parasite burden, haematocrit determination and anomalies in body tissues and organs in conjunction with determining gonad and

hepato-somatic indices, the condition factor, blood glucose levels, microscopy analyses of fish gills and metal concentrations in fish muscle tissues. Water quality in terms of pH, total dissolved solids and electrical conductivity, nutrients and some aqueous and sediment metals exhibited significant differences (p ˂ 0.05) between impoundments surveyed with water quality from good to poor being in the order of: Luphephe-Nwanedi Dams ˂ Rhenosterkop Dam ˂ Loskop Dam ˂ Phalaborwa Barrage ˂ Flag Boshielo Dam. The HAI scores varied significantly (p ˂ 0.0001) between impoundments and substantiated water quality variables results during once off surveys, indicating that the health of fish from Flag Boshielo Dam were most affected when compared to the health of fish surveyed from the other impoundments. Findings from monthly surveys conducted at Flag Boshielo Dam indicated better conditions in terms of water quality variables during a period of high inflow as opposed to periods of low inflow. The HAI scores obtained for fish exhibited significant (p ˂ 0.0001) differences between the months surveyed and were found to be in agreement with the water quality findings, indicating that flow regimes and water quality of an impoundment have an impact on fish health.
The condition and somatic indices findings did not seem to be sensitive enough to discriminate between the impoundments during the once off surveys. While during monthly surveys, these indices were significantly (p ˂ 0.0001) different between the months, showing to be affected by seasonal fluctuations. Knowledge of the HAI in conjunction with blood glucose levels, gill histopathology and the arithmetic mean thickness of gill epithelium (Har) best described the health of fish in both the once off and monthly surveys. In conclusion, the findings of this study emphasised the HAI premise that fish from more polluted sites would be more impacted as opposed to less impacted sites, making the HAI adapted by Madanire-Moyo et al. (2012) an effective and rapid biomonitoring tool that can be used in the field. Furthermore, this study proved that the HAI can be used either solely or in association with the parasite index (PI) or IPI depending on the objectives of the study, as no pronounced differences were evident when using HAI, HAI with PI and HAI with IPI. / National Research Foundation (NRF) and Water Research Commission (WRC)

A histology-based fish health assessment to determine the health status of two freshwater fish species from three hypertrophic impoundments in the North-West Province, South AfricaSikhakhane, Ngcebo Nkululeko Treasure 11 February 2014 (has links)
M.Sc. (Aquatic Health) / South Africa’s freshwater ecosystems are under threat due to anthropogenic activities. Recent reports have shown that the Hartbeespoort- (HBPD), Klipvoor- (KD) and Bospoort Dam (BD) impoundments situated in the North-West Province are polluted as a result of mining, industrial and agricultural activities. These impoundments are classified as being in a hypertrophic state. The health status of fish in these impoundments is of concern, and needs to be investigated. For this purpose, Clarias gariepinus and Cyprinus carpio were collected for a histology-based fish health assessment. These two species are present in the selected dams and are some of the preferred fish species for human consumption by the local people. The aim of the study was to determine the health status of these fish species in HPBD, KD and BD using a semi-quantitative histopathological assessment on selected target organs and compare the results to fish from a reference site, the Marico-Bosveld Dam (MBD). Gill nets were used for the collection of specimens. Fish were macroscopically examined, weighed and measured for the calculation of the condition factor. Blood samples were collected for haematocrit, leukocrit and total protein determination. Once sacrificed, a necropsy-based health assessment (HAI) was performed to determine any abnormalities within the visceral cavity. The selected target organs were excised and weighed for the calculation of selected biometric indices. Collected tissue samples were processed using standard histological techniques and the prepared slides were stained using Haematoxylin and Eosin. Macroscopically, a number of fish from polluted sites exhibited liver with fatty change and focal discoloration as well as severe parasites within the visceral cavity. As a result, higher HAI values were recorded in polluted sites compared to the reference site. The haematocrit values varied from normal to below and above the normal range. Leukocrit values were within the normal range except for C. gariepinus from HBPD, which was above the normal range. Total protein values were within the normal range for both species for all sites. The condition factor and hepatosomatic index (HSI) values were within the accepted range except for the HSI of C. gariepinus from MBD. The mean organ index for all study sites fell within class 1 with the exception of the liver index from HBPD, KD and BD which was in class 2. Class 1 (index value <10) is indicative of slight histological alterations while class 2 (index value 10-25) translates to moderate histological alterations. Overall, a higher prevalence of histopathological alterations was noted among fish from polluted sites compared to fish from the reference site. Hence, on the basis of the macroscopic and histopathological results, the fish from the polluted sites were more affected in terms of these parameters compared to fish from the reference site. The results therefore provide valuable information for future bio-monitoring studies in the HBPD, KD and BD impoundments.
